Chloride Test Kit - TPTZ Method

This document provides instructions for using a photometric colorimetric test to determine the concentration of chloride ions in a sample. The test uses a mercury(II)-TPTZ complex that forms a blue complex with chloride ions, and the absorbance change at 590nm is proportional to the chloride concentration. The test is linear up to 500mmol/L chloride and samples over this concentration must be diluted. Reagents, quality controls, and performance characteristics are also described.

Chloride ions react with a mercury(II)-2,4,6-tri-(2-pyridyl)-s-triazine (TPTZ) c [mmol/l] = c [mval/l]
complex to form mercury(II)-chloride. The liberated TPTZ reacts with
iron(II) ions yielding a blue coloured complex. The resulting absorbance c [mmol/l] x 3.55 = c [mg/dl]
