I found following lines only on index page but not on any other pages.

It don’t seem that pages are cached because website take lots of time to load.

21 seconds is too long, you may need to fix that first.

Is this a Supercache issue? Does the site load fast without Supercache enabled?

Site do not load fast without Supercache,so I installed supercache plugin.

I don’t think Supercache was designed for this. If your pages are taking longer than 20 seconds to load, you may need to clean up your code, plugins, etc. as most visitors won’t wait that long.
I guess you may be running into timeout issues…
