SAKAGUCHI TEST

OBJECTIVES:

To detect amino acid guanidinium group like arginine.

MATERIALS:

APPARATUSES TO MATERIALS TO SAMPLES TO BE TESTED:


BE USED: BE BROUGHT:

 3 Test tubes N/A  1 mL of 1% arginine + 2 drops of a-naphthol +


 1 Graduated 2 mL sodium hypochlorite + 1 mL of urea
Cylinder solution
 1 mL of 1% glycine + 2 drops of a-naphthol + 2
mL sodium hypochlorite + 1 mL of urea
solution
 1 mL of 5% egg white (albumin) + 2 drops of
a-naphthol + 2 mL sodium hypochlorite + 1 mL
of urea solution

PROCEDURE:

1. Put 1mL of 1% arginine, 1 mL of 1% glycine, 1 mL of 5% egg white (albumin) in to


different dry test tubes.
2. Add 2 drops of α-naphthol to the three samples and mix well.
3. Now add 2 mL sodium hypochlorite to all test tubes.
4. Immediately add 1mL of urea solution to establish the red complex formed.
