ANALISIS Protein

1. P.K. N total cara Gunning


- Timbang 0,7 – 3,5 g bhn (dihaluskan), mskk
dlm labu Kjeldahl, + 10 g K2S atau Na2SO4
anh, + 15 – 25 ml H2SO4 pkt (Kl perlu di+ 0,1 –
0,3 g CuSO4) dikocok.
- Panaskan pd pemanas listrik atau api bunsen
dlm lemari asam dg api kecil. Setlh asap
hilang api dibesarkan, panaskan sp lrtan
jernih.
- Buat percobaan blanko (tanpa sampel).
- Setlh labu + cairan dingin, + 200 ml aq dem +
1 g serbuk Zn + lrtan NaOH 45% sp cairan
bersifat basa. Sgr pasang labu pada alat
destilasi.
- Panaskan labu sampai amonia menguap
semua, destilat ditampung dlm Erlenmeyer
yg berisi 100 ml HCl 0,1 N + indik fenol merah
atau metil merah.
- Destilasi diakhiri setlh volume destilat 150 ml atau sp destilat
tak bersft basa.
- Keleb HCl 0,1 N dlm destilat dititrasi dg lrtan baku NaOH 0,1N.

Perhitungan:
% N = (ml NaOH blanko – ml NaOH sampel) X NNaOH x 14,008
g sampel x 10

% protein = % N x faktor (lht tabel)


Konversi dari kadar N menjadi kdr protein berbagai macam bhn

2. P.K. Protein dlm susu cara Formol
- Pipet 10,0 ml susu atau cairan protein ke dlm Erlenmeyer 125
ml + 20 ml aq dem + 0,4 ml lrtan K-ox jenuh (K-ox : air = 1:3) +
1 ml p.p. 1%. Diamkan 2 menit.
- Titrasi lrtan dg NaOH 0,1 N sp wrn spt wrn standard di bwh ini
atau sp merah jambu.
- Warna standard: 10,0 ml susu + 10 ml aq dem + 0,4 ml K-ox
jenuh + 1 tts 0,01% indik rosanilin-chlorida (lamp 17).
- Setlh wrn tercapai + 2 ml lrtan formaldehid 40%, titrasi
kembali dg lrtan NaOH p wrn spt wrn standard lagi.
- Catat volume titrasi kedua.
- Titrasi blanko: 20 ml aq dem + 0,4 ml lrtan K-ox jenuh + 1 ml
ind p.p. + 2 ml lrtan formaldehid, titrasi dg NaOH.
- Titrasi terkoreksi: titrasi kedua dikurangi titrasi blanko = titrasi
formol.
- Utk mengetahui % protein dibuat percobaan serupa dg
menggnk lrtan yg tlh diket kdr prot nya (mis dg cara Kjeldahl)
- Untuk susu dapat digunakan faktor 1,83:

% prot susu = 1,83 x ml titrasi formol

% kasein = 1,63 x ml titrasi formol

% N = titrasi formol x NNaOH x 14,008/g bahan x 10
