Vibrio spp. bacteria species are heterotrophic and halophilic bacteria, which are commonly found in various habitats such as brackish, marine waters and animals. They are widely used as an indicator of water quality and cultured animal health. This study investigates the diversity and abundance of Vibrionaceae populations in the rearing water of three distinct consumer types of aquaculture commodities, namely abalone, lobster and sea cucumber, during six months sampling. We isolated Vibrionaceae bacteria from the rearing tanks of three different consumer types of aquaculture commodities. We then inoculated 100 µL of undiluted to 10-5 diluted water samples onto petriplate containing selective Thiosulphate Bile Salt Sucrose agar (TCBS). Techniques for producing Holothuria scabra or sandfish sea cucumbers from aquaculture have been developed rapidly in the recent years to address the issues of decreasing sandfish population and increasing market demand. During the process of culturing H. scabra, the early stages of sandfish (i.e. larvae and juveniles) are the most critical of the development. The gold mining wastewater effluent is potential source for isolating cyanides-degrading bacteria, since cyanide is commonly used in gold extraction process in the mining industry. An indigenous bacterial strain LP3, capable of growing on and utilizing of a high concentration of cyanomethane (up to 1.0 Molar), could be isolated from Cikotok gold mine effluent. Based on 16S rDNA sequence, the strain was identified as Rhodococcus pyridinivorans. During the growth on cyanomethane (CH3CN), ethanamide (CH3ONH2) and ethanoic acid (CH3COOH) were detected in the growth media, indicating that nitrile hydratase and amidase involved in the metabolism of the substrate. Plantaricins, one of bacteriocin produced by Lactobacillus plantarum, are already known to have activities against several pathogenic bacterium. L. plantarum U10 isolated from Btempoyak,^an Indonesian fermented food, produced one kind of plantaricin designated as plantaricin W (plnW). The plnW is suggested as a putative membrane location of protein and has similar conserved motif which is important as immunity to bacteriocin itself. Thus, due to study about this plantaricin, several constructs have been cloned and protein was analyzed in Lactococcus lactis. In this study, plnW gene was successfully cloned into vector NICE system pNZ8148 and created the transformant named L. lactis NZ3900 pNZ8148-WU10. PlnW protein was 25.3 kDa in size. The concentration of expressed protein was significantly increased by 10 ng/mL nisin induction. Furthermore, PlnW exhibited protease activity with value of 2.22 ± 0.05 U/mL and specific activity about 1.65 ± 0.03 U/mg protein with 50 ng/mL nisin induction. Immunity study showed that the PlnW had immunity activity especially against plantaricin and rendered L. lactis recombinant an immunity broadly to other bacteriocins such as pediocin, fermentcin, and acidocin.

Lactobacillus plantarum SK(5) is lactic acid bacteria that isolated from bekasam
and has lipases activity. The purpose of this study was to produce lipases of       
L. plantarum SK(5) from Bekasam and to characterize the lipases based on pH
and temperature treatments. This study was divided into three stages: 1)
verification of SK(5) isolate; 2) lipases production from L. plantarum SK(5); and
3) lipases characterization of elected treatment. Isolate L. plantarum SK(5) shows
an ability to degrade lipids in olive oil on the early phase of isolate growth and the
isolate will produce lipases during those isolate growth (48 hours). The activity of
lipases of L. plantarum SK(5) was the highest observed at the early of stationary
phase that hour of the 30th of 25,417±3,683 units/mg. Lipases that produced at
30th hour has an activity in the range of pH of 4-9 and the range of temperature of
40-80 °C.
