Vibrio alginolyticus and Streptococcus agalactiae are important bacterial pathogens that yielded ... more Vibrio alginolyticus and Streptococcus agalactiae are important bacterial pathogens that yielded high losses in Nile tilapia in Egypt. The present study aimed to check the protective efficacy of inactivated whole-cell bivalent vaccines against these pathogens using incomplete Freund's adjuvant and Montanide™ IMS 1312 VG as adjuvants. The antibody titers have been determined at different weeks post-vaccination (WPV). Moreover, the protection levels against the challenged bacterial pathogens have also been examined in relation to the time-dependent protection at different WPV. The results revealed that serum antibodies were generated in all immunized fish at 1st WPV, peaked at 4th WPV, continued, and gradually decreased from 6th WPV to 14th WPV in all vaccinated groups. In addition, vaccines induced significantly higher protection of the immunized tilapia, manifested by higher survival rates. We noticed that the antibody levels and survival rates of the vaccinated fish by a vaccine adjuvanted by Montanide™ IMS 1312 VG were higher than those produced by a vaccine adjuvanted by incomplete Freund's adjuvant at different time points. Moreover, no external clinical signs, visceral adhesions, or internal lesions were recorded in the vaccinated tilapia, demonstrating the safety of the formulated vaccines. According to the aforementioned findings, we could suggest that the prepared bivalent vaccines, using the two adjuvant types, are safe and highly protective and could be utilized as promising candidate vaccines to increase the resistance of Nile tilapia against V. alginolyticus and S. agalactiae infections. Moreover, Montanide™ IMS 1312 VG enhanced the immuno-protectivity and exhibited optimum immune response and earlier protection compared to the vaccine adjuvanted by incomplete Freund's adjuvant, demonstrating its added value during the preparation of tilapia vaccines.

Histopathological outcomes designating the toxicological aspects of Fumonisin B1 on Cultured Nile Tilapia, Oreochromis niloticus

Fumonisins are mycotoxins produced by Fusarium verticillioides and F. proliferatum. They occur wo... more Fumonisins are mycotoxins produced by Fusarium verticillioides and F. proliferatum. They occur worldwide and are found predominantly in maize and in maize-based animal feeds. A total of 100 Nile tilapia, Oreochromis niloticus were grouped and fed ration contaminated with different levels of Fumonisin B1 and observed for one week to determine the median lethal dose (LD50). Another experiment was operated to determine the clinicopathological picture of O. niloticus when experimentally fed with ration containing 1/10 and ½ of the LD50 of Fumonisin B1 and then observed for another six weeks. The clinical signs postmortem (PM) lesions and mortalities were recorded daily. Kidney, gills, brain, liver and spleen were sampled and examined for any histopathological alterations. It was found that the LD50 of Fumonisin B1 was 0.6 ppm. Additionally, there were no any pathognomonic clinical signs. The results of histopathological sections were in the form of hepatotoxic and nephrotoxic alterations, hyperplasia of gill filaments, ischemic neuronal injury and demyelination of the brain and depletion of the lymphoid elements and melanomacrophage centers of spleen.

Photobacteriosis in some wild and cultured freshwater fishes in Egypt

Two hundred and seventy wild and cultured fishes (Mugil cephalus, Mugil capito and Nile tilapia) ... more Two hundred and seventy wild and cultured fishes (Mugil cephalus, Mugil capito and Nile tilapia) were collected from Alexandria, and investigated for the isolation of Photobacterium. The recovered bacteria were studied for the virulence, pathogenicity and antimicrobial sensitivity. The pathogenesis of the most virulent isolate was done experimentally through pathological investigations. Monitoring of the water quality was also carried out. Three isolates (ph1, ph2 and ph3) of Photobacterium damselae subsp. damselae were obtained from Mugil cephalus, Mugil capito and Nile tilapia, respectively. The first two recovered isolates caused 20% mortality among Oreochromis niloticus and Cyprinus carpio, while the third one (ph3) caused 40% and 30% mortalities among the two fish species, respectively. The infection by these bacteria was accompanied by increase in the unionized ammonia (0.08 mg/L), decrease in the dissolved oxygen (1.8 mg/L) and high pH value (9.8). The experimentally infected fishes showed skin darkening and hemorrhaging of the caudal fin and operculum. Internally, whitish pin-sized nodules were seen in the liver, spleen and kidneys. Histopathologically, the Photobacterium, during the acute phase of the disease, induced degenerative changes in the parenchymatous organs of the infected fishes with marked brain lesions and hematopoietic involvement, while in the chronic stage, granuloma formation and focal necrosis in the internal organs was evident. The three recorded isolates of the Photobacterium were sensitive to erythromycin, streptomycin and ampicillin, but totally resistant to chloramphenicol, oxytetracycline, oxalinic acid, kanamycin and ciprofloxacin. On the other hand, Ph1 isolate was also sensitive to furazolidone while Ph3 was sensitive to neomycin. We were able to isolate three isolates of Photobacterium damselae subspecies damselae, which varied in virulence and antimicrobial sensitivity, from the wild and cultured freshwater fishes at Alexandria, Egypt as a new recorded pathogen in this environment.

