Affective states can be inferred from responses to ambiguous and threatening stimuli, using Judgement Bias Tasks (JBTs) and Attention Bias Tasks (ABTs). We investigated the separate and interactive effects of personality and housing conditions on dairy cattle affective states. We assessed personality in 48 heifers using Open-Field, Novel-Object and Runway tests. Personality effects on responses to the JBT and to the ABT were examined when heifers were housed under reference conditions. Heifers were subsequently housed under positive or negative conditions, and housing effects on animal responses in both tasks were investigated while controlling for personality. A Principal Component Analysis revealed three personality traits labelled Activity, Fearfulness and Sociability. Under reference conditions, personality influenced heifers’ responses to the JBT and to the ABT, therefore questioning the tasks’ generalizability across individuals. Internal and external feeding on zooplankton may provide scleractinian corals with important nutrients. However, the latter process has never been properly quantified. To quantify the dynamics of zooplankton capture, digestion and release for a scleractinian coral, we performed detailed video analyses of Galaxea fascicularis feeding on Artemia nauplii. A highly dynamic process of prey capture, digestion and release was observed. A single G. fascicularis polyp (N=3) captured 558±67 and released 383±75 Artemia nauplii over a 6 h interval. On average, 98.6% of prey captured was not ingested. Instead, prey items were clustered into aggregates that were digested externally by mesenterial filaments. In addition, we employed carbon, nitrogen and phosphorus analysis of zooplankton before and after digestion by G. fascicularis colonies (N=6). For total organic carbon, 43.1% (0.298±0.148 μg Artemia–1) was lost after 6 h of digestion. For total organic nitrogen, total organic phosphorus and orthophosphate (PO43–), these values were 51.3% (0.059±0.028 μg Artemia–1), 50.9% (0.009±0.004 μg Artemia–1) and 84.6% (0.0019±0.0008 μg Artemia–1), respectively. For extracoelenteric zooplankton feeding alone, total estimated nutrient inputs for G. fascicularis colonies were 76.5±0.0 μg organic carbon, 15.2±0.0 μg organic nitrogen, 2.3±0.2 μg organic phosphorus and 0.5±0.8 μg inorganic phosphorus per cm2 coral tissue per day. These values exceed calculations based on intracoelenteric feeding by up to two orders of magnitude. Our results demonstrate that extracoelenteric zooplankton feeding is a key mechanism of nutrient acquisition for a scleractinian coral. These results are of importance to coral aquaculture and our understanding of benthic–pelagic coupling on coral reefs.

"Background

In differentiated gonochoristic species, a bipotential gonad develops into an ovary or testis during sex differentiation. Knowledge about this process is necessary to improve methods for masculinizing genetically female Atlantic cod for the subsequent purpose of producing allfemale populations.

Methods

Gonads were examined histologically in juveniles from 14 to 39 mm total body length (TL). Number and size of germ cells were determined in a subset of the samples. Relevant genes were cloned, and mRNA levels determined by qPCR of amh, cyp19a1a; dax1 (nr0b2); shp (nr0b2a) and sox9b in a mixed-sex and an all-female population ranging from 12-49 mm TL.

Results

Individuals between 14-20 mm TL could be separated in two subgroups based on gonad size and germ cell number. Ovarian cavity formation was observed in some individuals from 18- 20 mm TL. The mixed sex population displayed bimodal expression patterns as regards cyp19a1a (starting at 12 mm TL) and amh (starting at 20 mm TL) mRNA levels. After approximately 30 mm TL, cyp19a1a and amh displayed a gradual increase in both sexes. No apparent, sex-dependent expression patterns were found for dax1, shp or sox9b transcripts. However, shp levels were high until the larvae reached around 35 mm TL and then dropped to low levels, while dax1 remained low until 35 mm TL, and then increased sharply.

Conclusions

The morphological sex differentiation in females commenced between 14-20 mm TL, and ovarian cavities were evident by 18-20 mm TL. Testis development occurred later, and was morphologically evident after 30 mm TL. This pattern was corroborated with sexually dimorphic expression patterns of cyp19a1a from 12-13 mm TL, and a male-specific increase in amh from 20 mm TL."

