Enrichment of live foods by essential nutrients such as highly unsaturated fatty acids is an impo... more Enrichment of live foods by essential nutrients such as highly unsaturated fatty acids is an important tool for improvement of larval fish quality. In this study, nutritional effects of Artemia urmiana enriched by fish and plant oils on growth, survival rate, body compositions and resistance against thermal, salinity and hypoxic stresses in rainbow trout, Oncorhynchus mykiss were examined. Six food treatments comprising a commercial feed, non-enriched Artemia nauplii and the nauplii enriched by either of fish, sunflower, canola and soybean oils were used in triplicates. The fish fed with Artemia enriched by fish, sunflower and canola oils had significantly higher (p<0.05) survival rate, total length, wet and dry weights, specific growth rate and lower food conversion ratio than those fed the commercial feed. However, there were no significant differences in growth indices between the fish fed fish oil-enriched and plant oil-enriched Artemia. Minimum eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were reported in the fish fed with the commercial feed, while maximum EPA and DHA were in the fish fed canola oil-enriched and nonenriched Artemia, respectively. The fish fed canola oil enriched Artemia had significantly higher (p<0.05) resistance against environmental stresses compared to fish fed the commercial diet. Results showed that the plant oils, mainly canola and sunflower oils can be used for Artemia enrichment as a suitable substitute for the more expensive and rare fish oil for improving growth and resistance to environmental stresses in rainbow trout larvae.

A 30-day study was carried out to determine the influence of different weaning strategies (WS) on... more A 30-day study was carried out to determine the influence of different weaning strategies (WS) on growth and survival rates, stress resistance, digestive enzyme activities and whole body fatty acid profiles in yellowfin seabream (Acanthopagrus latus) larvae. Thus, eight WS were designed by combining live foods (LF) (rotifers and Artemia) and a microdiet (MD) (Gemma Micro TM , Skretting). In the control group (100 LF), larvae were fed only LF (100%), including enriched rotifers (from 2 to 20 days post hatching, dph) and Artemia (nauplii + enriched metanauplii, from 18 to 30 dph). The other treatments were based on different proportions of LF with the MD: 75LF-25MD, 50LF-50MD, 25LF-75MD and 100 MD in which 25, 50, 75 and 100% of LF that applied in the control group were replaced with a fixed ration of MD, respectively. In addition, for determining the appropriate time of weaning, three WS were applied including: sudden weaning (SW): larvae were weaned on to MD with only rotifer from 2 to 5 days post hatch (dph); early weaning (EW): larvae were weaned on to MD with only rotifer from 2 to 14 dah and late weaning (LW): larvae were weaned on to MD with rotifer (from 2 to 14 dph) and Artemia nauplii (from 12 to 20 dph). Larvae were hand-fed with the MD hourly from 08:00 h to 18:00 h (11 times a day). Results showed that the lowest survival rates were found among larvae from 100MD (1.5%) and SW (1.9%) groups, whereas the highest survival rates were found in larvae from 100LF (31.0 ± 1.7%), 75LF-25MD (34.7 ± 4.5%) and 50LF-50MD (36.2 ± 4.0%). Larvae from the 50LF-50MD group showed the highest resistance against air exposure (70.0%) and osmotic stress (100.0%) challenges. At the end of the trial, larvae from 100LF, 75LF-25MD, 50LF-50MD and LW groups showed higher growth performance than the other groups. At 30 dph, larvae from the LW group had the highest level of alkaline phosphatase to leucine-alanine peptidase ratio, while those from the 100MD, SW and EW treatments showed the lowest ones. The fatty acid profile of larvae reflected that of the diet. In particular, larvae from 100MD and SW groups had generally lower levels of n-3 long chain polyunsaturated fatty acids compared to the other treatments. In conclusion, results of the present study indicated the time of weaning and proportion of LF and MD remarkably affect larvae performance in terms of growth, survival, digestive function and stress tolerance. In addition, the replacement of LF with MD up to 50% after mouth opening of larvae neither have detrimental effects on growth, survival and digestive enzymes activities, meanwhile enhanced stress resistance of A. latus larvae.

