Phenol Red Fermentation Indole

1. 0.1 mL organism to each fermentation tube 1. 0.1 mL of organism in each tryptone broth tube
(Glucose, Sucrose, Lactose) 2. Shake gently. Incubate
2. Shake gently; Incubate. 3. + 1 mL ether/xylene; shake well
Methyl Red 4. Once ether rises to the surface, + 0.5 mL Erlich’s
1. 0.1 mL in buffered glucose medium reagent, tilt tube.
2. Incubate. Tripe Sugar Iron Agar
3. + 5 drps of methyl red indicator 1. streak then stab to 1/2 inch from the bottom of
Catalase TSI slant
1. pick a portion (w/ loop), place on slide 2. Incubate.
2. + a drop of 3% hydrogen peroxide. Nitrate Reduction
Starch Hydrolysis 1. 0.1 mL into each nitrate tube
1. 15-20 mL agar to 3 petri plates 2. Shake gently. Incubate
2. Inoculate with each of the cultures 3. + 2 drops of reagent A, shake. +2 drops reagent B,
3. Incubate (inverted). shake.
Gelatinase Urease activity
1. Inoculate gelatin tube by stabbing gelatin medium 1. 0.1 mL into each urea broth
down to the bottom 2. Shake gently. Incubate.
2. Incubate. Ice bath, 5 minutes Citrate
1. Streak into Simmon’s citrate slant.
2. Incubate.
