This technique make us able grow cells outside of their natural environment (in vitro). In this technique, we can control and monitor the cell’s physicochemical environment such as pH and temperature. In this practice, MCF-7 (human breast cancer cell lines) which are continuous (non-primary culture) is used.
This experiment was performed to observe the effect of different hormone on the callus induction ... more This experiment was performed to observe the effect of different hormone on the callus induction using MS Media supplemented with 2, 4-D (2,4-Dichlorophenoxi acetic acid) and BAP (6-benzylaminopurine) 100mg/L respectively. The purposed of this were to practice proper sterilization technique as well as preparing the media for callus induction in callus culture and cell suspension. Green bean seeds used as source of explants and to prevent contamination, 20% sodium hypochlorite and 70% ethanol were used. Other aseptic measure has been applied. The callus formation was monitored every week and in the end of the sixth week, results obtained. Media that contained BAP shows the best callus formation and least contamination while MSO media does not produce callus at all, but it formed roots. 2, 4-D media shows moderate callus formation and contaminated by bacteria. Overall, BAP media is the best for callus induction and cell suspension. In the future, to avoid contamination and produce better callus each steps have to be done more carefully. Besides, longer duration for this experiment would give better results on the observation of callus development.