Many scleractinian coral species host epizoic acoelomorph
flatworms, both in aquaculture and in situ. These symbiotic
flatworms may impair coral growth and health through lightshading, mucus removal and disruption of heterotrophic
feeding. To quantify the effect of epizoic flatworms on
zooplankton feeding, we conducted video analyses of single
polyps of Galaxea fascicularis (Linnaeus 1767) grazing on
Artemia nauplii in the presence and absence of symbiotic
flatworms. 18S DNA analysis revealed that flatworms
inhabiting G. fascicularis belonged to the genus Waminoa
(Convolutidae), which were hosted at a density of 3.660.4
individuals polyp21. Polyps hosting flatworms exhibited prey
capture rates of 2.262.5, 3.464.5 and 2.763.4 nauplii polyp21
30 min21 at prey concentrations of 250, 500 and 1,000 nauplii
L21, respectively. Polyps that had their flatworms removed
displayed prey capture rates of 2.761.6, 4.864.1 and 16.9610.3
nauplii polyp21 30 min21. Significant main and interactive
effects of flatworm presence and ambient prey concentration
were found, reflected by the fact that flatworms significantly
impaired host feeding rates at the highest prey density of 1,000
nauplii L21. In addition, flatworms displayed kleptoparasitism,
removing between 0.160.3 and 0.661.1 nauplii 30 min21 from
the oral disc of their host, or 5.363.3 to 50.062.1% of prey
acquired by the coral. We suggest classifying the coralassociated
Waminoa sp. as an epizoic parasite, as its presence
may negatively affect growth and health of the host.

Several factors may affect heterotrophic feeding of benthic marine invertebrates, including water flow rate and polyp context (i.e. the presence of neighbouring polyps). We tested the interactive effects of water flow rate and polyp context on zooplankton feeding by the scleractinian coral Galaxea fascicularis. Single polyps and colonies were incubated in a flow cell for 30 minutes with an ambient Artemia nauplii concentration of 10,000 L-1 and water flow rates ranging from 1.25 to 40 cm s-1. Water flow rate and polyp context showed significant main and interactive effects on feeding rates of G. fascicularis polyps. More specifically, feeding rates were optimal at flow rates of 1.25 cm s-1 for single polyps and 5 to 10 cm s-1 for polyps inhabiting colonies. The presence of epizoic acoelomorph flatworms may have negatively affected the observed feeding rates, especially at high flow. Our results demonstrate that water flow affects coral feeding and thus heterotrophic nutrient input at both a polyp and colony level. These findings are of relevance to our understanding of how biotic and abiotic factors interact on coral heterotrophy, and may serve to optimise coral aquaculture.

Heterotrophy is known to stimulate calcification of scleractinian corals, possibly through enhanced organic matrix synthesis and photosynthesis, and increased supply of metabolic DIC. In contrast to the positive long-term effects of heterotrophy, inhibition of calcification has been observed during feeding, which may be explained by a temporal oxygen limitation in coral tissue. To test this hypothesis, we measured the short-term effects of zooplankton feeding on light and dark calcification rates of the scleractinian coral Galaxea fascicularis (n = 4) at oxygen saturation levels ranging from 13 to 280%. Significant main and interactive effects of oxygen, heterotrophy and light on calcification rates were found (three-way factorial repeated measures ANOVA, p<0.05). Light and dark calcification rates of unfed corals were severely affected by hypoxia and hyperoxia, with optimal rates at 110% saturation. Light calcification rates of fed corals exhibited a similar trend, with highest rates at 150% saturation. In contrast, dark calcification rates of fed corals were close to zero under all oxygen saturations. We conclude that oxygen exerts a strong control over light and dark calcification rates of corals, and propose that in situ calcification rates are highly dynamic. Nevertheless, the inhibitory effect of heterotrophy on dark calcification appears to be oxygen-independent. We hypothesize that dark calcification is impaired during zooplankton feeding by a temporal decrease of the pH and aragonite saturation state of the calcifying medium, caused by increased respiration rates. This may invoke a transient reallocation of metabolic energy to soft tissue growth and organic matrix synthesis. These insights enhance our understanding of how oxygen and heterotrophy affect coral calcification, both in situ as well as in aquaculture.

