in central, western and southern Sudan. Hydatid cysts were present in 59% (466/779) of camels, 6% (299/4893) of cattle, 11% (1180/10,422) of sheep and 2% (106/5565) of goats, with little variation among different geographical areas. 532 of these cysts were examined by PCR and could be overwhelmingly (98.7%) allocated to Echinococcus canadensis G6/7 (all of 215 cysts from camels, 112 of 114 cysts from cattle, 134 of 138 cysts from sheep, and all of 65 cysts from goats); the genotype G6 was identified by sequencing 13 of these isolates. Only 2 cysts from cattle belonged to Echinococcus ortleppi. The mean number of cysts per infected animal was much higher in camels (5.1) than in the other species (1.0-1.3), and cyst fertility was higher in camels and cattle (74% and 77%) than in goats and sheep (31% and 19%). Fertile cysts from five human patients from hospitals in Khartoum and Juba belonged to E. canadensis (G6). This study confirms the predominance of the 'camel strain' in Sudan and the infectivity of this strain for humans. This is the first genetic characterization of human CE in Sudan.

New information was collected on cystic echinococcosis in livestock (camels, cattle and sheep) and humans in the central region of Sudan. The livestock data were collected in abattoir-based surveys in the towns of Omdurman, Tamboul and Wad Madani, between 1998 and 2001, and covered a total of 8205 animals. The highest prevalence of infection was found in the camels (44.6% of 242 infected), followed by the sheep (6.9% of 5595) and cattle (3.0% of 2368). Records were made of the sizes of the 1320 hydatid cysts detected in the livestock (907 in sheep, 71 in cattle, and 342 in camels), whether or not each cyst was fertile, and where it occurred in the body of the host. Cysts collected from cattle and camels where much more likely to be fertile (22% and 24%, respectively) than those from sheep (1%). Camels and cattle therefore appear to be the principal intermediate hosts for Echinococcus granulosus in central Sudan, whereas sheep apparently play a marginal role in transmission. In 2002, as a preliminary assessment of the public-health impact of the disease, 300 residents of a rural village 60 km west of Wad Madani were surveyed using a portable ultrasound scanner. Only one (0.33%) of the villagers investigated was found infected. The implications of these finding are discussed in terms of the various strains of E. granulosus and the role of each in human disease.
The objectives of the present study were to investigate strain identification of Echinococcus granulosus infecting camel and human in Qalyubia, Egypt. Therefore partial sequences were generated after gel purification of nested PCR amplified products of mitochondrial NADH 1gene of Echinococcus granulosus complex. Sequences were further examined by sequence analysis and subsequent phylogeny to compare these sequences to those from known strains of E.granulosus circulating globally and retrieved from GenBank. All isolates are homologous to the camel strain, E. canadensis (G6) genotype. Nucleotide mutations generate polymorphism at position of 275 nucleotide, where a thymine replaced a cytosine and at the levels of 385 and 386 nucleotides, where two cytosine substituted a guanine and a thymine respectively. KF815488 Egypt showed typical identity (99.5%) with JN637176 Sudan, HM853659 Iran, AF386533 France and AJ237637 Poland with 0.5% diversion.. Phylogenetic analysis showed a robust tree clustering all isolates with sequences belonging to the camel genotype (G6) variant with strong bootstrap values at relevant nodes and the evolutionary distance between groups is very short. There are two mutations in the sequences of amino acids at the position of 92, where an Alanine is changed to a Valine and at the position of 129, where a Valine is transformed to a Proline. Our record of a single genotype determined a strain which could be incriminated for camel and human infectivity and responsible for its persistence in the endemic areas. Cystic echinococcosis (CE) is a zoonosis caused by the cestodes of the Echinococcus species. Its life cycle involves dogs and other canids as definitive hosts for the intestinal tapeworm, as well as domestic and wild ungulates as intermediate hosts for the tissue-invading metacestode (larval) stage. Cystic echinococcosis has been considered one of the major parasitic zoonoses which is associated with severe economic losses. The present study was undertaken to investigate the occurrence, organ distribution, cyst fertility, and viability of cystic echinococcosis in slaughtered camels and cattle from various abattoirs in Assiut Governorate, Egypt. The work also involved morphological, morphometric, and molecular identification of the parasite. In Algeria, cystic