This study was to achieve the aim of this study, the following points were done : 1) Evaluation of the virulence of isolated Pseudomonas flourescence. 2) And effect of isolated strain of Pseudomonas fluorescence on different blood parameters and serum enzyms. Examination the potency of locally prepared bacterin against isolated strain of Pseudomonas flourescence. the clinical signs were manifested as, loss of scales from some areas of the body with excessive mucus allover the body surface and petechial haemorrhages over the body. The lymphocyte showed a tendancy to decreasing from the 1 st week to the 4 th .While, the monocyte level decreased from the 1 st week to the 3 rd week. The Phagocytic activity and index increased progressively from the 1 st week to the 4 th week of the experiment. The results also showed that, the total protein, albumin, globulin and albumin/globulin ratio differ significantly among infected and control group and among different weeks. The antibody titer and RLP the antibody titer in the groups treated with P. Flourescence only of lower RLP than that of the control groups.

Introduction The increasing frequency of multidrug-resistant Pseudomonas aeruginosa (MDRPA) strai... more Introduction The increasing frequency of multidrug-resistant Pseudomonas aeruginosa (MDRPA) strains is concerning, as efficacious antimicrobial options are severely limited. Risk factors for MDRPA infection include prolonged hospitalization, exposure to antimicrobial therapy, and immunocompromised states such as human immunodeficiency virus infection. The emergence of MDRPA isolates during therapy was reported in 27%-72% of patients with initially susceptible P. aeruginosa isolates. Patients with severe MDRPA infections should be treated with combination therapy, consisting of an antipseudomonal β-lactam with an aminoglycoside or fluoroquinolone, rather than aminoglycoside and fluoroquinolone combinations, to provide adequate therapy and improve patient outcomes (1). P. aeruginosa produces a number of virulence factors, which, after colonization, can cause extensive tissue damage, bloodstream invasion, and dissemination (2). Pathogenesis is based on multiple virulence factors: endotoxin, exotoxins, and enzymes. Its endotoxin, like that of other gram-negative bacteria, causes the symptoms of sepsis and septic shock (3). Most strains of P. aeruginosa produce 2 exotoxins, exotoxin A and exoenzyme S; a variety of cytotoxic substances including phospholipases, pyocyanin, and proteases; and an alginate-like exopolysaccharide that is responsible for the mucoid phenotype. The importance of these putative virulence factors depends upon the site and nature of infection. Proteases play a key role in corneal ulceration, are important in burn infection, and are associated with chronic pulmonary colonization (4). Chitinase produced by P. aeruginosa has been investigated as a virulence factor associated with plant diseases (5). Folders et al. (6) reported that ChiC chitinase was produced by clinical isolates of P. aeruginosa. These virulence factors help the bacteria Background/aim: Virulent Pseudomonas aeruginosa is frequently life-threatening and often challenging to treat, and the emergence of multidrug-resistant isolates presents a critical problem for patients. The aim of the study was concerned with molecular analysis of the virulence factors and antimicrobial resistance profile of multidrug-resistant P. aeruginosa (MDRPA). Materials and methods: Out of 44 MDRPA isolates, 12 isolates representing different resistance profiles and sources of samples were selected for further molecular studies. Polymerase chain reaction (PCR) approaches were applied to identify the genes implicated in antimicrobial resistance or virulence factors in the selected MDRPA isolates. Results: Multidrug-resistance (pstS), β-lactamase (IMP7, IMP10, IMP13, and IMP25), and extended spectrum β-lactamase (blaOXA50) genes were detected in all of the selected MDRPA isolates. However, only 4 (33%) MDRPA isolates were positive for the presence of the extended spectrum β-lactamase (blaOXA2) gene. Furthermore, the hemolytic phospholipase C precursor gene (plcH) was detected in all PCR products of the tested MDRPA isolates while the exotoxin A (toxA) gene was absent. Other virulence genes were detected with variable percentage in tested isolates. Conclusion: The statistical analysis revealed a significantly positive correlation (r = 0.779, P = 0.002) between virulence factors and antimicrobial resistance marker profiles of the tested MDRPA isolates.

