USING THE MICROSCOPE AND EXAMINING CELL STRUCTURES

PURPOSE: 17
 to become familiar with the characteristics of images produced by microscopes
 to examine a typical plant / animal cell and draw a labelled biological diagram

MATERIALS:
 microscope  “e” slides
 prepared slide of generalized animal cell  prepared slide of generalized plant cell

PROCEDURE:

1. Place the LETTER “e” face up on the stage. Draw the letter as it appears with the
naked eye and on LOW POWER under the microscope. [2]

NAKED EYE MICROSCOPE (LOW)

2. How has the position of the letter changed when viewed with the microscope? [1]

3. View the letter “e” under MEDIUM and HIGH POWER. Draw what you can see. Be sure
to put the letter in the middle of the field of view before changing from medium to high
power. [2]
MEDIUM HIGH

4. Return to LOW POWER and adjust using the coarse adjustment knob. Move the
letter “e” slide as described below and record the movement of the image. [4]

5. Obtain a slide of the ANIMAL CELL and place it on the stage of the microscope. Adjust
the focus on low power using the coarse adjustment knob to view specimen.

6. Rotate the revolving nosepiece until the medium power objective clicks into place.
Adjust the focus using the fine adjustment knob to view specimen.

7. Repeat step 6 using the high power objective.


*Use extra caution when adjusting the focus to prevent slide from cracking.

8. When you are done, turn the revolving nosepiece so the lowest power objective lens is
in place, and remove slide from the stage.

9. Repeat steps 5-8 using the slide of plant cell.

10. Make a proper biological drawing of ONE PLANT CELL under HIGH POWER ON THE
BACK of this handout. Review rules for biological drawings!!!