echinococcosis (CE) is a serious economic and public health problem. The common sheep/dog cycle is usually considered as the major source of human contamination. But to date the main strain of Echinococcus granulosus involved in the human contamination and the role of other hosts are still unknown. This paper reports an original work performed in northern Algeria combining field observations and molecular analysis. In a first step, examination of 6237 carcasses in slaughterhouses showed high infection and fertility rates in cattle and dromedaries. Then, in a second step, we used a molecular biology approach to identify the E. granulosus strain(s) involved. Forty-six samples from various origins were collected. They were analysed using comparison of PCR-amplified DNA sequences with one genomic (BG 1/3) and two mitochondrial (COI and NDI) targets. Results show the presence of a "sheep" strain of E. granulosus in North Algeria circulating between cattle and ovines and infectious to humans, whereas in South Algeria, a "camel" strain and a "sheep" strain were found to circulate in camels and in sheep, respectively. This study also reports an ambiguous genotype which resembled the "sheep" strain genotype (Gl) on the basis of the partial COI gene sequence, whereas on the basis of the partial NDI gene sequence, it was similar either to the "sheep" strain (Gl) or to the "camel" strain (G6). The aim of the present study was to determine the molecular characteristics of Echinococcus granulosus sensu lato from faecal samples of dogs and the hydatid cyst from liver of bovine, camel, cattle, and sheep and the immunodiagnosis of Hydatid cyst in human sera samples from the Gaza Strip, in Palestine. A total of 38 faecal samples were collected near farms and local markets where stray dogs were grouping there. Post-slaughtering, the livers of the bovine, camel, cattle, and sheep were examined for cysts. Where 30 suspected hydatid cysts were collected, the number and size of cysts in each liver were recorded. For serology, 23 sera specimens were collected from farmers within the Gaza Strip. According to hospital records in Gaza, 12 patients had attended three different hospitals and had tested positive for hydatid cyst/cystic echinococcosis by CT scans and histopathology. ELISA results from the 23 human patients showed that 3/23 (13%) tested positive for hydatid disease. Of the 30 bovine and cattle harboring cysts, 14/30 (46.6%) were positive for hydatid cyst/cystic echinococcosis of which 23 (76.7%) were males and 7 (23.3%) were females. The present study shows that 6/38 (15.7%) were positive for E. granulosus sensu lato from faecal analysis in dogs. Subsequent sequencing of both dogs and cattle confirmed infection by the G1 strain. The cattle/dog strain (G1) of E.granulosus sensu lato suggests that this infection is cosmopolitan in its distribution. It is concluded that the detection of hydatid cyst/ cystic echinococcosis in the examined hosts either human or animals should be considered among physicians and a large sample size is recommended in future research.

The objectives of this epidemiological study on cystic echinococcosis (CE) in Morocco (2001-2004) were to update the prevalence of CE in different animal species living in the most important areas of the country and to collect protoscoleces and germinal layers for genetic research purposes. The post mortem inspection concerned 2948 sheep, 2337 goats, 618 cattle, 482 camels and 455 equines (325 horses, 60 mules and 70 donkeys) in five different regions: the Rif (Mediterranean coast and high mountains of the Rif), the Loukkos (Atlantic northwest plain), the center (Rabat and Casablanca regions), the Middle Atlas mountains and the south (arid and semi desert areas). The global CE infection prevalence rates obtained were 22.98% in cattle, 10.58% in sheep, 12.03% in camels, 17.80% in equines and 1.88% in goats. Iran is an important endemic focus area of cystic hydatid disease where several intermediate host species are commonly infected with Echinococcus granulosus . Annually, most livers are shipped out, stored away, or destroyed due to contamination among livestock. For instance, in Aleshtar (Lorestan province), liver and lung losses due to hydatid cysts during 2002-2003 were approximately 864,360,000 Rials and 35,530,000 Rials for sheep and cow, respectively. In other words, given the risk of human contamination with this parasite, the costs associated with diagnosis, treatment, and surgery are important. Epidemiology studies have been performed in different part of Iran, but there is no international estimate of infection in livestock. Molecular studies have indicated the presence of G1 and G6 genotypes in different parts of Iran. We discuss these molecular studies in this article.

