Aim: To Prepare Paracetamol granules by wet granulation method

Requirements:
1. Chemicals: Paracetamol, Acacia, Lactose, Starch paste, talc,
magnesium stearate, distilled water.
2. Glasswares: Measuring cylinder, beaker, mortar and pestle,
granulating sieve, spatula.
3. Instrument: Hot air oven/Tray Dryer
Theory:
Paracetamol tablet belongs to a group of medicines called analgesics
and it is used to treat pain.
Granulation is the process of formation of granules from a powdery
or solid substance, producing a granular material. In wet granulation,
granules are formed by the addition of a granulation liquid onto a
powder bed.
The granulation liquid contains a solvent or carrier material that must
be volatile so that it can be removed by drying, and depending on the
intended application, be non-toxic.

Procedure:
1. Weigh and pass paracetamol powder through sieve no 44.
2. Mix paracetamol and starch powder uniformly in mortar and
pestle.
3. Prepare 15% starch paste in boiling water and stir until it
becomes translucent.
4. Add starch paste dropwise in mortar to get cohesive mass.
Record quantity of starch paste used for granulation.
5. Screen prepared cohesive mass through sieve no 16 and collect
it on granulating tray.
6. Dry granules in tray at 50oC for 30 minutes. Pass the dried
granules through sieve no 22 to get uniform particle size.
Result: The Paracetamol granules were prepared by the wet
granulation method and stored in well closed container.
