Set up the frog muscle preparation as follows:
1. We can observe a muscle twitch by removing the gastrocnemius
from a frog and administering a small stimulus to a muscle. Compress
the femur in the femur clamp. The muscle was left attached to part of
the femur,
2. Adjust the clamps so that the thread is vertical and the
displacement transducer rod rests just on the upper stop with no slack
in the thread. For the Gastrocnemius muscle, one end of a thin, straight
wire was tied to the tendon and the other end attached to an isometric
lever.
3. Position the stimulator electrodes so that they lie against the muscle about
midway between the knee and the tendon; the two electrodes should not
touch one another, but both must be in contact with the muscle. In here,
we will determine the threshold stimulus. A threshold stimulation is the
smallest amount of stimulation that will actually result in a contraction. A
minimum amount of stimulation is needed to induce a muscular
contraction. We can stimulate the muscle with an external power source. By
gradually increasing the amount of stimulus strength, we can find the
threshold level of stimulation.
4. The Double Right Angle Clamp will ensure to support the gastrocnemius
muscle in the whole experiment. Adjust the thread tension by moving the
transducer up or down, until the lever is horizontal. Make sure that the
thread is aligned at a 90 degree angle to the transducer lever. It holds and
provides support to the rod that contains pen allowing the pen to write on
the graphing paper in your kymograph. In the vertical bar those clamp are
use to hold tranduscer rods, and another rod for holding the frog muscle
usually gastronecmius muscle
