Thank you for purchasing this product, the UV Performance Validation Software for the Shimadzu
UV-Visible Spectrophotometer (hereafter referred to as "this software").
This manual describes how to use this software. Read the manual thoroughly before using this
software and use this software in accordance with the manual's instructions. Keep this manual for
future reference.
This manual assumes that you are familiar with the basic operations of the Windows operating
system. For Windows operations, refer to the instruction manual and other documents included with
the relevant product.

Warranty
Shimadzu provides the following warranty for this product.

1. Period: Please contact your Shimadzu representative for information about the
period of this warranty.
2. Description: If a product/part failure occurs for reasons attributable to Shimadzu during
the warranty period, Shimadzu will repair or replace the product/part free
of charge. However, in the case of products which are usually available on
the market only for a short time, such as personal computers and their
peripherals/parts, Shimadzu may not be able to provide identical
replacement products.
3. Limitation of 1. In no event will Shimadzu be liable for any lost revenue, profit or data,
or for special, indirect, consequential, incidental or punitive damages,

This chapter explains the installation process relevant to this software (software and instrument
setups, starting and exiting software, etc.).

1.2 Installing Software 1
The following explains the installation procedure for this software.

This software is compatible with Windows 7, Windows Vista, and Windows XP operating systems (All
Edition 32-bit). Check the operating system running on your personal computer (hereafter called a "PC") 1
before installing this software.

1 Turn on the power to the PC. After Windows has started, insert the installation CD of
this software into the CD-ROM drive.

In the Windows XP operating system, the setup program starts automatically and the window
shown in step 4 appears. If the setup program does not start automatically, take the following steps
to start it manually. 1
This example assumes that drive "E" is assigned to the CD-ROM drive. For "E:\", substitute the

correct drive assigned to the CD-ROM drive of your PC.

1 Click (start), and then click [Run...].


The [Run] window appears.
2 Enter "E:\autorun.exe", and click [OK]. 1

2 When the [AutoPlay] window appears, click [Run autorun.exe].

If the [AutoPlay] window does not appear, take the following steps to start the setup program. This
example assumes that drive "E" is assigned to the CD-ROM drive. For "E:\", substitute the correct
drive assigned to the CD-ROM drive of your PC.

1 Click (Start).
2 Enter "E:\AutoRun", and press the "Enter" key.

4 Read the precautions and click [Next].

The following window appears if this software is found on your PC during a software upgrade.
Click [Remove] to uninstall the software.

5 Enter the appropriate information for [User Name] and [Organization], then click
[Next].

You may enter up to 50 characters for the [User Name] and up to 80 characters for the
[Organization].

6 Configure the instrument information.

1 Select [InstrumentType] of the instrument to be connected.


2 Enter [InstrumentName] and [SerialNumber].
3 Check that the settings and the entered data are correct, and click [Next].

In UV-2600/2700 series, the serial number is loaded when they are connected. Therefore the
serial number field is disabled. Also note that the [InstrumentType] selection cannot be
changed after installation.
Note that for UV-1600/2400PC/2500PC series, the information selected and entered for
[InstrumentType] and [SerialNumber] cannot be changed after installation.

7 Check the destination folder to install the software, and click [Next].

To change the destination folder, take the following steps:


1. Click [Change].
2. Select or enter the destination folder to install the software, and click [OK].

Folder securities for Windows XP or later versions are stricter than those of earlier versions. Therefore,
creating new sub folders or saving files in the destination folder may be restricted. To change the
destination folder used for installation, check the folder security settings in advance.

1 Double-click the shortcut icon on your desktop.

2 Select the account and enter the password, then click [OK].

For information about accounts, see "2.7.1 Restricting Functions Based on Account".

Logging in is possible immediately after installation without entering a password.

The software starts.


Immediately after installation, the software initially starts with a default (recommended) test item and
other conditions ( "8.1 Recommended Value List"). Subsequently, the software will start with the
conditions that you configured during the last operation.

