THE AUTHOR: IFBB PRO LEO

M AY R H O F E R

My name is Leo Mayrhofer. I was born in 1992. I am an IFBB Pro Bodybuilder and have
been coaching and personally training people (from “regular Joes” to Mr. Olympia
competitors) for over a decade now.
I started self-studying the human anatomy, biochemistry, nutrition and training science rst
by myself, then a few years at a German University until I moved to the States at the very
end of 2019.
Of course my curious mind keeps me studying, learning and looking at data, outcomes
and new information constantly- including what I observe with my clients, colleagues and
myself.

I started serious training in the gym at the age of 18. I have always been trying to absorb
every knowledge I can gather to improve and get more ef cient at what I do every day.

Until the age of 25 I stayed natural and

competed in natural bodybuilding shows. By
now, I have done over 30 shows in total of
which more than 10 were IFBB Pro Shows.

The most prominent names in the bodybuilding

industry I coached and trained are Urs
Kalecinski (about two years, turning pro and
rst pro-show), 2x female bodybuilding
champion and vice Ms. Olympia Helle Trevino
and IFBB Pro Dr. Thomas Connelly.

Currently, I work at Gold s Gym Venice as an

independent Personal Trainer, I am the host of
the Overcome Podcast (Youtube, Rumble,
Spotify…) and I coach a few clients online (as
I am very responsible and want everyone to
get the full experience and attention, I dont
take on too many!).

WHAT ARE PEPTIDES & WHY USE

THEM?

Peptides are short chains of amino acids, typically ranging from 2 to 50 amino acids. They
function as metabolic signaling messengers in the body, interacting with speci c receptors
on cells to regulate various physiological processes. By acting as signaling molecules,
peptides in uence hormone release, metabolism, immune function, and tissue repair.

Their speci c sequences allow them to trigger targeted effects, such as promoting muscle
growth, enhancing fat burning (lipolysis), or aiding in healing. Unlike larger proteins,
peptides provide a more focused approach with fewer side effects, making them essential
for many metabolic functions.

So why would someone want to use peptides? I guess that is pretty obvious when looking
at all the properties described before.

If dosed correctly (current data and all my anecdotal experience with athletes and
colleagues shows) most peptides have essentially no side effects while providing good
bene ts. The most prevalent side effect amongst peptides is nausea. But only a few
actually could induce nausea, so looking at most pro les I created in this book you will see
that only a certain, small group of them have shown to be able to cause nausea, with
those I would recommend any user to start with a very low dosage and adjust slowly
according to personal tolerance.

PEPTIDES AND STEROIDS IN

SYNERGISM
APPROAC H TO GROW TH WITH HEALTHIER
ALTERNATIVES

Peptides and anabolic steroids can work synergistically to enhance muscle growth and
performance. Steroids, like testosterone, boost protein synthesis and nitrogen retention,
creating an anabolic environment. Peptides, such as growth hormone-releasing peptides
(GHRPs) like Ipamorelin or CJC-1295, amplify this by stimulating natural growth hormone
(GH) release, which enhances recovery, fat metabolism, and muscle repair. Together, they
amplify gains—steroids provide the raw anabolic power, while peptides ne-tune GH and
IGF-1 (insulin-like growth factor-1) pathways for sustained growth and reduced catabolism.

Beyond anabolics, peptides offer pathways to grow without the heavy androgenic load of
steroids, which can strain organs and disrupt hormonal balance. For instance, BPC-157
promotes tendon and ligament healing, while TB-500 boosts tissue regeneration—both
support training intensity without androgenic side effects. Unlike steroids, which rarely
offer health bene ts and often increase risks (e.g., liver toxicity, cardiovascular strain),
many peptides provide therapeutic perks: BPC-157 aids gut health, and GH peptides
improve sleep and skin quality.

