EXPERIMENT NO.9
COLLECTION, PACKING AND PRESERVATION OF
SEMEN AND SEMEN STAINED CLOTHING

I. Objective: To learn the proper technique of packing and preserving

seminal fluid and semen stained clothing.

II. Materials and Reagents: Semen stained clothing (under clothing

or bed clothing) cardboard or similar materials, paper bag or a large
box, tying material, chalk and 10%formalin solution.

III. Discussion:
One of the most common physical evidence to be found in connection
with sexual offense like cases of adultery, rape and bestiality is the semen
and seminal stain. The search for this evidence should be the first objective
of the investigator. Semen maybe found fresh in the vaginal or rectal
contents of the victim or it may be noted as dried secretion on the hand skin
around the genitals or sometimes present as dry stains or under clothing,
bed clothing and other objects.
The seizure of the wearing apparel of the victim and suspect must be
made as soon as possible because it has often happened that an important
trace has been destroyed by the washing of the clothes and in dried
condition semen are easily cracked, chipped away and lost.
Semen is a type of perishable evidence that requires special
preservative to maintain its evidential values. During hot weather where
there is danger of putrefaction, seminal fluid maybe preserved by a few
drops of a 10% solution of formalin. Since these items of evidence are
perishable and easily putrefy great care must be exercised in handling and
packing.

IV. Procedure and Observation:

I. Semen Stained Clothing
a. Encircle with chalk the area of the clothing in which the stain was
found. Mark with date and initial as far away from stained area as
possible. Air dry the stain if still moist.
b. Place gently the clothing bearing suspected semen stains
between two sheets of cardboards or similar material and tie
together. The specimen should not be rolled on packing.
c. Place in a breathable container as paper bag or large box the
packed clothing.
d. Label the container by inscribing thereon the case information.
Observation:
1. Why is moist air dried before packing?

2. When packing, semen stained clothing should not be rolled.

Why?

3. What is the purpose of encircling with chalk the stained areas of

the clothing?

II. Fluid Semen

If fluid semen is found place it in a test tube and add a few drops
of 10% solution of formalin and then cover it with cork. Seal with
masking tape in such manner that it cannot be opened without
breaking the seal. Label or tie a tag to the test tube bearing the
initials or name of the investigator and the date of the sealing
(abbreviated form).

Observation:
1. Why is seminal fluid preserved by a few drops of a 10% solution
of formalin especially during hot weather?

V. Conclusion:
1. In the search for semen and seminal stain evidence where you
can find semen as fresh?

2. Where can you find it as a stain?

3. What is putrefaction?

4. Explain: Seminal stain is perishable.
