Theory-
Micrometry-
It is a scale used to measure microscopic objects it
is expressed in micron.
The technique of micrometer evaluation is useful
for measurement of dimension of starch
grain,calcium oxalate crystals,fibres and cell
content in powdered or unground crude drugs.
The size of an object or part of it is can be
measured with help of 2 types micrometer
1-stage micrometer
2-eye piece micrometer
1-Stage micrometer:
It is a slide it contain a standard scale length of
1mm which is divided into 100 divisions
1 division of stage
micrometer=1/100mm=0.01mm
2-Eye piece micrometer
It is a circle of glass with a scale etched on the
surface. It is suitable for insertion inside the
ocular and during the operation of measurement.
A convenient form is a linear scale of 1mm divided
into 100 division
The value of its division varies with the
combination of eye piece and objective lenses.
Hence, calibration of one division of it is essential
during practical work it order to get exact value of
its one division in terms of microns
Callibration of one division of eyepiece
micrometer-
1-put the eyepiece micrometer on the
diaphragm of the ocular
2-put the stage micrometer on the
stage of microscope
3-See the lines of two micrometer
coincide count the number of lines
required to coincide
Characteristics of starch grain
-Rice starch
Nature- simple and compound
Shape-polyhedral with sharp angles
Size- 4 to 10u but average grains are about 6-4
compound grains may contain upto 150
components and may measure upto 30u in width
and 20u in length
Hilum and striations-no striations are visible and
only very occasionaly a hilum can be distinguished
as small dot.
-Potato starch
Nature-mostly simple but occasionally
Shape-subspherical or ovoid or often slight
flattened.
Size- Subspherical grains are upto 35u in length
and ovoid grains vary from 30 to 100u
Hilum and striations-The hilum occurs as a small
point situated towards narrow ends of grain and is
surrounded by well marked concentric striations.
Procedure
1-Calibrate an eyepiece micrometer using a stage
micrometer and calculate the factor.
2-Mount a little quantity of crude drug powder.
3-measure the diameter of isolated starch grain in
the mount by focusing them on the scales of
eyepiece micrometer. Note the number of
division covered by starch grains.
4-Calculate the dimension of 50 starch grains
multiply the division by the factor so as to get
actual dimension of starch grain in microns.
5-calculate the average value and give the range
for the dimension of starch grains.
Calculation-
1 division of stage micrometer=10um
3 division of eyepiece micrometer=5 division of
stage micrometer
now, 1 division of stage micrometer=10um
5 division of stage micrometer=5x10=50um
3 division of eyepiece micrometer=50um
1 division of eyepiece micrometer=50x1/3=16.66um
