For anybody else having this issue, using the Inspect Element of Google Chrome pinpointed this to a SMTP plugin still being accidentally in debug mode and reporting lots of SMTP responses back which were interpreted as a fail. Disabling the debugging has fixed this.
I will mark as resolved.

Whenever I submit a comment as a user either logged in or not (and not as admin), I get the following error: “Sorry, but there was an error posting your comment. Please try again later.” However, the comment is sat in wp-admin in an unapproved state awaiting approval (and I get the email), so it hasn’t exactly failed. I have just confirmed I get the same error on your demo site also (tested here http://demo.themepacific.com/plugin-tiled-gallery-carousel/2013/10/13/tiled-gallery-carousel-without-jetpack-wordpress-plugin/#jp-carousel-10). It also happens in WordPress 2.1.18, so it’s not a new issue. I do not have Askimet enabled and no other spam filters.
I have seen various forum posts which point to a patch, but when checking the jetpack-carousel.php file, this patch has been applied, and does not seem to fix this issue.
I cannot work out what it is semi-failing on, and am wrongly tempted to just re-word line 119:
'comment_post_error' => __( 'Sorry, but there was an error posting your comment. Please try again later.', 'themepacific_gallery' ),which is obviously very dangerous in the long run.
It would be great if a fix could be found for this so I don’t have to resort to such drastic measures.
