Incubate culture for 24 hrs at 37 ̊C

After 24 hrs incubation of plate at 37 ̊C


Growth

Pick colony from quadrant 4 and perform Gram stain ( All stain and alchohol 1 min and Iodine 3 min)
Identification – GRAM NEGATIVE BACILLI (GNB) GN card

GRAM POSITIVE COCCI (GPC) GP card

YEAST YST card

ANAEROBES AND CORYNEBACTERIA ANC card

NEISSERIA AND HAEMOPHILUS NH card

GRAM POSIVE BACILLUS SPECIES BCL card

Antibiotic Sensitivity GNB

GNB AST N280 card (for LF colonies)

AST N281 card (for NLF colonies)

GPC AST GP 628 card (Staphylococcus & Enterococcus)

AST ST03 card (Streptococcus)


YST AST YS08 (for Yeast AST)
Vitek 2 Compact

A. Bench Work
1. Take 2 plastic tubes (no.1 and no.2) for I isolate and place it on cassette
2. Take 3ml suspension solution in each tube from dispensette.
3. Mark tube 1 as ID tube ( for identification), and mark tube 2 as AST tube
(for sensitivity)
4. Pick similar isolated colonies from Quadrant 4 from the plate, and make
a suspension in tube no.1.
5. Fix the turbidity of tube no.1 upto

Bacteria (GN & GP) : 0.50 – 0.63 Mcfarland

Yeast and BCL : 1.8 – 2.2 Mcfarland

NH and ANC : 2.7 to 3.3

6. Transfer 145 µl (for GNB) or 280 µl (for GPC and YST) from tube no.1 to
tube no.2
7. Place ID card in tube no.1 and AST card in tube no.2
1. Enter; Patient ID= Lab ID; Name = Patient; Location = Patient location;
Physician = Dr’s Name ; Lab ID = Lab ID = ACCESSION ; Type = Bench
Name ; Source = Sample Type
2. Click “OK”.

3. Click “ Isolate View”.


4. Set on left view bar; View By = Date Tested
5. Filter By = Show all
6. Click on the current date folder on the navigation tree.
7. Click on the Isolate no. or name on the navigation tree.
8. On active the report will appear.
1. For Print out

Click on “print” icon (Top 2nd last Icon)

Uncheck Lab Report and Check Lab Report.

Click “Print All”.

2. For AST Offline test (ALL GPC Results)


Staphylococcus aureus; Benzylpenicillin MIC >=0.5; β-lactamase test +
Staphylococcus spp. ; Benzylpenicillin MIC >=0.25; β-lactamase test +
For all others put β-lactamase test –
3. For blocking Antibiotic from report list
Click on the box in attached to the antibiotic name on the list on screen
4. Report Review

Click on review Icon (Top right 2nd Icon)


5. Report Approve
Log Out of current username password
Log in by microbiologist Username Password
Go to the report through Isolate View.
