and NF-H. All three subunits contain a well conserved glutamate (E)-rich region called "E-segment" in the N terminus of the tail region. Although the E-segments of NF-L and NF-M are phosphorylated by casein kinases, it has not been observed in NF-H. Using mass spectrometric analysis, we identified phosphorylation of the E-segment of NF-H, prepared from rat spinal cords, at Ser-493 and Ser-501 in the Ser-Pro sequences. The E-segment kinase was isolated from rat brain extract using column chromatography and identified as glycogen synthase kinase (GSK) 3. GSK3 was shown to phosphorylate at Ser-493 in vitro by phosphopeptide mapping and sitedirected mutagenesis, and in vivo in HEK293 cells using the phospho-Ser-493 antibody, but did not phosphorylate Ser-501. GSK3 preferred Ser-493 to the KSP-repeated sequences for phosphorylation sites in the NF-H tail domain. Moreover, Ser-493 was a better phosphorylation site for GSK3 than other proline-directed protein kinases, Cdk5/p35 and ERK. GSK3 in the spinal cord extract was associated with NF cytoskeletons. Taken together, we concluded that Ser-493 in the E-segment of NF-H is phosphorylated by GSK3 in rat spinal cords. molecular weight subunit of neurofilament; NF-M, middle molecular weight subunit of neurofilament; NF-H, high molecular weight subunit of neurofilament; DTT, dithiothreitol; Pipes, 1,4-piperazinediethanesulfonic acid; MOPS, 4-morpholinepropanesulfonic acid; CA-MEK, constitutively active mitogen-activated protein kinase kinase; pS493, phosphoserine 493.

Neutralizing polyclonal antibody to respiratory syncytial virus (RSV) has been shown to be an effective prophylactic agent when administered intravenously in high-risk infants. This study describes the generation of a humanized monoclonal antibody, MEDI-493, that recognizes a conserved neutralizing epitope on the F glycoprotein of RSV. The affinity of MEDI-493 was found to be equal to or slightly better than an isotype-matched chimeric derivative of the parent antibody. In plaque reduction, microneutralization, and fusion-inhibition assays, MEDI-493 was significantly more potent than the polyclonal preparation. Broad neutralization of a panel of 57 clinical isolates of the RSV A and B subtypes was demonstrated. Pretreatment of cotton rats with MEDI-493 resulted in 99% reduction of lung RSV titers at a dose of 2.5 mg/kg, corresponding to a serum concentration of 25 -30 mg/mL. Further, MEDI-493 did not induce increased RSV infection or pathology in either a primary or a secondary challenge.

Therefore, an investigation was undertaken to infer genes governing Al tolerance in pea. To study the inheritance of Al tolerance, tolerant lines ‘Azad P1’ and ‘PC-55-11-1-2’ were crossed with sensitive lines ‘PC-493-5’ and ‘PSM-2’. Parental, F1, F2 and backcross generations were grown in a nutrient solution containing 30 ppm of Al for haematoxylin staining and root regrowth and classified for tolerance by staining of root tips and root regrowth. The F1 hybrids responded similarly to the tolerant parents indicating dominance of Al tolerance over sensitivity. Segregation for tolerance vs. sensitivity in F2 fitted well with the 3 : 1 ratio expected for a single gene. The backcrosses involving dominant parents showed the dominant reaction on all the plants while those involving susceptible parents segregated into one tolerant to one sensitive ratio. Experimental results showed that Al tolerance is a monogenic dominant trait that can be easily transferred to desirable lines through backcross breeding programme. An understanding of the inheritance of aluminum (Al) tolerance is important to breed for Al tolerant genotypes of pea (Pisum sativum L.). Abstract. Frequency patterns of chitinozoans, scolecodonts (polychaete jaws), and conodonts in the upper Llandovery and lower Wenlock of the Paatsalu core, western Estonia, are described. Chitinozoans are represented by nearly 50 species and constitute the most abundant group, outnumbering scolecodonts and conodonts by 10œ100 times. Jawed polychaetes with about 60 species and up to thousands of specimens per kilogram of rock are the most diverse but least varying group.

At Site 493, DSDP Leg 66, dioritic basement was reached below lower Miocene (NN1 Zone, 22-24 Ma) terrigenous sediments. Petrographical, mineralogical (including microprobe analyses), and chemical features of the dioritic rocks reveal their magmatic affinity with the calc-alkaline series. Furthermore, their radiometric age (35.3 m.y.) links the basement to the Sierra Madre Occidental in Mexico and to mid-Tertiary volcanic arcs

We assessed the efficacy of simultaneous agonism at the glucagon-like peptide-1 receptor (GLP-1R) and the melanocortin-4 receptor (MC4R) for the treatment of obesity and diabetes in rodents. Diet-induced obese (DIO) mice were chronically treated with either the long-acting GLP-1R agonist liraglutide, the MC4R agonist RM-493 or a combination of RM-493 and liraglutide. Co-treatment of DIO mice with RM-493 and liraglutide improves body weight loss and enhances glycemic control and cholesterol metabolism beyond what can be achieved with either mono-therapy. The superior metabolic efficacy of this combination therapy is attributed to the anorectic and glycemic actions of both drugs, along with the ability of RM-493 to increase energy expenditure. Interestingly, compared to mice treated with liraglutide alone, hypothalamic Glp-1r expression was higher in mice treated with the combination therapy after both acute and chronic treatment. Further, RM-493 enhanced hypothalamic Mc4r expression....

The leakage current and the conduction mechanisms in Ta O layers on silicon with thickness in the range of 20}80 nm, obtained by reactive sputtering of Ta in an Ar/O mixture have been investigated. Some dielectric and electrical properties, important for the application of the layers as storage capacitors in high-density dynamic random access memories (DRAM) and as a gate dielectric in metal oxide silicon transistors (MOSTs) are also considered. The results show that for as-deposited layers the leakage currents are higher for samples obtained at higher deposition temperature. The e!ect of postdeposition oxygen annealing depends on the thickness of Ta O layers. For thicker layers (40 nm), the leakage current after annealing increases and the e!ect is stronger for layers deposited at T "493 K. It has been established that for thinner oxides (25 nm) the annealing strongly improves the leakage currents (the density of leakage current is 10\ A/cm at applied "elds of about 1 MV/cm, which is low enough to satisfy the demands of 64 and 256 Mbit DRAM). For as-deposited samples the conduction mechanism is Poole Frenkel. After annealing depending on the "eld strength, di!erent types of conduction mechanisms occur: for electric "elds in the range 0.8}1.3 MV/cm, the conduction mechanism is dominated by electrode limited Schottky emission and for higher "elds ('1.5 MV/cm) it is bulk limited Poole Frenkel emission. The results are discussed in terms of bulk traps in the initial Ta O and their modi"cation after oxygen annealing.