1.4 Configuring Communication Port


Configure the communication port in the [Instrument Settings] window.
In this window, you can change the instrument name specified during installation, and check
information about the instrument currently connected.

You can only connect to the type of instruments that you configured during installation.
Do not connect to other instruments even if the type of the other instruments is the same as the ones you
configured. Each instrument has its own unique serial number.

See "2.1 Intended Instrument Types".

The following shows how to configure the communication port.


If your PC has two or more communication ports, check the number of the port to which the
communication cable is connected.

A UV-2600/2700 is connected via USB, but it uses the communication port inside the PC.
Refer to the "UV-2600/2700 Instruction Manual" and check the connected communication port number
using the device manager.

1 Click [Configure] from the [Instrument] menu.

2 From the [Communications Port] list, select the port to which the instrument's
communication cable is connected.

You can only establish communication with the type of instruments that you configured during installation.
Do not connect to other instruments even if the type of the other instruments is the same as the ones you

To simply view previous test results, communication with the instrument does not need to be established.

1 Turn on the power switch of the instrument (spectrophotometer).

2 Click [Connect].

When communication with the instrument has been established, the [Connect] button on the tool button
bar changes to [Disconnect], and [Start] is enabled (turns green).

Set the password required for logging in.
Note that the administrator account is required to set or change passwords.

1 Click [Administrative] from the [Security] menu.

2 Select the account to be changed, and enter a new password (enter it twice for
confirmation).
You may enter up to 30 alphanumeric characters. The password is case sensitive.

4 Click [Close].
The [Administrative] window closes.

2 2 Software Overview

This software tests the performance of the spectrophotometer using predefined procedures
(measuring parameters) and stores the results. This process is referred to as "validation" in the
software and in the Installation Manual.

This software controls the spectrophotometer from a personal computer (PC), collects and processes
data, then makes a pass or fail decision based on predefined acceptance criteria. 2
This software only checks the performance of a spectrophotometer. It will not operate correctly if optional
accessories are attached to the spectrophotometer.

2.1 Intended Instrument Types
This software can control and test the following instrument types. 2
• UV-1600 series (UV-1601/UV-1601PC/UV-1650PC)
• UV-2400PC series (UV-2401PC/UV-2450) 2
• UV-2500PC series (UV-2501PC/UV-2550)
• UV-2600 series 2
• UV-2700 series

Note that items that can be tested and measuring parameters that can be configured vary depending on

the instrument type. Therefore, this software can only control (test) a single instrument type after
installation. 2
Be sure to only use this software for a single instrument under a single (PC) environment.
The type and serial number of the instrument to be tested must be specified when the software is
being installed.

2 Software Overview

• The instrument type cannot be changed after installation. If you want to change the instrument type,
reinstall the software. Note that if the instrument type is changed, the test condition file currently used
can no longer be used.
• For the UV-1600/2400PC/2500PC series, the serial number or the instrument type cannot be changed
after installation. A different instrument of the same instrument type can be connected, but the serial
number specified during installation will be recorded in the test result file. As a result, the instrument
information contained in the result file will be different from that of the actual instrument. Therefore, do
not use this software for a different instrument.
• For the UV-2600/2700 series, the instrument serial number is stored in the internal memory. When the
instrument is connected, this software loads the stored information, and thus a different instrument can
be connected and controlled (tested) as long as it is the same instrument type as the one initially
configured. As the instrument name is set as part of the instrument information in the software
( "6.4 [Instrument] Menu"), the name must be changed each time you change instruments. If you
forget to change the setting, obtained test results may become invalid. Therefore, it is recommended that
you only use this software for a single instrument under a single environment (PC).

2 Software Overview

2.2.1 File Property Pane


Enter information about the test results file.
Before starting a test, you must specify the folder to save the file to, the result file name, and the
analyst name.

See "4.3.2 Entering Information About the Test Results File".

The settings for the destination folder and the result file name are saved in the test condition file.