Zearalenone (ZEA) as mycoestrogen is found in human foods and animal feeds. Its estrogenic potenc... more Zearalenone (ZEA) as mycoestrogen is found in human foods and animal feeds. Its estrogenic potency depends on its biotransformation fate. The hepatic biotransformation of ZEA in two species of sturgeon fish (Acipenser persicus and Huso huso) was investigated. ZEA was incubated with the hepatic microsomal and postmitochondrial sub-fractions in the presence of NADPH and the metabolites were determined by means of HPLC. Moreover, the rate of glucuronidation for ZEA and its metabolites were estimated in the presence of uridine diphosphateglucuronic acid. zearalenol (-ZOL) was found to be the major metabolite of ZEA by both subfractions. Enzymatic kinetics studies revealed that the maximum velocity (Vmax) in microsomal and post-mitochondrial fractions for -ZOL production was found 5-and 7-folds in Huso huso and 8-and 12-folds in A. persicus higher than that for -ZOL production, respectively. The H. huso hepatic post-mitochondrial fraction mainly glucurinated ZEA while in A. persicus, the metabolites and in particular -ZOL were glucuronidated. Data suggest that the hepatic biotransformation of ZEA in studied sturgeons resulted in detoxification of ZEA as the main metabolite tends to be -ZOL with weaker estrogenic property. Moreover, clear differences in glucuronidation profile are indicating interspecies variety in hepatic biotransformation of ZEA.

The importance of start feeding on growth, survival and quality of larvae and role of Artemia as ... more The importance of start feeding on growth, survival and quality of larvae and role of Artemia as a nutritious live food with ability of carrying various essential nutrients is a well documented fact. In this study the possibility of replacing fish oil with vegetable oils for enrichment of A. urmiana nauplii and its use in start feeding of rainbow trout larvae were investigated. Six feeding treatments were used in two phases to feed the rainbow trout larvae from the beginning of the exogenous feeding for a period of two months. The feeding treatments during the first 10 10 days included: 1) Commercial diet, 2) A. urmiana nauplii enriched with fish oil, 3) A. urmiana nauplii enriched with Sunflower oil, 4) A. urmiana nauplii enriched with Canola oil, 5) A. urmiana nauplii enriched with Soybean oil, 6) Newly hatched un-enriched A. urmiana nauplii. The larvae in all groups were fed on commercial diet during the second phase from day 11 until end the experiment. Survival percentage of the larvae fed on Artemia nauplii enriched with canola and sunflower oils were significantly higher than those fed on commercial diet. Total length, wet weight and dry weight of the larvae in all treatments received Artemia nauplii during first phase were significantly higher compared to those fed on commercial feed from the beginning. Highest growth was obtained in the larvae fed on Artemia nauplii enriched with canola oil. The results of this research showed that vegetable oils can play essential role in enrichment of Artemia nauplii and feeding the rainbow trout.

Its estrogenic potency mainly depends on its biotransformation fate. The hepatic biotransformation of ZEA in rainbow trout was investigated in this study. Various concentrations of ZEA were separately incubated with the hepatic microsomal and post-mitochondrial sub-fractions in the presence of NADPH, and the metabolites were determined by means of HPLC. Moreover, the rate of glucuronidation for ZEA and its reduced metabolites were estimated in the presence of uridine diphosphate glucuronic acid. β-zearalenol (β-ZOL) was found to be the major metabolite of ZEA by both sub-cellular fractions. The enzymatic kinetics analyses indicated that the α-ZOL and β-ZOL production by microsomal fraction were 8-and 2-fold higher than those by postmitochondrial fraction, respectively. High percentages of ZEA and its metabolites are conjugated with glucuronic acid at the lower concentrations. Data suggest that the hepatic biotransformation of ZEA in rainbow trout resulted in its detoxification as the main metabolite tends to be β-ZOL with weak estrogenic property. Moreover, at certain concentrations, the produced metabolites are entirely conjugated with glucuronic acid, which may consequently cause a prolonged duration of action due to entero-hepatic cycle.

