Calculation of protein concentration of 3 samples for Biuret and Bradford


Assays

Biuret :

Using the trend line equation given in Figure 1, y = 7E-05x, we can find the
concentration of protein by substituting y with the Absorbance. Using this equation
for biuret samples

Sample 1: y = 7E-05x

0.024 = (7x10-5 ) x

X = 343 µg/ml

Sample 2: y = 7E-05x

0.0415 = (7x10-5 ) x

X = 593 µg/ml

Sample 3: y = 7E-05x

0.0615 = (7x10-5 ) x

X = 879 µg/ml

Bradford :

Using the trend line equation given in Figure 2, y= 0.0022x, we can find the
concentration of protein by substituting y with the Absorbance. Using this equation
for Bradford samples

Sample 1: y = 0.0022x

1.514 = 0.0022x

X = 688 µg/ml

Sample 2: y = 0.0022x

1.569 = 0.0022x

X = 713 µg/ml

Sample 3: y = 0.0022x

1.648 = 0.0022x

X = 749µg/ml
