Read the Photosynthesis Lab Info posted on Schoology Complete the pre-lab information (typed) on a separate piece of paper Do the lab as instructed Complete data analysis and discussion: final lab report will be completed in class on Thursday/Friday 11/14/15.

Prelab Questions 1. We will measure the amount of DPIP remaining as an indicator of photosynthesis. Give a concise 4-5 sentence description of the events in photosynthesis that are occurring as DPIP is reduced to DPIPH2 and turns from blue to clear. 2. Describe how a spectrophotometer detects light. 3. What is the importance of each of the 5 cuvettes you will be using? Be sure to include which ones are controls and which are experimental. 4. Why must you always put the cuvette into the colorimeter lined up with the reference line? 5. State your hypothesis about the relative amount of light transmittance you expect to see in each experimental cuvette. 6. Read through the procedure and make a data table. Analysis and Discussion Keep in mind: You will be using class data. What is the purpose of taking class data? When explaining an experiment using class data, should you average it or talk about all the data individually? All work for calculations should be shown and/or explained in your report. 1. Make a graph of transmittance vs. time for your groups and classes experimental data (only one cuvette is experimental). Then make graph(s) that show class data for all controls. Label each graph appropriately. 2. Final experimental trial averages. Show your work! Place a label with the rate on the graph. 3. Describe the shape and end-point of the graphed class data curves. Explain why we get these results, in terms of the role of chloroplasts, light and DPIP. Make sure you cite specific data and evidence as part of your answers. 4. Explain why this procedure cant assess the activity of the dark reaction. Your answer should include explanation of how DPIP and NADP operate. 5. Briefly explain how you could use this experimental design to test the relative effectiveness of red, blue and green light on photosynthesis. Include the following: a. What technical problem will you encounter trying to test photosynthesis in blue light? b. What hypotheses would you make? Justify your hypothesis. c. What controls will you set up? Why? (Be specific)
