Objective:
Objective: Reference guide to operate SimplicityChrom™ Software.

Scope: Perkin Elmer LC equipped with SimplicityChrom Software.

Responsibility:

 Analyst/ Chemist are responsible for proper use and implementation of SOP.
 Group Leader and Head of the department shall ensure implementation of SOP.

Instrument Procedure:

 Start)
Start)up
A. Switch on the instrument
a. Switch on the computer.
b. Switch on the power button of the LC instrument.
c. Install LC column and Set up the system.
d. Keep degassed Mobile phase bottle on solvent reservoir and set up the system.
e. Condition the column 30 minutes prior to analysis.

B. Handling of the software


a. Click on the SimplicityChrom icon in desktop.

b. following window will appear. Select the project folder.

 Analysis:
A. Project creation:
a. For new project creation click on Lab Management.

b. Following add project
project winow will appears, Click on Add Project. Then press Next button.

c. Type applicable project name and description. Then press next button.

d. Click on Add instrument, select instrument, and press finish button.

B. Add instrument
a. Click on Lab Management tab in Home and Click on Instrument Systems.

b. Select add system from top left corner, following window will appear.

c. Add HPLC Pump, HPLC autosampler, Column Oven, applicable detector from add instrument option and LC 300 Display from
add display option.
d. Click on communication setting for all components and check the connection by selecting the Test Connection.

e. Click on Save and enabled the instrument by clicking on connect from top right corner.

C. Acquisition Method Creation.


Creation.
a. Select the system from browse system and take control.

b. Once the instrument in locked. Click on Acquisition Panel. Following window (Workspace) will appear.

c. Modify the listing default method parameters for the selected instrument.

d. In the Acquisition panel toolbar, click the Save icon. The Save Acquisition Method page is displayed.
e. Enter a name for the acquisition method and then click Save.

D. Sequence Creation
a. Navigate to Sequence panel. Click on New and one empty sequence grid will be displayed.

b. Enter the desired value and details in appropriate grid. Click on save. The save Sequence page will be displayed.
c. In the Name box enter the desired name for the sequence then click on save.

E. Starting the Sequence and Editing the Sequence


a. Once the Sequence is created and saved start the sequence by pressing start icon.

b. Run queue panel will be displayed. In Run queue panel sequence can be paused and editing.

c. After editing run can be start by pressing start/Resume button.

F. Data Integration

a. `Open the acquired data by clicking on open data icon and select the acquired data.

b. Click on chromatogram panel to obtain the chromatogram plot. In chromatogram plot click on edit integration and select the
desired integration event to integrate peak.

c. To label the peaks open processing method panel. In processing method panel click on compound table and add peak name,
assign RT and its window.

G. Linearity Calculation/Y)
Calculation/Y)Intercept
a. For Linearity Calculation open the processing method. In compound table add the number of levels used in linearity sequence.

b. In compound table number columns will generated which will be equal to number of levels were added. Enter the appropriate
concentration of linearity levels.
c. Click on Result set review panel and enter the sample type standard for linearity levels. Enter the level in result set review
column.

d. Click on Calibration curve panel.

H. Report generation.
a. Click on the print option, following window will appear

b. Select the desired template report and click on right corner icon to print report.

I. After completion of analysis switched off the instrument for safety purpose.
