4-Methylpyrazole, in a dose producing inhibition of alcohol dehydrogenase (alcohol:NAD + oxidored... more 4-Methylpyrazole, in a dose producing inhibition of alcohol dehydrogenase (alcohol:NAD + oxidoreductase, EC 1.1.1.1), was given alone or together with ethanol (10%) as sole drinking fluid to growing rats for up to 38 weeks. Their weight curves remained normal. Electron microscopy of liver, kidney, and heart revealed no changes related to treatment. Hematologic analysis showed normal values for blood and bone marrow. Several clinical chemical parameters showed no impairment of liver or kidney function, except for an enhancement of the microsomal drug-metabolizing activity after concurrent administration of 4-methylpyrazole and ethanol. A study on rats receiving 4-methylpyrazole and ethanol indicated a mutual interaction of the two compounds or the metabolites, leading to increased concentration in the blood of the compounds and reduced formation of 4-hydroxymethylpyrazole, the primary metabolite of 4-methylpyrazole. In 2003, a joint effort between the Swedish government, a number of companies in the building and... more In 2003, a joint effort between the Swedish government, a number of companies in the building and construction sectors, some municipalities, insurance companies and banks set a target that by 2009, all new buildings and 30% of existing Swedish buildings should be rated using a voluntary environmental rating tool. In a major research programme finished in 2008, a tool was developed to be used in this context. The tool covers three assessment areas: Energy, Indoor environment and Material & Chemicals. These areas are split into 11 aspects with one or a few indicators. Rating criteria are specified for each indicator, stipulating requirements for a rating Gold, Silver, Bronze and Rated. Indicator results can then be aggregated to aspect, area and a single rating for building level for enhanced result communication. The tool builds on previous experiences regarding environmental building rating tools and therefore includes some special characteristics which aim to tackle some of the criticism directed towards the first generation of such tools. At the time of writing, the first buildings have received official ratings and an independent stakeholder group is promoting broader implementation of the tool.

Macaccr,~asciculalis, were chosen as a model to investigate the metabolism of 4.methylpyrazole and its interaction with methanol. Following a single dose of 4-methylpyrazole, the elimination of 4-methylpyrazole from the plasma followed a linear course; at a dose of ,50 mgikg (424 /LmoUkg) the constant rate of elimination of 4-methglpyrazole was approximately 13 pmollkg/h with disappearance of the dose within 35 h. This rate of elimination \vas slower when the smaller dose of 20 mg 4-methylpyrazole was administered. The primary metabolite of 4-methylpyrazole. 4-hydroxymethylpyrazole, accumulated to levels at most 10% af those of 4-methylpyrazole and did not persist in the plasma after the elimination of I-methylpyrazole. The concurrent administration of methanol with 4-methylpyrazole inhibited the disappearance of 4-methylpyrazole from the plasma about 25%, which suggested an interaction between the metabolism of the two compounds. As expected from its potent inhibitory effect on alcohol dehydrogenase. 4-methylpyrazole produced a profound and dose-dependent inhibition of the rate of elimination of methanol from the plasma. In addition, 4-methylpyrazole at a plasma level of greater than 10 PM was capable of preventing the accumulation of formic acid in methanol poisoned monkeys. In such a way 4-methglpyrazole repeatedly injected in various dose combinations was able to reverse methanol toxicity in the monkey. These studies indicated the usefulness of the monkey as a model to study 4-methylpyrazole activity and toxicity in light of its possible use to treat methanol poisoning in man.

No effects were observed on the weight curves. Hematologic analyses sho\ved normal values for blood and bone marrow. Studies of liver function \+?th transaminase, bilirubin ant1 albumin did not reveal any functional changes. Kidney function was normal as judged by creatinine and normal electrolytes. Electronmicroscopy of liver, kidney, and heart did not rexreal any changes related to treatment. Combined treatment of ethanol and +methylpyrazolr caused an increase of the microsomal drugmetabolizing activity. Chronic administration of ethanol and 4.methylpyrazoie indicated that there is a mutual interwtion in the metabolism of ethanol and 4-methylpyrazolr, leatlinp to a higher concentration of both ethanol and 4-methylpyrazole in the blood. Acute experiments, where alcohol dehydrogenase is saturated with ethanol, indicated a much hlo\\.er elimination of 4-methylpyrazole. Administration of ethanol and 4.methylpyrdzole in acute experiments showed a lower concentration of 4-hydroxymeth~lpyrazole in the blood indicating that ethanol interferes with the 4-methylpgrazole-anti/or 4-hgdrosymethyl-p~razole-metabolizing enzymes. The present investigation has shown that the acute and chronic toxicity of 4.meth~lp~razole alone or in combination with ethanol is minimal at doses that are effective in blocking ethanol metabolism in the rat. Because of its IOM toxicity and powerful inhibitory capacity, 4-methglpyrazole should be a potential tool for experimental clinical investigation of alcohol metabolism and its effects. 4-Methylpyrazole is also a potential therapeutic agent in methanol or ethylene glycol poisoning.
