providing the necessary materials and encouragement.
Objective
To determine and compare the caffeine content in different brands of tea and coffee using
simple extraction and analysis techniques.
Theory
Caffeine is a naturally occurring stimulant found in coffee beans, tea leaves, and certain
soft drinks. Chemically, it is an alkaloid of the purine group with the formula
C8H10N4O2.
It acts as a central nervous system stimulant and temporarily wards off drowsiness and
restores alertness.
Caffeine is slightly soluble in water but highly soluble in organic solvents like
chloroform and dichloromethane. It can be extracted using these solvents from beverages
like tea and coffee.
This principle of differential solubility forms the basis of this experiment.
Materials Required
- Tea leaves (2 brands)
- Coffee powder (2 brands)
- Distilled water
- Separating funnel
- Beaker, funnel, filter paper
- Dichloromethane (or ethyl acetate/chloroform)
- Evaporating dish
- Water bath
- Weighing balance
Procedure
Step 1: Preparation of Tea/Coffee Solution
1. Boil 100 mL of distilled water.
2. Add 5 g of tea leaves or coffee powder.
3. Boil for 5 minutes.
4. Filter the solution.

Step 2: Extraction of Caffeine


1. Pour the solution into a separating funnel.
2. Add 20 mL of dichloromethane.
3. Shake gently and allow layers to separate.
4. Remove the organic layer.
5. Repeat the extraction.
6. Combine all organic layers.

Step 3: Evaporation
1. Transfer the combined organic extracts to an evaporating dish.
2. Heat gently in a water bath.
3. Dry and weigh the remaining caffeine.

Calculations
Caffeine content per gram = Extracted caffeine (mg) / Sample used (g)
Example for Coffee Brand A:
= 52 mg / 5 g = 10.4 mg/g
Result
- Coffee contains more caffeine than tea.
- Brand A has more caffeine in both categories.
- The method is efficient for qualitative comparison.
Conclusion
This project successfully demonstrates the extraction and comparison of caffeine from tea
and coffee. Caffeine content varies depending on the brand and type of beverage. Such
experiments are helpful in understanding organic compound isolation techniques.
