DNA ANALYSIS ASSIGNMENT

SBI4U

Several types of mutations can affect an organism's genes. The extent of the effect varies
widely, from no difference to the production of a dysfunctional protein, or no protein at all. In this
assignment you will be demonstrating how gene mutations affect the production of proteins.

PART A: [12TI, 14A]

1. Your teacher will assign you a DNA template strand. Draw a data table similar to Table 1,
using correct table formatting, in your notes and record the wild type sequence of the template
strand.

2. Write out the mRNA sequence that would be created by transcription of the original wild type
template. /2T

3. Translate the mRNA from step 2 into protein. Record the sequence of amino acids in your
polypeptide chain. /2T

4. Perform each of the following changes to your original DNA sequence. Transcribe each new
sequence and translate it. Record the resulting amino acid sequence. Always compare back to
the original wild type protein to see the impact of mutation. /4T
a) Replace the 3rd T with a G.

b) Delete the 7th base.
c) Insert an A between the 11th and 12th bases.
d) Delete the 8th, 9th and 10th bases.

5. For each change you made in step 4, identify the type of mutation that occurred and the effect
of that mutation on the polypeptide chain (8A)

6. What would happen if a nucleotide was deleted in each of the following regions of a gene?/6A
a. the promoter region
b. an exon
c. an intron

PART B – Communication /12

Research a genetic disorder that is caused by a substitution, addition or deletion mutation.

Find out which gene is involved, and how the protein is altered as a result. Describe the effect of
the disorder on an individual's health and lifestyle. Consider the cost of treatment.

You will be marked with the following checklist:

a. Naming a disorder that is caused by a substitution, addition or deletion mutation (1C)
b. Naming the gene that is involved, and how the protein is altered as a result (2C)
c. Describing how the disorder impacts an individual's health and or lifestyle (3C)
d. A brief description of the cost of treatment (1C)

You will also be given a communication mark of 5 marks for your use of scientific terminology,
proper spelling and grammar and organization.
References must be embedded and in APA format.