By leaning on peptides, athletes can reduce reliance on steroids, lowering androgenic

stress and fostering longevity. Pairing these with resistance training, nutrition, and recovery
optimizes growth while keeping health in check—proving you don t need to max out on
anabolics to break plateaus.

THE MOST COMMON, MILD SIDE

EFFECT AND FIX

Several peptides, particularly those that in uence growth hormone (GH) secretion,
in ammation, or immune responses, can cause histamine release as part of their biological
activity. Histamine is a naturally occurring compound in the body, involved in immune
responses, in ammation, and as a neurotransmitter. When peptides stimulate the immune
system or affect blood vessels, histamine can be released, leading to symptoms like
ushing, redness, or itchiness.

Here are the major peptides that may cause histamine release or similar reactions:

CJC-1295 (with or without DAC)

GHRP-6
GHRP-2
BPC-157
Melanotan II
PT-141 (Bremelanotide)
Thymosin Alpha-1
IGF-1
Hexarelin

Why Do Peptides Cause Histamine Release?

• Histamine release from peptides usually happens because of their immune-

modulating effects or their in uence on the vascular system. Some peptides
can increase the permeability of blood vessels, which can lead to uid leaking
out of blood vessels into surrounding tissue, causing redness or swelling (like in
allergic reactions).
• Other peptides may activate mast cells (which release histamine) or directly
stimulate the immune system, leading to the release of histamine as part of the
body's response to the peptide.
fl
fl

fl
[email protected] 21 Jul 2025

Managing Histamine Release Symptoms:

• Lowering the dose: If you notice ushing or other symptoms after taking a
peptide, reducing the dose might help.
• Taking antihistamines: Over-the-counter antihistamines can sometimes help
manage mild histamine-related symptoms. A low dose of Cetirizine should resolve the
problem.
• Changing injection sites: Injecting peptides into different areas or rotating
injection sites can help reduce localized reactions.

CONCLUSION

If the ushing isn t mild and majorly decreasing after 20-30min without the use of an
antihistamine you either overdose or should not keep using the certain peptide that caused
it. Mild ushing can be resolved as described above.

CAUTION! LONGTERM
CONSUMPTION AND OVERDOSING

Peptides have become a popular tool for those looking to accelerate recovery, improve
performance, or enhance overall wellness. However, it s important to understand the risks
that come with their use, especially when consumed in excess. One of the main concerns
with peptides is that they trigger a cascade of growth mechanisms in the body. While a
peptide like GHRP is designed to stimulate the release of growth hormones for tissue repair
and muscle growth, it doesn't just affect the targeted pathways. In fact, it can activate
other growth factors that aren't always bene cial. These unintended growth signals can
lead to unpredictable side effects that might not show up right away, making them dif cult
to diagnose early. Over time, these side effects can have a negative impact on your
health, potentially contributing to things like tumor growth or abnormal cell proliferation.

Another major issue with many peptides is that they stimulate collagen synthesis—a key
component for tissue repair and recovery. While collagen is necessary for healing, an
overproduction can cause problems. If collagen is produced in excessive amounts,
especially in areas like the skin, joints, or internal organs, it can lead to brosis. This results
in scar tissue buildup, which can make tendons and ligaments overly stiff or cause them to
lose exibility. Over time, excessive collagen can lead to pulmonary brosis, cardiac
brosis, or joint stiffness, which may not show immediate symptoms but can cause serious
long-term issues.

Peptides can certainly provide bene ts for growth and recovery, but the effects aren't
always as targeted as you'd expect. When overconsumed, they can lead to unintended
consequences like excess growth factor activation or collagen overproduction, which may
not be apparent at rst but can have lasting and potentially harmful effects on your health.
It's essential to be cautious and consult with healthcare professionals to make sure you're
using peptides in a safe and effective way.

HUMAN GROW TH HORMONE

Human Growth Hormone (HGH) is a hormone naturally produced by the pituitary gland,
and it plays a crucial role in growth, cell regeneration, and maintaining healthy tissues,
including muscles and bones. It has become popular in bodybuilding and performance
enhancement for its potential bene ts, but it is important to understand the complexities,
including its use, dosages, sources, and the potential side effects.