You may create a new folder in the dialog box, but is may take a few seconds to
create the new folder after you click the "new folder" button.

If a file with the same name exists in the destination folder, the date and time is
added to the end of the file name (as shown in the following example) indicating

Example: If the test started on September 9, 2011 at 10:30 AM and 56:21
seconds,

Enter the name of operator responsible for the test.

highlighted in different colors to indicate the state of progress or results.

5 Batch of Test(s) Row colors
Orange: Indicates that the item has been selected to check the test result. The
result of the selected test item is displayed in the Result pane.
Green: Indicates that the item is currently being tested.
Aqua: Indicates that the item has been tested, aborted, or skipped.
Character colors
Red: Indicates that the test failed.
Displays the overall pass/fail decision of the result for each test item.
Passed: All tests for the selected test item had satisfactory results.
Failed: A test for one or more selected test items had unsatisfactory
results or the test was aborted (or untested).
6 Result Example for "WL Accuracy (Deuterium Discharge Lamp)": If the
result of wavelength 486.0 nm is determined as "Passed", while
testing for wavelength 656.1 nm was aborted and determined as
"Failed", then the overall result becomes "Failed".
Not excuted: The item was not tested.
Displays the name of the condition file currently loaded.
7 Condition file name If a file was created and has not yet been saved, a file name is not displayed in
this field.

For information about the result file, see "2.5 File Types".

• The "total test time" or "remaining test time" does not include standby time (i.e., the wait time required
for the instrument to stabilize) during a test.
• The "total test time" includes the time required for testing all items with checked boxes in the Batch of
Test pane.
• Both the "total test time" and "remaining test time" are only intended for use as a rough indicator. For
some items, the discrepancy may be a few minutes.

When a test has started, the result of the current test is displayed in real time in a window (called
View Test window). You can also display the result of a test item already completed (called View
Result window).
The windows can be switched using [View Test] or [Results] on the tool button bar or the [View]
menu. ( "6.2 [View] Menu")

The View Result window appears automatically when the test has been completed. To start another test,
configure test conditions or load the existing test conditions.

For the procedure to switch the windows, see "4.4 Checking Test Results in Process".

2.5 File Types

2.5 File Types


This software can create two types of files.

„ Test results file (*.pvr)


When the test processes are completed, a result file is created automatically and saved in the specified
folder.
Included in the result file are measurement and decision results, waveform data (spectrum and time
course), measuring parameters, and an event log, which can be checked or printed after being loaded

using this software.

For the method used to check test results, see "4.4 Checking Test Results in Process".

„ Parameter file (*.pvm)


A test condition file includes the following information:
• Items to be tested (test items) and their order
• Detailed conditions for respective test items (wavelength, acceptance criteria, etc.)
• Information related to test tools (such as verified value, expiration date, control number of
reference filter)
• Names of the destination folder and file where results are saved*
* The names of the destination folder and file are those entered in the file property pane in the Main window
( "2.2.1 File Property Pane") when you save the condition file.

For more details about configuring, saving, or loading a condition file, see "4.2 Test Conditions".

At the time of installation, the "UVPerfval-Data" folder is created in the install drive as the initial
destination folder for the result file and the condition file. This folder will not be removed even when the
software is uninstalled.

graph". The supported paper size and orientation is A4 portrait. If you print data in letter size, you

This software has three accounts, and four functions to which access can be restricted for each
account. Access to those functions by logged in accounts can be restricted by assigning the functions

„ Registering frequently used test conditions


This software can register up to three frequently used test condition files.
For example, test conditions for periodic inspection (such as test items and the acceptance criteria)
can be registered as a "frequently used condition" when the conditions are saved as a file. The
registered condition can be easily selected and loaded using a tool button in the Main window.

„ Listing test conditions


The current test conditions can be confirmed and printed in a list.
Use this function to make sure that the loaded test condition file has no errors. This function can be
used even if you are not authorized to edit the conditions or view the Edit window.
From the [Tests] menu, click [View Conditions] to open the Preview window and view the test
conditions.