Background: Echinococcus granulosus (EG) complex, the cause of cystic echinococcosis (CE), infects humans and several other animal species worldwide and hence the disease is of public health importance. Ten genetic variants, or genotypes designated as (G1-G10), are distributed worldwide based on genetic diversity. The objective of this study was to provide some sequence data and phylogeny of EG isolates recovered from the Sudanese one-humped camel (Camelus dromedaries). Fifty samples of hydatid cysts were collected from the one-humped camels (Camelus dromedaries) at Taboul slaughter house, central Sudan. DNAs were extracted from protoscolices and/or associated germinal layers of hydatid cysts using a commercial kit. The mitochondrial NADH dehydrogenase subunit 1 (NADH1) gene and the cytochrome C oxidase subunit 1 (cox1) gene were used as targets for polymerase chain reaction (PCR) amplification. The PCR products were purified and partial sequences were generated. Sequences were further examined by sequence analysis and subsequent phylogeny to compare these sequences to those from known strains of EG circulating globally. Results: The identity of the PCR products were confirmed as NADH1 and cox1 nucleotide sequences using the Basic Local Alignment Search Tool (BLAST) of NCBI (National Center for Biotechnology Information, Bethesda, MD). The phylogenetic analysis showed that 98% (n = 49) of the isolates clustered with Echinococcus canadensis genotype 6 (G6), whereas only one isolate (2%) clustered with Echinococcus ortleppi (G5). Conclusions: This investigation expands on the existing sequence data generated from EG isolates recovered from camel in the Sudan. The circulation of the cattle genotype (G5) in the one-humped camel is reported here for the first time.

Little is known about the genotypes of Echinococcus spp. and their life cycles in eastern Iran. We analysed the partial sequences of the nad1 and cox1 genes from 17 isolates from hydatid cyst-infected patients (n=9), camels (n=5) and sheep (n=3) in Birjand, eastern Iran. A new primer pair was also used to amplify the long fragment (1180 bp) of the cox1 gene. All camel and eight human isolates were G6 strains of Echinococcus canadensis while one human isolate and the three sheep isolates were G1 genotypes (sheep strain) of E. granulosus sensu stricto (s.s.). Nad1 and cox1 sequence analyses showed high G6 genetic homogeneity, similar to previously reported G6 strains from southeast and central Iran, Sudan and Mauritania. Low nucleotide and haplotype diversity similar to G6 strains from Russia (Altai republic) and Kazakhstan was also found, consistent with a bottleneck effect. In this study, G6 was the most common Echinococcus genotype. Genetic homogeneity of east, southeast and central Iranian G6 and its low genetic diversity may be due limited mobility and contact between humans and camels from other regions because of large, inhospitable deserts.