HGH AND PERFORMANCE ENHANCEMENT:

Athletes, bodybuilders, and those seeking to improve physical appearance or

performance often use HGH for its anabolic (muscle-building) properties and recovery
bene ts. It is believed to promote fat loss, increase muscle mass, improve strength, and
enhance recovery. However, the use of HGH for performance enhancement is illegal in
most sports, and its use without medical supervision can be risky.

Generic vs. Pharmaceutical HGH

-Pharmaceutical HGH (Brand-name): These products are FDA-approved, typically

manufactured by major pharmaceutical companies like Genentech (Saizen), Novo Nordisk
(Norditropin), Eli Lilly (Humatrope), and others. They are produced using recombinant
DNA technology and are highly regulated to ensure purity and effectiveness.

Pharma HGH is generally considered to be safer and more reliable in terms of quality
and dosing compared to generic versions. These are used in medical treatments like
growth hormone de ciencies, Turner syndrome, and adult HGH de ciency.

Generic HGH: These are often produced by smaller, less regulated companies, and while
they may contain similar or the same active ingredient, their quality control, ef cacy, and
safety standards may not be as stringent as pharmaceutical-grade HGH. Generic HGH is
often sought after because it may be less expensive, but it carries a higher risk of
impurities, incorrect dosing, or suboptimal results. If it is tested though a unit of generic
HGH is just as potent as a unit of pharmaceutical brand-name HGH.

PHARMACEUTICAL HGH PRODUCTS:

Some well-known pharmaceutical HGH products include:

1. Saizen (Genentech)

2. Norditropin (Somatropin) (Novo Nordisk)

3. Humatrope (Eli Lilly)

4. Omnitrope (Sandoz)

5. Tev-Tropin (Teva Pharmaceuticals)

These HGH products are used medically to treat growth hormone de ciencies and other
conditions but are also commonly used off-label for performance enhancement and anti-
aging purposes.

E F F E C T I V E D O SAG E S F O R B O DY B U I L D I N G :

The use of HGH in bodybuilding and performance enhancement varies, but some common
approaches include:

-Low-dose therapy: 1–2 IU (International Units) per day (good levels within very healthy
natural range)

- Used for maintaining muscle mass, fat loss, and general wellness. This is generally seen
as the safest approach.

- Moderate-dose therapy: 3–4 IU per day

- Often used for muscle building, fat loss, and recovery, especially in bodybuilders or
athletes looking for more signi cant improvements (slightly to moderate supernatural
levels)

- High-dose therapy: 5–10 IU or more per day

- This dose is used by those seeking maximal muscle gain and recovery, though it carries
a higher risk of side effects and is not generally recommended for long-term use. It is not
yet established if hyperplasia, the building of new muscle bers can be induced by taking
high levels of HGH for a prolong period of time but the rumors are out there.

It's important to note that the dose is usually tailored to the individual s goals, experience,
and tolerance to HGH. Dosage cycles may vary, with some users taking HGH for months
on end, while others use it in shorter cycles to avoid long-term side effects.

Controlling blood sugar levels become very crucial at higher dosages. Common practice is
using most of the amount used at night time with taking Berberine and a ber supplement
to keep insulin sensitivity normal.
Many also add Berberine to most meals that contain carbohydrates. One is also well

advised to focus on not overeating carbohydrates and also in an offseason scenario not to
eat more than needed and rather create a caloric surplus by eating more protein.

ENDOGENOUS HGH PRODUCTION:

At an age of 15–25 years old natural HGH production is usually at its peak and varies
signi cantly throughout life. From age 15 to 25, normal HGH secretion can range from 1
to even 3 IU per day in rare cases.

HGH production is highest during puberty when growth is most rapid. The secretion
decreases with age, typically slowly decreasing after the age of 30.

