This present study was carried out to evaluate antioxidant activities, total phenolic and total f... more This present study was carried out to evaluate antioxidant activities, total phenolic and total flavonoid contents of ethyl acetate extract of Mengkudu fruit and its fractions. Ethyl acetate extract was fractionated by column chromatography and yielded 15 fractions based on the identical TLC (thin layer chromatography) profile. Antioxidant activities in ethyl acetate extract and its fractions were determined by radical scavenging assay using DPPH (2,2-diphenyl-1- picrylhydrazyl) radical. The total phenolic and total flavonoid contents were determined spectrophotometrically. Among 15 fractions of ethyl acetate extract evaluated, fraction 8 (IC50 5,49 μg/mL) and fraction 7 (IC50 7,90 μg/mL) revealed antioxidant activities that higher than that of vitamin E (IC50 8,27 μg/mL). This attracted unethical players to blend these oils with lower price oils, therefore, its authentication by analysis of purity levels of oils is very important. This study highlighted the potential application of FTIR spectroscopy and multivariate calibrations for analysis of PSO, RBO, and SEO in ternary mixtures. Individual FTIR spectra of studied oils as well as in ternary mixtures with certain compositions were scanned and pre-processed. Therefore, UV-VIS spectrophotometry coupled with multivariate calibration of partial least square (PLS) for quantitative analysis of metamizole, thiamin and pyridoxin is developed in the presence of cyanocobalamine without any separation step. Methods: The calibration and validation samples are prepared. The calibration model is prepared by developing a series of sample mixture consisting these drugs in certain proportion. Cross validation of calibration sample using leave one out technique is used to identify the smaller set of components that provide the greatest predictive ability. The evaluation of calibration model was based on the coefficient of determination (R 2) and root mean square error of calibration (RMSEC). Results: The results showed that the coefficient of determination (R 2) for the relationship between actual values and predicted values for all studied drugs was higher than 0.99 indicating good accuracy. The RMSEC values obtained were relatively low, indicating good precision. The accuracy and presision results of developed method showed no significant difference compared to those obtained by official method of HPLC. Conclusion: The developed method (UV-VIS spectrophotometry in combination with PLS) was succesfully used for analysis of metamizole, thiamin and pyridoxin in tablet dosage form.

Physico-Chemical Characterization of Avocado (Persea americana Mill.) Oil from Three Indonesian Avocado Cultivars

A study was carried out to determine the physico-chemical characteristics of avocado oil derived ... more A study was carried out to determine the physico-chemical characteristics of avocado oil derived from three Indonesian avocado cultivars, namely Bantul (MAB), Purwokerto (MAP) and Garut (MAG). The extraction of avocado oil from avocado fruit was carried out using solvent extraction method. The avocado oil obtained from all samples had a green yellowish color. The iodine value of MAG is 88.7 g I 2 /100 g oil, slightly higher than MAB (87.0 g I 2 /100 g oil) and MAP (77.09 g I 2 /100 g oil) indicated that MAG contains more unsaturated fatty acid. The saponification values of avocado oil were 193.1 mg KOH/g oil for MAB, 198.4 mg KOH/g oil for MAP and 153.17 mg KOH/g oil for MAG, respectively. The peroxide values of MAB, MAP and MAP were 166.1, 124.7 and 14.9 meq kgG 1 oil, respectively. The Conjugated Dienes (CDs) and Conjugated Trienes (CTs) value of MAB, MAP and MAG were significantly different in the specific absorptivity range value from 2.6-3.7. The MAG had lowest CDs and CTs value. The anisidine value for avocado oil samples ranged from 10.59-11.36. There were no significant differences in the anisidine value among avocado oil samples. Avocado oil samples had high amounts of total unsaturated fatty acids, i.e., MAB (55.7%), MAP (62.8%) and MAG (68.9%), respectively. Thermal analysis by Differential Scanning Calorimetry (DSC) showed that avocado oil from three different cultivar had different melting and crystallization profile. Principal component analysis was used to classify each sample based on their DSC parameters. The results showed that by using the melting and crystallization proWles the discrimination of three avocado oils was very clear.

In the study, we examined the effects of marmin on the contraction of guinea pig-isolated trachea stimulated by several inducers, namely histamine, metacholine, compound 48/80. We also evaluated its action against contraction induced by extracellular or intracellular calcium ion. The possibility of marmin to potentiate the relaxation effect of isoprenaline was also studied. Marmin added in the organ bath at 10 min prior to the agonist inhibited the contraction elicited by histamine and metacholine in a concentration-dependent manner. Moreover, marmin antagonized the histamine-induced contraction in competitive manner. Marmin mildly potentiated the relaxation effect of isoprenaline.