„ Selecting "Tested" or "Not Tested" (Selecting Batch of Test)


You can select items to be tested without changing test conditions.
For example, if the test fails for only one item as a result of testing multiple items, you can load the

The "Tested" or "Not Tested" setting is not saved in the test condition file.

2.7.3 Automatic Standby for Test


It is recommended that some items be tested in stable temperature and lamp conditions of the
instrument.

See "3.3 Automatic Test Items".


If the recommended amount of time for instrument stabilization* has not passed before a new test
starts, the new test is put on standby until the instrument stabilization time has elapsed, after which
the test starts automatically.
It is also possible to forcibly restart the test during the instrument stabilization time.

3.2 Semi-Automatic Test Items


These test items require a reference filter or other test tools for testing.
If two or more test tools are used for testing, an operator will be required to insert or remove the test
tools according to messages displayed during the test.
Use an accessory such as the 6-Cell Multi Cell Sample Compartment or CPS-240A to change the test
tools automatically during the test.

Table 3-1

Test Item Description


Measures the absorbance or transmittance of a transmittance calibration filter*, and
determines, based on the criterion, whether or not to accept the error between the
measured value and the calibration reference value (verified value) attached to the
filter.
You can register several filters with different transmittance to test with all or any of
the filters. For more accurate testing, it is recommended that you set an approximate
(about ± 3 °C) sample (filter) temperature specified in the calibration certificate of
Photometric Accuracy the calibration filter to be used.
The 6-Cell Electronic Temperature Control Cell Holder (CPS-240A) can be used to
control the filter temperature during the test.

To set acceptance criteria values, add accuracy of the calibration filter specified in
the certificate.
Measures the photometric value (absorbance or transmittance) using a transmittance
calibration filter* at a wavelength set arbitrarily. Repeats the measurement three

3.2 Semi-Automatic Test Items

Measures the relative stray light. The value is obtained by: {(Sum of light intensity
at wavelengths other than the set wavelength)/(Light intensity output from the
spectrometer at the set wavelength)} × 100.
The stray light value at the following wavelengths can be determined:
• 198 nm - When potassium chloride solution (12 g/l) is used (Only for UV-2600/
2700)
• 220 nm - When sodium iodide solution (10 g/l) is used
• 340 nm - When sodium nitrite solution (50 g/l) is used

Absorbance error caused by stray light is described in "8.3.1 What is Stray Light?".

Measures the absorbance spectrum of 0.02 %V/V toluene solution in hexane
("toluene/hexane solution") to obtain the absorbance ratio of the maximum (peak)
Resolution value in the vicinity of 269 nm and the minimum (valley) value in the vicinity of
(Absorbance Ratio) 266 nm, and then determines whether or not to accept the value based on the
criterion.
This test uses the method specified in European Pharmacopoeia to check resolution.
Measures a wavelength calibration filter* to determine, based on the criterion,
whether or not to accept the error between the measured value and the reference

wavelength attached to the filter. To set the acceptance criterion value, add accuracy
(uncertainty) of the calibration filter specified in the certificate.
Measures an arbitrarily selected absorption peak three times using a wavelength

These items can be tested automatically from measurement to pass/fail decision once the test starts.

3.4.1 Decision Digit Count
A measurement result is displayed using the same number of digits (after the decimal point) as
defined for acceptance criteria.
If a criterion has three digits after the decimal point, then the fourth digit of the measured value is
rounded (a value of 5 or higher is rounded up while a value of 4 or lower is rounded down). The
rounded (displayed) value is used to determine whether it passes or fails the criterion.

For some test items, the digits can be changed. 3

Before testing for semi-automatic test items, add test tools to be used, and enter the relevant information
(control number, expiration data, wavelength, etc.).

4.3 Performing a Test


This section explains the operations before starting a test.