This study included nine samples collected from the infected (liver and lung) divided into three equal samples (sheep, cattle and humans) collected from some Central Euphrates Provinces for the period from the beginning August 2019 till February 2020. PCR technique was performed for in detection and genotyping of Echiococcus granulosus hydatid cyst based on mitochondrial Cox1 gene and Nad1 genes in isolates. This technique was carried out according to method described by Nikmanesh et al. (2014). The present study aimed to showed haplotype genetic and the frequency percentage of it in the Central Euphrates region. the result showed the H15 haplotype was a higher frequency followed it (H10, H11) and then (H5). The frequency percentage of the haplotype genetic in the current study of the taken samples (8) that the prevalence and frequency of haplotype genetic H15 was 37.5%, followed by the genotype (H10,H11) at 25% for both them , and the lowest was the genotype (H5) at 12.5% The human samples registered 100% for (H15) , the sheep registered 50 % for H5, H10.While cattle samples registered 33.3 % for H10 and 66.6 for H11. Isolation H15 is the essential in the infection to humans as a result of dogs were fed hydatid cysts of cattle origin, and there is no relationship between sheep and humans, while there is a relationship between sheep and cattle in the Haplotype (H10).

Cystic Echinococcosis (CE) caused by Echinococcus granulosus sensu lato (s.l.) is a neglected helminth zoonosis affecting humans and various animal species. Human CE has been reported in almost all countries of sub-Saharan Africa but its prevalence and public health impact are subject to large geographical variations. The reasons for these differences are not well understood; among other factors, occurrence of different species/genotypes of E. granulosus s.l. has been suggested. CE is very common in all livestock species in Ethiopia; human CE is poorly documented in the country. The aim of this study was to assess the fertility and molecularly characterize hydatid cysts collected from cattle, camels, goats and pigs from different parts of the country. From the 137 samples characterized by PCR-RFLP and sequencing, 115 (83.9%) were identified as E. granulosus s.s. (G1, common sheep strain), 6 (4.4%) as Echinococcus ortleppi (G5, cattle strain) and 16 (11.7%) as Echinococcus intermedius (G6/7, camel strain). In cattle, E. granulosus s.s. and E. ortleppi were found; in camels and goats, E. granulosus s.s. and E. intermedius; two cysts found in pigs were identified as E. granulosus s.s. and E. ortleppi, respectively. All cysts recovered from goats and pigs were sterile, while fertility was 34% and 50% in cysts from cattle and camels, respectively. In cattle, 31% of E. granulosus s.s. cysts were fertile, showing the importance of cattle in the transmission of the "sheep strain". Next to E. granulosus s.s., E. intermedius (camel strain) was the predominant species: 34.4% of the cysts collected from camels and 62.5% from goats were identified as E. intermedius. These animals originated from the drier Central, Eastern and Southern parts of the country. For the first time, we showed the presence of CE in pigs in Ethiopia. The presence of these strains and especially the fact that the zoonotic E. granulosus s.s. and E. intermedius are dominant, make CE an important public health concern in Ethiopia.

Cystic echinococcosis (CE) or hydatidosis is a widespread zoonose in the world. In Tunisia, despite the deployed prevention program, CE remains a serious public health problem. With a human surgical annual incidence averaging 12.6/100000 inhabitants, Tunisia is one of the most endemic areas amongst the Mediterranean countries. Three Echinococcus species have been described: E. granulosus sensu stricto (G1 and G3 genotypes), E. canadensis, and E. equinus. CE, which commonly starts during childhood and described as a young adult disease, may be observed at any age. The liver and the lungs are the most commonly involved organs but the cyst can occur almost anywhere in the body. In production animals, the prevalence of CE is ranged from 16.42% to 40.42% in sheep, 8.56% in cattle, 6% in dromedaries, 2.9% in goats, and 8.48% in donkeys. The elevated number of stray and semi-stray dogs and their frequent contamination by E. granulosus infected viscera is the major cause of the CE spread. A high prevalence of E. granulosus infection has been reported in Tunisian dogs ranging from 3.75% to 27.1%, depending on the regions. Thus, the sanitary education concerning hydatidosis should be reinforced and efforts should be made to implement a targeted educational program.