In order to find a marker for differentiating between a bisexual and a parthenogenetic Artemia st... more In order to find a marker for differentiating between a bisexual and a parthenogenetic Artemia strain, Exon-7 of the Na ⁄ K ATPase a 1 subunit gene was screened by RFLP technique. The results revealed a constant synonymous SNP (single nucleotide polymorphism) in digestion by the Tru1I enzyme that was consistent with these two types of Artemia. This SNP was identified as an accurate molecular marker for discrimination between bisexual and parthenogenetic Artemia. According to the Nei's genetic distance (1973), the lowest genetic distance was found between individuals from Artemia urmiana Gü nther 1890 and parthenogenetic populations, making the described marker the first marker to easily distinguish between these two cooccurring species.

The simultaneous effects of processing factors such as ripening time (25-75 days), ripening tempe... more The simultaneous effects of processing factors such as ripening time (25-75 days), ripening temperature (4-14°C) and brine concentration (10-13%) on biogenic amines content, proteolysis and sensory score of Iranian white brine cheese were studied, in 12 cheeses. Response surface methodology (RSM) was used to minimise biogenic amines content. At low level of ripening time, biogenic amines content decreased with increasing levels of brine concentration but at high level of ripening time, brine concentration had inverse effect. Ripening time showed quadratic effect on biogenic amines content. Based on biogenic amines content and sensory score, the optimum conditions were 13% brine and ripening at 9-14°C for 43-65 days.

The present study was performed to elucidate the effects of total replacement of fish oil with ve... more The present study was performed to elucidate the effects of total replacement of fish oil with vegetable oils on growth performance and foregut histological indices of the rainbow trout. Fishes with average initial weight of 15.10±0.20 g were transferred into 15 (300 L) tanks (50 fishes/tank) and reared for 60 consecutive days using different experimental diets. Kilka fish oil (FO) was fully replaced with different vegetable oil sources including canola oil (CO), safflower oil (SO), linseed oil (LO) and a mixture (CSLO) of CO (40%), SO (30%) and LO (30%) as dietary lipid sources. The samples were fixed at 10% neutral-buffered formalin and paraffin sections were prepared and stained with hematoxylin and eosin and periodic acid-Schiff methods. Growth performances in CO and CSLO groups were better than other groups. Moreover, in these groups, the number of goblet cells was decreased; however thickness and height of intestinal folds and thickness of tunica mucosa and tunica sub-mucosa were increased in comparison to FO group. Overall, performance and histological findings did not show any negative effect due to canola and mixed vegetable oils feeding compared to FO group.

Data suggest that the hepatic biotransformation of ZEA in rainbow trout resulted in its detoxification as the main metabolite tends to be β-ZOL with weak estrogenic property. Moreover, at certain concentrations, the produced metabolites are entirely conjugated with glucuronic acid, which may consequently cause a prolonged duration of action due to entero-hepatic cycle.

In order to find a marker for differentiating between a bisexual and a parthenogenetic Artemia st... more In order to find a marker for differentiating between a bisexual and a parthenogenetic Artemia strain, Exon-7 of the Na ⁄ K ATPase a 1 subunit gene was screened by RFLP technique. The results revealed a constant synonymous SNP (single nucleotide polymorphism) in digestion by the Tru1I enzyme that was consistent with these two types of Artemia. This SNP was identified as an accurate molecular marker for discrimination between bisexual and parthenogenetic Artemia. According to the Nei's genetic distance (1973), the lowest genetic distance was found between individuals from Artemia urmiana Gü nther 1890 and parthenogenetic populations, making the described marker the first marker to easily distinguish between these two cooccurring species.

The simultaneous effects of processing factors such as ripening time (25-75 days), ripening tempe... more The simultaneous effects of processing factors such as ripening time (25-75 days), ripening temperature (4-14°C) and brine concentration (10-13%) on biogenic amines content, proteolysis and sensory score of Iranian white brine cheese were studied, in 12 cheeses. Response surface methodology (RSM) was used to minimise biogenic amines content. At low level of ripening time, biogenic amines content decreased with increasing levels of brine concentration but at high level of ripening time, brine concentration had inverse effect. Ripening time showed quadratic effect on biogenic amines content. Based on biogenic amines content and sensory score, the optimum conditions were 13% brine and ripening at 9-14°C for 43-65 days.