In this study, Pseudomonas aerogenosa was isolated during an outbreaks among cultured Oreochromis niloticus. The isolated strain from examined fish were indol-ve , nitrate +ve , glucose +ve, lactose-ve and sensitive to Amikacin and Gentamicin .The most clinical signs in both natural and experimental infected fish were darkening of body, loss of scales, tail rot and congestion of all internal organs.

The present work investigated experimentally a rapid, effective and inexpensive control to strept... more The present work investigated experimentally a rapid, effective and inexpensive control to streptococcosis, and the role of Streptococcus iniae as a major cause for mass mortalities of cultured Oreochromis niloticus in Kafrelsheikh, Egypt, in the summer of 2017; mass mortality was associated with poor water quality, and cultured naturally infected O. niloticus showed corneal opacity, congestion, and deep haemorrhagic ulceration in the dorsal region. S. iniae was identified as a causative agent by traditional and PCR techniques. The highest incidence of S. iniae was in the liver. Choosing vaccination as a control was experimentally investigated; the superiority of Streptococcus agalactiae FKC bacterin was related to the antibody titre, which was higher than S. iniae bacterin. The relative level of protection values were 79% and 23% in fish challenged with S. agalactiae and vaccinated with FKC of either S. agalactiae or S. iniae, respectively. These values were 56% and 74% in fish challenged with S. iniae and vaccinated with FKC of either S. agalactiae or S.i niae, respectively. In conclusions, autogenous bacterins of S. agalactiae or S.iniae induced cross-protection against S. agalactiae and S. iniae, in O. niloticus. The protection was superior to a S. agalactiae FKC vaccine, which could be a useful means for the prevention and control of streptococcosis.

The aim of the study was concerned with molecular analysis of the virulence factors and antimicrobial resistance profile of multidrug-resistant P. aeruginosa (MDRPA). Materials and methods: Out of 44 MDRPA isolates, 12 isolates representing different resistance profiles and sources of samples were selected for further molecular studies. Polymerase chain reaction (PCR) approaches were applied to identify the genes implicated in antimicrobial resistance or virulence factors in the selected MDRPA isolates. Results: Multidrug-resistance (pstS), β-lactamase (IMP7, IMP10, IMP13, and IMP25), and extended spectrum β-lactamase (blaOXA50) genes were detected in all of the selected MDRPA isolates. However, only 4 (33%) MDRPA isolates were positive for the presence of the extended spectrum β-lactamase (blaOXA2) gene. Furthermore, the hemolytic phospholipase C precursor gene (plcH) was detected in all PCR products of the tested MDRPA isolates while the exotoxin A (toxA) gene was absent. Other virulence genes were detected with variable percentage in tested isolates. Conclusion: The statistical analysis revealed a significantly positive correlation (r = 0.779, P = 0.002) between virulence factors and antimicrobial resistance marker profiles of the tested MDRPA isolates.

In this study, Pseudomonas aerogenosa was isolated during an outbreaks among cultured Oreochromis niloticus. The isolated strain from examined fish were indol-ve , nitrate +ve , glucose +ve, lactose-ve and sensitive to Amikacin and Gentamicin .The most clinical signs in both natural and experimental infected fish were darkening of body, loss of scales, tail rot and congestion of all internal organs.

The present work investigated experimentally a rapid, effective and inexpensive control to strept... more The present work investigated experimentally a rapid, effective and inexpensive control to streptococcosis, and the role of Streptococcus iniae as a major cause for mass mortalities of cultured Oreochromis niloticus in Kafrelsheikh, Egypt, in the summer of 2017; mass mortality was associated with poor water quality, and cultured naturally infected O. niloticus showed corneal opacity, congestion, and deep haemorrhagic ulceration in the dorsal region. S. iniae was identified as a causative agent by traditional and PCR techniques. The highest incidence of S. iniae was in the liver. Choosing vaccination as a control was experimentally investigated; the superiority of Streptococcus agalactiae FKC bacterin was related to the antibody titre, which was higher than S. iniae bacterin. The relative level of protection values were 79% and 23% in fish challenged with S. agalactiae and vaccinated with FKC of either S. agalactiae or S. iniae, respectively. These values were 56% and 74% in fish challenged with S. iniae and vaccinated with FKC of either S. agalactiae or S.i niae, respectively. In conclusions, autogenous bacterins of S. agalactiae or S.iniae induced cross-protection against S. agalactiae and S. iniae, in O. niloticus. The protection was superior to a S. agalactiae FKC vaccine, which could be a useful means for the prevention and control of streptococcosis.
