Formulation Project Report Suspension Paracetamol

This document reports on a formulation project to create a paracetamol suspension. It introduces paracetamol and suspension requirements, then describes the project to create a 100ml suspension containing paracetamol, CMC Na, tween 80, propylene glycol, syrup simplex, and aquadest. The methods, materials, and results are presented, showing a homogeneous but cloudy and precipitate-containing suspension. Testing found changes in viscosity and pH, meaning the formulation did not meet requirements for circulation and consumption. In conclusion, the formulation was a paracetamol syrup rather than a true suspension.

INTRODUCTION
● Paracetamol, also known as
acetaminophen, is a medication used to
treat fever and mild to moderate pain. At
a standard dose, paracetamol only slightly
decreases body temperature; it is inferior
to ibuprofen in that respect, and the
benefits of its use for fever are unclear.
INTRODUCTION
● According to The International
Pharmacopoeia, Paracetamol oral suspension
is a suspension of Paracetamol in a suitable
vehicle, which may be flavoured. It contains
not less than 90.0% and not more than
110.0% of the amount of paracetamol
(C8H9NO2) stated on the label.

Methods
1. balance the scales, weigh the ingredients in the recipe
2. calibrate the bottle
3. Cook the water to a boil, then wait until the temperature reaches (±80° 𝑐)
4. Develop CMC Na over hot water (±80°𝑐) by sprinkling it evenly, then wait (±15 )
5. In another mortar, grind the paracetamol ad finely, then add tween 80, grind ad homogeneously.
[Link] propylene glycol to the pct+tween 80 mortar, grind ad homogeneously (mixture I)
7. Crush the CMC Na that has been developed ad mucilage
8. Put the mucilage into the mixed mortar (1), grind it to form a corpus suspension
9. Add syrup simplex into the mortar, grind ad homogeneous
10. Dilute with aquadest, then put into a bottle
11. Add aquadest ad calibration limit
12. Cover the bottle, shake ad homogeneous
RESULT AND DISCUSSION
The paracetamol suspension was mixed homogeneously. which opposes the
slightly cloudy nature of the suspension and there is a precipitate from the
formation of paracetamol and Na CMC.

In the result of suspension there is foam which is quite disturbing the appearance
of the suspension, This is due to the saponification of the tween 80 substance.
except if the foam is filtered before packaging.

Tween 80 has HLB 15, which is a detergent and solubilizing agent. tween 80 also
causes the suspension to become clear because the paracetamol is completely
dissolved.

Conclusion
Results of our research were compared with conditions, respectively criteria, set by
International Pharmacopeia, respectively British Pharmacopeia (BP) and Indonesia
Pharmacopeia.
it can be concluded that the formulation in the recipe is not a paracetamol suspension
formulation, but a paracetamol syrup formulation.
in some experiments there was a change in viscosity and pH.
This shows that the suspension made does not meet the requirements for circulation and
consumption