Cystic echinococcosis (CE) is a neglected zoonotic disease with considerable economic and public health burden worldwide, particularly affecting developing countries like Ethiopia. To initiate effective prevention and control of CE, comprehensive data on the local lifecycles of the various species/genotypes of Echinococcus are needed. In the present study, conducted in eastern Ethiopia, a total of 1106 livestock animals were examined at three slaughterhouses, which resulted in combined prevalence of morphologically and molecularly confirmed CE of 8.4% (75/891) in cattle, 1.1% (1/95) in sheep, 0.0% (0/95) in goats and 12.0% (3/25) in camels. All cystic lesions recovered during post mortem examination were assessed for cyst condition and underwent molecular characterization by PCR and sequencing of a 1081 bp fragment of the mitochondrial cox1 gene. A total of 175 cysts belonged to E. granulosus s.s. (n = 165), E. ortleppi (n = 6) and E. canadensis G6/7 (n = 4). Of all examined cysts, only 14 were fertile and contained protoscoleces, all from the lungs of cattle: 5 were E. granulosus s.s., 6 E. ortleppi and 3 E. canadensis G6/7. In sheep, only one sterile liver cyst of E. granulosus s.s. was found, while in camels seven sterile or caseated/calcified cysts of E. granulosus s.s. and E. canadensis G6/7 were found in liver and lungs. In conclusion, the prevalence of CE was rather low compared to other regions of Ethiopia, and, based on the number of fertile cysts, three Echinococcus spp. contributed almost equally to transmission. Cattle seem to be, epidemiologically, the most important livestock species. Our data provide a substantial basis for more detailed investigations of the transmission dynamics of CE in the study area.

Cystic echinococcosis (CE), caused by hydatid cysts, is a widespread and hazardous disease in humans and animals worldwide. The aim of the current study was to investigate the genetic variations among Echinococcus granulosus cyst strains isolated from sheep, camel, pig, and donkey using RAPD-PCR analysis. Seven primers of arbitrary sequences were used in the PCR reactions. The screened primers gave total patterns ranging from 27 to 39 reproducible bands for each isolate. Each population isolate gave its specific pattern. Although distinct polymorphic patterns were obtained among the four isolates, there were several shared bands among them in each primer used. A comparison of the different RAPD-PCR patterns showed that primers P1, P3, and OPH 04 yielded band patterns that revealed a high degree of divergence among the four isolates of E. granulosus that allowed easy distinction between them. The remaining primers (P2, P4, P5, and OPH14) amplified DNA fragments that were common to two or more isolates but diversified in the other two or three isolates. The study revealed that the most closely related isolates were of donkey and camel where the similarity coefficent between them ranging from 53 % to 78 %, followed by isolates of pig and sheep (sc040 % to 68 %), while the similarity coefficent between isolates of camel and sheep was 33-45 %, between camel and pig was 36 to 57 %, between donkey and pig was 37 to 52 %, and between donkey and sheep was 35 to 54 % which means that they more or distant from each other. In conclusion, hydatid cysts isolated from camel may have the genotypic characters of donkey strain.

Cystic echinococcosis is a neglected, zoonotic disease in Turkey. The disease is commonly seen in rural areas where the local population is in close contact with livestock and dogs. This research aimed to molecularly identify of hydatid cysts in cattle and human isolates from Konya, Turkey. Following sample collection, direct microscopy was performed. After direct examination, total DNA was extracted, and positive PCR products of cox 1 mitochondrial gene (~875 bp) were sequenced. A total of 83 hydatid cysts (cattle n = 57 and human n = 26), 82 were identified as Echinococcus granulosus sensu stricto (G1-G3 genotypes), and one human isolate was characterized as Echinococcus equinus (G4 genotype). Fertility rates of cysts belonging to cattle for liver and lung cysts were 93.3% and 80%, respectively. Out of 26 human originated isolates, 18 (69.2%) of cysts were found to be fertile. To the best of our knowledge, this is the first report of E. equinus from human host in Turkey.
