Introduction:-
Propofol is an intravenous sedative-hypnotic.
Most common intravenous agent used for induction and
maintain ace of Anaesthesia.
USES
• Propofol slows the activity of your brain and nervous system.
• Propofol is used to put you to sleep and keep you asleep during general anaesthesia
for surgery or other medical procedures. It is used in adults as well as children 2
months and older.
• Propofol is also used to sedate a patient who is under critical care and needs a
mechanical ventilator (breathing machine).

HOW IS PROPOFOL GIVEN?
• Propofol is injected into a vein through an IV. A healthcare provider will give you this
injection.
• You will relax and fall asleep very quickly after propofol is injected.
• Your breathing, blood pressure, oxygen levels, kidney function, and other vital signs will
be watched closely while you are under the effects of propofol.
DOSAGE & ADMINISTRATION
• Healthy Adults Less than 55 Years of Age: 40 mg every 10 seconds until induction onset (2to 2.5 mg/kg)
• Elderly, Debilitated Patients: 20 mg every 10 seconds until induction onset (1to 1.5 mg/kg)
• Cardiac Anaesthesia: 20 mg every 10 seconds until induction onset (0to 1.5 mg/kg)
• Neurosurgical Patients: 40 mg every 10 seconds until induction onset (1to 2 mg/kg)
• Paediatric – healthy 3 years of age or older: 2to 2.5 mg/kg administrated over 20-30 seconds .
INDICATIONS
• Because of its early induction, early & smooth recovery , inactive metabolism & anti
emetic effects it is the IV agent of choice for day care surgery.
• Along with opioids (alfentanil or remifentanil) it is the agent of choice for total
intravenous anaesthesia(TIVA).
• Propofol Injection can be used to produce sedation in ICU patient.
• Agent of choice for induction in susceptible individuals for induction for malignant
hyperthermia.
CONTRAINDICATIONS
• Hypersensitivity to propofol which contains soyabean,oil and egg phospholipid.
• Obstetrical procedures(Propofol crosses the placenta and maybe associated with neonatal
depression).
• Propofol is not recommended for use in nursing mothers because propofol has been
reported to be excreted in human milk and the effects of oral absorption of small amounts
of propofol are still known.
• Propofol not recommended for children below the age of 3 years because safety and
effectiveness have not been established yet.