4.3.1 Checking Test Tools


Some test items require apparatuses such as reference filters and solutions as well as reagents (test
tools) for testing.
Add test tools to be used or edit reference values in the Detailed Condition Setting window for each
test item.

See "4.2.1 Configuring Test Conditions". For test tools required for each test item, see "3.2 Semi-
Automatic Test Items" and "3.3 Automatic Test Items".

Before starting a test, make sure that all the required test tools are ready and usable (check the
expiration date of each test tool).

If the expiration date of a tool has passed, the test cannot be started.

5.2 Test Operations


Because automatic testing does not use test tools, all processes, from the start to end of each test and
the transition to the next test, are carried out automatically.

• This software automatically turns the lamps on and off before and after a test. Be sure to turn on the
power switch of the "Mercury Lamp Unit" before starting a test. For the position of the power switch,
refer to the Instruction Manual of the "Mercury Lamp Unit".
• An optional "Mercury Lamp Unit" is required for "WL Accuracy (Mercury Lamp)" and "WL
Repeatability (Mercury Lamp)". Before starting the test, install the lamp unit onto the spectrophotometer
according to the instructions in the included Instruction Manual.

5 Test Items

„ Checking stray light using a mesh filter


UV-2700 series is a low stray light instrument as it has a specification of maximum 0.00005 %T at
220 nm.
To check the performance at the recommended 0.001 %T or higher level, the cell holder on the
reference side must be left blank for testing. However, to check the stay light value at lower than
0.001 %T within the instrument specification range (by setting the number of decision digits to 4 to 5
after the decimal point), the light on the reference side must be dimmed and the measurement range
must be enlarged.
In that case, insert a test tool into the cell holder on the sample side and a mesh filter
( "3.2 Semi-Automatic Test Items") into the cell holder on the reference side at "Operation
message 1" and start the test.

• For UV-1600/2400PC/2600 series, no mesh filters are required as the value level is out of the instrument
specification range.
• UV-2500PC series is also an instrument with low stray light, but this software cannot test a stray light
value lower than 0.001 %T.

To check a stray light value lower than 0.001 %T within the instrument specification range, see
"8.2.2 Testing Stray Light Using Mesh Filter" to use "UVProbe Software" for testing.

8.2 Test Method Using UVProbe


This section describes the method used to measure and test special items this software cannot test by
using the UVProbe Software, which can control spectrophotometers and process their data.

8.2.1 Testing Resolution Using Benzene Vapor

WARNING
When handling benzene, take appropriate safety measures by wearing adequate working
clothes, protective goggles, masks, and gloves in accordance with the Material Safety Data
Sheet (MSDS) guidelines created by the relevant reagent manufacturers.
Failure to observe this may result in an accident.
If an accident involving chemicals occurs, take relevant first-aid and emergency measures according
to the instructions provided by the relevant organizations.

1 Encapsulate approximately 2 μl (a half drop) of benzene in the quartz cell with an


airtight stopper (optical path length: 10 mm).

2 Set the measurement mode of the spectrophotometer to absorbance, the scanning


speed to low, and the slit width to 0.1 nm.

3 Set the wavelength scanning range to between 252 nm and 248 nm, and after
baseline correction, set the above quartz cell in the sample compartment and measure 8
spectrum.

4 Adjust concentration of the benzene vapor so that the absorption peak in the range
251 nm to 249 nm (encircled in the following figure) will be 0.5 Abs ±0.2 Abs.

To attenuate the concentration, open the cell plug and leave it for a while. To increase the
concentration, warm up the cell with body heat, or add approximately 1 μl of benzene.

8.2.2 Testing Stray Light Using Mesh Filter


The following describes the procedure for testing the stray lights (of 220 nm and 340 nm) using mesh
filter P/N 206-82299-91 for UV-2500PC series.

