COLLECTION, PACKING, AND PRESERVATION OF SEMEN AND
SEMEN STAINED CLOTHING
INTRODUCTION
Semen, also known as seminal fluid, is an organic fluid that may contain spermatozoa. It is secreted by
the gonads (sexual glands) and other sexual organs of male or hermaphroditic animals. It can fertilize
female ova. In humans, seminal fluid contains several components besides spermatozoa such as
proteolytic and other enzymes, fructose, These are elements of seminal fluid which promote the
survival of spermatozoa, and provide a medium through which they can move or "swim".
On the other hand, seminal stain is a stain left on surface of under clothing and
bed sheet after sex. Both semen and seminal stain are perishable. In this experiment, you will learn
the proper way to collect, pack and preserve semen and semen stain evidences.
I. OBJECTIVE:
To perform the proper technique to collect, pack and preserve seminal fluid and semen stained
clothing.
Ii. MATERIALS/REAGENTS
1. semen stained clothing (under clothing or bed clothing)
2. cardboard or similar materials
3. paper bag or large box
4. tying material, labeling pen
6. chalk, test tube with screw cap, droppers
7. 10% formalin solution
III. PROCEDURE:
Construct a crime scene showing the presence of semen stained clothing and fluid semen. Show by
taking pictures, the steps in examining a crime scene and the proper collection, packing, and
preservation of fluid semen and semen stained clothing evidences following the procedures below.
EXPERIMENT NO. 8
1. SEMEN STAINED CLOTHING
a. Encircle with chalk the area of the clothing in which the stain was found. Mark with date and initial
as far as possible. Air dry the stain if it still moist.
b. Place gently the clothing bearing suspected semen stains between two sheets of cardboards or
similar material and tie together. The specimen should not be rolled on packing.
c. Place the packed clothing in a breathable container like paper bag or large box
d. Label the container by inscribing thereon the case information.

The first thing we did was codoned the area of the incident then we started to investigate the area
for the purpose of in oder to identify and locate the possible dry stain of semenal fluid in the clothing
/bedsheet of the victim’s bed inside her home

when we are able to identify and found the possible dry stained of seminal fluid on the bedsheet of
the victims bed we took photographs and sketch as part of a legal procedure of the investigation ,we
found out it was not yet completely dried so we first wait for 15 to 30 minutes inorder for it to
completely air dry , after that we encircle the area of dried stain using a chalk and we cut the part or
area of bed sheet slowly to avoid rolling or damaging of the collection of specimen of the dry seminal
fluid on clothing or bedsheet of the victim that contains the area encircled of dry seminal fluid.

After that for the collection and preservation of dry stained seminal fluid we put the sample specimen
of dried stain of seminal fluid from the piece of bedsheet and Place gently the clothing bearing
suspected semen stains between two sheets of cardboards and tie it together safetly.

Afrter that we place the sample collected inside a paper bag to avoid moisture,contamination or
growth of bacteria on the specimen sample collected and labelled it .

2. FLUID SEMEN
If flaid semen is found, place it in a test tube and add a few drops of 10% solution of formalin and
then cover it. Seal with masking tape in such a manner that I cannot be opened without freaking the
seal Label or tie a tag to the test tube she bearing the case mumber, initials of name of the
investigator and the date of collection.

During the investigation,we found a fresh seminal fluid in liquid form or drops of seminal fluid on the
floor, we first took a photograph and sketch on it.
After tht then we took and collected the fresh seminal fuid samples using a clean dropper and place
it inside the container using the test tube, then we add a few drops of tuluol or 10% of solution of
formalin in order to preserve it , then we cover it and seal with a plastic tape in order for the sample
specimen to avoid leaked or contamination and in oder for it be preserve well and properly for the
safety and security of the sample specimen.

of the investigator and the date of collection.

1. Seizure of wearing apparel must be done as soon as possible. It often happened that important
traces has been destroyed by washing the clothes, chemise, panties, trousers, skirts, are the most
common parts of wearing apparel carrying seminal stain.
2. In packing of wearing apparel there should be no friction between the apparel and the stain. The
packing of wearing apparel or objects carrying seminal stain must be made in such a manner that
there is no friction against the stain. Semen in dried condition is very brittle and is liable to break into
small particles which can be lost, Friction may cause the breaking of the spermatozoa
