Water determination with Karl Fischer titration
Principle:
Karl Fischer titration is used for many substances as a reference
method, and is a chemical analysis based on the oxidation of sulfur
dioxide by iodine in a methanolic hydroxide solution. In principle, the
following chemical reaction takes place:
H2O + I2 + SO2 + CH3OH + 3RN -> [RNH]SO4CH3 + 2[RNH]I
The titration can be performed volumetrically or coulometrically.
In the volumetric method a Karl Fischer solution containing iodine is
added until the first trace of excess iodine is present. The amount of
iodine converted is determined from the burette volume of the iodine-
containing Karl Fischer solution.
In the coulometric procedure, the iodine participating in the reaction is
generated directly in the titration cell by electrochemical oxidation of
iodide until again a trace of unreacted iodine is detected. Faraday's law
can be used to calculate the amount of iodine generated from the
quantity of electricity required.
Application:
The Karl Fischer titration is a moisture determination method specific for
water and is suitable for samples with a high water content (titrimetry)
and also for those with water content in the ppm range (coulometry). It
was originally developed for nonaqueous liquids, but is also suitable for
solids if these are soluble or if the water they contain can be removed by
heating in a stream of gas or by extraction.
Advantages:
Accurate reference method, coulometry also suitable for trace analysis
and water detection.
Restriction:
The working method must be adapted to the specific sample.

A titration is a technique where a solution of known concentration is used to
determine the concentration of an unknown solution. Typically, the titrant (the know
solution) is added from a buret to a known quantity of the analyte (the unknown
solution) until the reaction is complete. Knowing the volume of titrant added allows
the determination of the concentration of the unknown. Often, an indicator is used
to usually signal the end of the reaction, the endpoint.
Chlorophyll in Water
Chlorophyll in various forms is bound within living cells of photosynthetic organisms, such
as phytoplankton and cyanobacteria (blue-green algae). The amount of chlorophyll found in
a water sample is used as a measure of the concentration of phytoplankton. These
measurements contribute to the understanding of the general biological “health” of
the system, such as its trophic status or primary production. Chlorophyll measurements can
also identify algal bloom events and their effects on water quality and anticipate toxic algal
blooms.
Chlorophyll fluoresces when irradiated with light of a particular wavelength (435-470 nm).
For field measurements, in situ fluorometers induce chlorophyll to fluoresce by shining a
beam of light of the proper wavelength into the water and then measuring the higher
wavelength light which is emitted. These real-time chlorophyll measurements complement
extractive lab analysis.
Blue-Green Algae
Blue-green algae (BGA), also known as cyanobacteria, can range in colors from blues,
greens, reds, and black. BGA can reduce nitrogen and carbon in water, but can also
deplete dissolved oxygen when overabundant. Monitoring BGA is important because they
pose a serious threat to water quality, ecosystem stability, surface drinking water supplies,
and public health through toxin production and the large biomass produced in algal blooms.
YSI measures blue-green algae in real-time through the in vivo fluorometry technique. This
method directly detects the fluorescence of a specific pigment in living algal cells and
determines relative algal biomass. The BGA sensor does not receive interference from
chlorophyll or turbidity.