„ Obtaining a stray light value at 220 nm

1 Start the UVProbe Software and enter the spectrum measurement mode (spectrum
module).

2 Set measuring parameters as follows.

3 Make sure that nothing is inserted into cell holders and start baseline correction.

4 After baseline correction is completed, set [Go To WL] to 220 nm, insert a mesh filter
into the cell holder on the sample side, and close the sample compartment cover.

5 Write down the transmittance displayed in the lower right of the window.
Let this value be "transmittance A".
In sample compartment > Sample (S) side: Mesh filter, Reference (R) side: Air

6 Move the mesh filter from the sample side cell holder to the reference side, and in its
place, insert a shutter block into the sample side cell holder.
In sample compartment > S side: Shutter block, R side: Mesh filter

7 Press [Start] to start measurement, and after the measurement is completed, click
[Point Pick] from the [Operations] menu, and write down the transmittance at 220 nm.
Let this value be "transmittance B".
* Click [Settings] from the [View] menu to set the number of digits to 3 after the decimal point to be
displayed for Y-axis value (photometric value).

8 Remove the shutter block from the sample side cell holder, and in its place, insert a
quartz cell containing 10 g/l of sodium iodide (NaI) solution into the cell holder.
In sample compartment > S side: Cell with NaI solution, R side: Mesh filter

9 Press [Start] to start measurement, and after the measurement is completed, click
[Point Pick] from the [Operations] menu, and wite down the transmittance at 220 nm.
Let this value be "transmittance C".

As the stray light value at 340 nm must be measured while the WI lamp is used, set the light source switch
wavelength to 330 nm before starting measurement.

8.3 Reference Data


8.3.1 What is Stray Light?
Stray light is the total of the light of the wavelength deviated from a certain spectrum width that has
the set wavelength of the spectrometer placed in the center and the light emitted from spectrometer
that passes the side of the sample instead of transmitting it.
For example, if 0.1 % of stray light is included in the monochromatic light when a sample with the
wavelength of λo and absorbance of 2 (transmittance 1 %) is measured, the transmitted light will
have 1.1 % of transmittance (absorbance 1.959), causing 2 % error. It is because 0.1 % stray light is
added in addition to 1 % of the light with the wavelength of λo.
As known from this, the higher the sample absorbance is, the larger the error due to stray light
becomes.
To reduce stray light, it is generally effective to form a double-monochromator by connecting two
monochromators. Although it is expensive due to the complex mechanism, the amount of stray light
is reduced from one-fiftieth (1/50) to one-one thousandth (1/1000) of that of normal single
monochromater.

8.3.2 Influence by Hydrogen Emission Line


A micro amount of hydrogen atoms are in the deuterium lamp used for the resolution and other tests.
This emission line spectrum of hydrogen atoms occurs at 656.28 nm, which is in proximity to
656.10 nm of the emission line of the deuterium lamp. This may affect test results of resolution
obtained using UV-2400/2500 series when testing resolution (spectral band width) with the 0.2 nm
slit width.
Figure A below shows the emission line spectra at 656.1 nm, measured by setting the slit width to
0.1 nm and data interval to 0.01 nm using Shimadzu spectrophotometer UV-3600. "H" is a spectrum
generated by hydrogen, and "D" by deuterium.
Figure B shows the emission line spectra when this is measured by setting the slit width to 0.2 nm. As
the hydrogen's spectrum overlaps, two spectra appear to be a single emission line spectrum. The
resolution (half width) at this time is 0.24 nm.
The default (recommended) value as an acceptance criterion is less than 0.21 nm for UV-2400/2500
series. As hydrogen atoms have a significant affect in this example, the result fails the criteria.
However, this is not because the spectrophotometer has a resolution-related failure.

When the spectrum is measured using UV-2400PC/2500PC series, the applicable minimum sampling
interval is 0.05 nm, and thereby a 0.1 nm resolution cannot be checked with the spectral band width of the
emission line.
To check a 0.1 nm resolution using UV-2400PC/2500PC series, you can only measure benzene vapor and
obtain its shape to determine the resolution.