Light spectrum plays a key role in the biology of symbiotic corals, with blue light resulting in higher coral growth, zooxanthellae density, chlorophyll a content and photosynthesis rates as compared to red light. However, it is still unclear whether these physiological processes are blue-enhanced or red-repressed. This study investigated the individual and combined effects of blue and red light on the health, zooxanthellae density, photophysiology and colouration of the scleractinian coral Stylophora pistillata over 6 weeks. Coral fragments were exposed to blue, red, and combined 50/50% blue red light, at two irradiance levels (128 and 256 mmol m 22 s 21 ). Light spectrum affected the health/survival, zooxanthellae density, and NDVI (a proxy for chlorophyll a content) of S. pistillata. Blue light resulted in highest survival rates, whereas red light resulted in low survival at 256 mmol m 22 s 21 . Blue light also resulted in higher zooxanthellae densities compared to red light at 256 mmol m 22 s 21 , and a higher NDVI compared to red and combined blue red light. Overall, our results suggest that red light negatively affects the health, survival, symbiont density and NDVI of S. pistillata, with a dominance of red over blue light for NDVI.

Coral reefs are essential to many nations, and are currently in global decline. Although climate models predict decreases in seawater pH (,0.3 units) and oxygen saturation (,5 percentage points), these are exceeded by the current daily pH and oxygen fluctuations on many reefs (pH 7.8-8.7 and 27-241% O 2 saturation). We investigated the effect of oxygen and pH fluctuations on coral calcification in the laboratory using the model species Acropora millepora. Light calcification rates were greatly enhanced (+178%) by increased seawater pH, but only at normoxia; hyperoxia completely negated this positive effect. Dark calcification rates were significantly inhibited (51-75%) at hypoxia, whereas pH had no effect. Our preliminary results suggest that within the current oxygen and pH range, oxygen has substantial control over coral growth, whereas the role of pH is limited. This has implications for reef formation in this era of rapid climate change, which is accompanied by a decrease in seawater oxygen saturation owing to higher water temperatures and coastal eutrophication.

Psychrophilic (6-15°C) anaerobic treatment of malting wastewater was investigated. A two-module expanded granular sludge bed reactor system with a total volume of 140 dm 3 was used to treat malting wastewater having a soluble and total chemical oxygen demand (COD) between 233 and 1778 mg dm -3 and between 317 and 4422 mg dm -3 , respectively. The removal efficiencies at 6°C were 47 and 71% of the soluble and volatile fatty acids (VFA) COD, at organic loading rates (OLR) ranging between 3.3 and 5.8 kg of COD m -3 day -1 . The removal efficiencies at 10-15°C were 67-78 and 90-96% of the soluble and VFA COD at an OLR between 2.8 and 12.3 kg of COD m -3 day -1 . The specific methanogenic activity of the sludge present in each module increased 2-3-fold during system operation for 400 days. The relatively high concentration of suspended solids in the influent (25% of the total COD) caused a deterioration of the sludge bed in the first reactor module. This was aggravated by excessive growth of acidifying biomass, which persisted in the first module sludge bed and resulted in granular sludge flotation. However, the second module could accommodate the increased OLR, thus providing a very high effluent quality (soluble COD < 200 mg dm -3 ) of the total system. The stability of module I concerning suspended solids could be restored by presettling the wastewater.