HGH is released in pulses, with the highest secretion occurring shortly after sleep onset
(during deep sleep) and at various times throughout the day.

WHEN TO ADMINISTER AND WHY:

HGH is typically administered subcutaneously in the medical eld (under the skin). In the
bodybuilding realm rather intramuscularly when done multiple times a day to prevent
blood sugar problems. The timing of HGH administration can impact its effectiveness:

Best time for administration:

HGH is released in pulses during sleep, and taking HGH 30 minutes or directly before bed
can maximize results by improving recovery and growth because it will increase time of
deep sleep. This is one of the most common strategies for enhancing growth and recovery.

Pre-workout: Some users prefer taking HGH pre-workout to optimize the fat burning
properties that HGH carries.

In cycles: Some people use HGH in cycles, such as 4–6 months on and 2–4 months off, to
avoid tolerance and mitigate side effects. When controlled perfectly and blood sugar
levels are stable it makes no sense to get off HGH when maximizing bodybuilding results is
the main goal.

BENEFITS OF HGH USE:

HGH is believed to provide several bene ts, especially in the context of performance
enhancement and bodybuilding:

1. Increased Muscle Mass: HGH promotes the growth of lean muscle by stimulating protein
synthesis and increasing the production of collagen in muscles and tendons. But this impact
is very minor and using it by itself won t be even close to as effective as a low dose
testosterone cycle.

2. Fat Loss: HGH has been shown to enhance lipolysis (fat breakdown), helping reduce
body fat, especially abdominal fat.

3. Improved Recovery: HGH accelerates tissue repair and recovery, reducing muscle
soreness after intense workouts and speeding up the healing of injuries.

4. Skin Health: HGH helps improve skin elasticity and reduces wrinkles, making it popular
in anti-aging circles. In higher dosages it might even do the opposite though as very high
levels of IGF1 seem to be very pro-aging as the newest longevity research suggests.

5. Bone Density: HGH can stimulate the production of bone-forming cells, helping improve
bone density, which is particularly important in aging individuals.

SIDE EFFECTS OF HGH USE:

While HGH can offer performance bene ts, it also carries several potential side effects,
especially when used improperly or in high doses:

1. Joint Pain: One of the most common side effects is joint and muscle pain, particularly at
higher doses.

2. Carpal Tunnel Syndrome: Some individuals develop numbness, tingling, or pain in the
wrists or hands, which can be a sign of carpal tunnel syndrome.

3. Water Retention: HGH can cause water retention, leading to bloating and an increase
in blood pressure.

4. Insulin Resistance: Long-term use of HGH can lead to insulin resistance, which may
increase the risk of developing Type 2 diabetes. A good diet and management is crucial.

5. Acromegaly: Prolonged use of HGH, especially at very high doses, can lead to
acromegaly, a condition in which the bones and tissues enlarge abnormally.

6. Heart Issues: There have been concerns that long-term HGH use may increase the risk
of heart problems, by way of heart enlargement and high blood pressure.

7. Increased Risk of Cancer: HGH can stimulate the growth of certain tissues, including
potentially abnormal cells, raising concerns about cancer risk, particularly in those who
are predisposed to cancer. It is very unlikely to stimulate cancer occurrence but it will
de nitely increase the pace of spread and growth, if the individual already has cancer.

CONCLUSION:

While HGH has shown promise for muscle growth, fat loss, and performance
enhancement, it is not without signi cant risks. The distinction between pharmaceutical-
grade and generic HGH is crucial, as the former is generally safer and more reliable, but
if tested and pure Generic won t be less effective. For those considering HGH, moderation
and accessing tolerance is key, and it's advisable to consult with a healthcare professional
to monitor dosing and potential side effects.

Using HGH to enhance performance or appearance should always be done with caution,
and it s important to balance the potential bene ts with the risk of adverse effects,
especially over long periods of use.
