This paper presents an overview of the main actions and projects of the ANR TECSAN "CancerSensor"... more This paper presents an overview of the main actions and projects of the ANR TECSAN "CancerSensor" during the 2011-2014 period. The project aims to develop and validate a quantitative, non-invasive therapeutic tool to detect selected urinary modified nucleosides as biomarkers of colorectal cancer chemotherapy and to monitor in fine the efficiency of the chemotherapy. A methodology has been developed to generate a bulk acoustic wave molecularly imprinted polymer sensor (SAW-MIP) for the detection of various nucleosides and nucleotides of interest. A rapid and sensitive mass spectrometry method has been also developed.

Studies examining the mechanisms of hyperoxia-induced airway hyperresponsiveness have focused on contractile mechanisms, although excessive airway narrowing could be due to impaired relaxation. Our objective was to determine the effects of hyperoxia on airway structure and relaxing properties in juvenile rats exposed to an inspired fraction of oxygen (FiO(2)) of 0.5 within 24 hr of birth for 15 days, compared to a control group of air-exposed rats. We studied the 1) tracheal smooth muscle surface area; and 2) in vitro relaxation in precontracted (carbachol, 1.10(-3) M) isolated tracheal rings to increasing cumulative concentrations (10(-8) to 10(-4) M) of salbutamol. There was no significant difference in the amount of smooth muscle between the hyperoxia and air-exposed groups. Maximal relaxation of the isolated trachea to salbutamol was greater in the hyperoxia group than in the air-exposed group when normalized to smooth muscle surface area (-12.69 +/- 3.51 g/mm(2) vs. - 8.49 +/- 1.67 g/mm(2); P (MANOVA) < 0.05) or expressed as maximal relaxation induced by 10(-3) M theophylline (61% vs. 28%; P < 0.05). In conclusion, salbutamol-induced relaxation is enhanced (and not impaired) in newborn rats exposed to prolonged moderate hyperoxia.
This work deals with the design of a highly sensitive acoustic Love waves biosensor for heavy met... more This work deals with the design of a highly sensitive acoustic Love waves biosensor for heavy metal detection in liquid medium. Biorecepting Escherichia coli bacteria are immobilized on polyelectrolyte multilayers, and are used to detect the presence of cadmium by modification of viscoelastic properties of bacteria. The different setups of sensor elaboration are measured through the real-time resonant frequency shifts of the sensor. The impact of the addition of a titania mesoporous coating is studied, and compared to previous results [1]. Frequency variation for bacteria immobilization is doubled with titania compared with bare silicon dioxide surface. The improvement of the acoustic platform sensitivity is highlighted with cadmium as typical heavy metal. A complementary study was done by Atomic Force Microscopy (AFM) and allowed us to validate the assumptions.