The present study was performed to elucidate the effects of total replacement of fish oil with ve... more The present study was performed to elucidate the effects of total replacement of fish oil with vegetable oils on growth performance and foregut histological indices of the rainbow trout. Fishes with average initial weight of 15.10±0.20 g were transferred into 15 (300 L) tanks (50 fishes/tank) and reared for 60 consecutive days using different experimental diets. Kilka fish oil (FO) was fully replaced with different vegetable oil sources including canola oil (CO), safflower oil (SO), linseed oil (LO) and a mixture (CSLO) of CO (40%), SO (30%) and LO (30%) as dietary lipid sources. The samples were fixed at 10% neutral-buffered formalin and paraffin sections were prepared and stained with hematoxylin and eosin and periodic acid-Schiff methods. Growth performances in CO and CSLO groups were better than other groups. Moreover, in these groups, the number of goblet cells was decreased; however thickness and height of intestinal folds and thickness of tunica mucosa and tunica sub-mucosa were increased in comparison to FO group. Overall, performance and histological findings did not show any negative effect due to canola and mixed vegetable oils feeding compared to FO group.

NMR-based metabolomics was applied to explore metabolic impacts of diazinon on sea water adaptati... more NMR-based metabolomics was applied to explore metabolic impacts of diazinon on sea water adaptation of Persian sturgeon fingerlings, Acipenser persicus. Fingerlings were exposed to sub-lethal concentrations of diazinon in freshwater (FW) for 96 h (short-term trial) and 12 days (long-term trial) and then exposed in brackish water (BW) (12 mg L-1 salinity) for 24 h. After 96 h and 12 days exposure in FW, identified metabolites (amino acids, osmolytes, energy metabolites) showed different change-patterns compared to control group (P &amp;amp;lt; 0.05) as follow: (A) short-term trial: higher plasma levels of glucose, lactate (in all diazinon-exposed fish), acetate and acetoacetate (in 0.9 mg L-1diazinon treatment); lower levels of creatine (in all diazinon-exposed fish), trimethylamine-N-oxide, choline, taurine, betaine, N,N-dimethylglycine and almost all amino acids in fish exposed to high concentrations of diazinon (0.54 and 0.9 mg L-1 diazinon). (B) Long-term trial: higher plasma levels of lipid oxidation metabolites and almost all amino acids in fish exposed to 0.54 and 0.9 mg L-1 diazinon; lower levels of creatine, trimethylamine-N-oxide, N,N-dimethylglycine, betaine, choline (in all diazinon-exposed fish), glucose (in 0.54 and 0.9 mg L-1diazinon treatments) and taurine (in 0.9 mg L-1 diazinon treatment). When fish were exposed in BW for 24 h, the plasma levels of osmolytes decreased significantly in almost all experimental groups of short-term and long-term trial (P &amp;amp;lt; 0.05). In short-term trial, the plasma levels of glucose in all groups and lactate in 0.18 and 0.54 mg L-1 diazinon treatments increased after salinity challenge (P &amp;amp;lt; 0.05). However, a significant decrease was observed in lactate levels in 0.9 mg L-1 diazinon treatment (P &amp;amp;lt; 0.05). Also, the plasma levels of amino acids decreased mostly in fish of control group than exposed fish (P &amp;amp;lt; 0.05). The plasma glycerol concentration showed a significant decrease only in fish of 0.54 mg L-1 diazinon treatment (P &amp;amp;lt; 0.05). In long term trial, the energetic metabolites (acetate, acetoacetate, glycerol) showed significant increases mostly in fish exposed to high concentrations of diazinon (P &amp;amp;lt; 0.05). Phosphocreatine was detected only in groups exposed to 0.54 and 0.9 mg L-1 diazinon. Some amino acids decreased in control and diazinon-exposed groups while glycine (in control and 0.18 mg L-1 diazinon treatment), glutamine and alanine (in 0.9 mg L-1 diazinon treatment) elevated significantly after 24 h acclimation in BW (P &amp;amp;lt; 0.05). Our results may help to understand the effects of pesticides on fish osmoregulation from a metabolic approach.