An experiment was performed to investigate the effect of animal versus vegetable protein sources in the diet on the development of behavior in laying hens. A diet containing protein sources of only vegetable origin was compared with four diets, each containing one of four processed animal proteins (PAPs). Two PAPs (Daka-58 and Sonac-60) were classified as meat meals, whereas the remaining ones (Daka-40 and Sonac-50) were classified as meat and bone meals. The impact of a control diet (without PAP) versus four PAP diets (50 g/kg CP of animal origin) on behavior was determined. All diets were isocaloric (AMEn = 11.8 MJ/kg) and isonitrogenous (dig. lysine = 6.8 g/kg). Hens were housed in 40 floor pens (12 hens/pen, 8 pens/treatment) from 20 to 40 weeks of age. Supplementation of PAPs did not generally reduce feather pecking behavior. Nevertheless, Daka-40 and Sonac-50 fed hens showed a delay in the development of feather damage and, simultaneously, an increase in litter condition, foraging and walking behavior, and floor pecks compared to hens fed Sonac-60. These shifts seemed to be partly related with the intake of digestible glycine, available phosphorus, calcium, potassium, and sodium.

An experiment was conducted to investigate the effect of a prebiotic on performance of partridge. The experiment was carried out with a total of eighty-day-old male Chukar partridge (Alectoris chukar chukar) chicks in a completely randomized design. The dietary treatments consisted of a control and an experimental treatment, and each treatment was replicated four times with 10 chicks per replicate. The experimental period lasted 16 weeks with a starter period (0-8 weeks) and a grower period (9-16 weeks). The control group was fed the basal diet, while the experimental group was fed the basal diet supplemented with 0.18% prebiotic Aspergillus meal in each period. Performance parameters included body weight gain, cumulative feed consumption and cumulative FCR and were recorded at biweekly intervals. Carcass characteristics and blood parameters were measured at the end of the experiment (week 16). Although the results showed that prebiotic had no significant effect on body weight gain and cumulative feed conversion ratio; however, the supplementation of Aspergillus meal significantly (p < 0.05) decreased cumulative feed consumption. Also prebiotic significantly increased percentages of breast and gastrointestinal tract, decreased percentage of back-neck, decreased blood triglyceride and total cholesterol content and increased blood calcium content. From this study, it was concluded that dietary supplementation of 0.18% Aspergillus meal might offer beneficial effects on partridge feed consumption, carcass quality and blood cholesterol.

The effects of ambient temperature (T; 18 vs. 28°C), dietary energy level 90, 100, and 110% of NRC (1994) recommendations], and dietary lysine level [LYS; 105 vs. 120% of NRC (1994) recommendations], and their interactions on feed intake, BW gain, feed:gain ratio, and carcass yields of male turkeys were studied from 29 to 140 d of age. The experiment was designed as a split plot, including T as the main plot and E and LYS as the subplot, with 60 pens containing 10 male turkeys each. Overall, feed intake, BW gain, and feed:gain ratio were significantly lower at high compared with low T. Feed intake decreased linearly as E increased and was more pronounced at low compared with high T. Metabolizable energy intake increased more at high compared with low T as E increased. Turkeys that were fed the highest E gained less weight until 84 d of age than those fed the lowest E. An experiment with male turkeys was conducted to test the hypothesis that turkey production performance responds positively to extra crystalline amino acid supplementation (lysine, methionine, and threonine) when subjected to a high ambient temperature regimen (HT) in the grower period. Two diets were formulated to provide lysine, methionine, and threonine concentrations that either 1) met the breeder recommendations or 2) contained 10% higher lysine and methionine concentrations from 22 to 134 d of age and 10% higher threonine concentration from 22 to 68 d of age. Both diets were fed at two temperatures (15 or 25 C) from 42 d of age onward.

The effects of ambient temperature (T; 15 C vs. 30 C from 6 wk of age onwards), dietary Arg:Lys ratio (Arg:Lys ratio; 1.00 vs. 1.25), dietary electrolyte balance (DEB: 164 vs. 254 meq/kg), and their interactions on growth performance and carcass yields of male turkeys were studied. The experiment was designed as a split plot, including T × DEB as the main plot and Arg:Lys ratio as the sub-plot, with 24 pens containing 35 male turkeys each. Feed consumption, BW gain, mortality, and processing yields were measured. Temperature had a clear effect on performance during all age periods. Feed intake was significantly lower for the high T group compared with the low T group (322.7 vs. 432.3 g/bird per day; P < 0.001).
