Adult Swiss albino mice were inoculated intraperitoneally (i.p) with lo6 Ehrlich ascites carcinom... more Adult Swiss albino mice were inoculated intraperitoneally (i.p) with lo6 Ehrlich ascites carcinoma cells. Twenty-four hours later they were given an i.p. injection of lo-60 mg/kg of withaferin A (WA), isolated from the roots of Withania somnifera. The tumor growth and tumor free animal survival were studied for up to 120 days. In another experiment 30 mg/kg WA was injected i.p. to mice at 1, 3 or 5 days after tumor cell injection with or without acute abdominal exposure to 7.5 Gy gamma radiation and the tumor growth and 120-day survival were studied. WA inhibited tumor growth and increased tumor free survival in a dose-dependent manner. The drug EDSo for 120-day survival was-30 mg/kg body wt. Drug treatment before irradiation synergistically increased 120-day survival even in advanced tumors. A dose of 30 mg/kg seems to be optimum for combination with radiation.

The aqueous extract of polyherbal formulation (PHF) triglize was used for the study. The animals were fed with high fat diet to induce hyperlipidemia . The test (polyherbal formulation) and standard drugs were administered once dally as a single oral dose for 28 days. The test drug administered at the dose levels of 25, 50, 100 and 200 mg/kg and standard drug atorvastatin was administered at the dose level of 10 mg/kg. Weekly body weight variation, lipid profile (total cholesterol, triglyceride, HDL, VLDL and LDL levels, HDL ratio and atherogenic index) were analyzed and day of the termination the liver was isolated from the all the animals and subjected to histopathological evaluation. End of the experiments liver markers like ASAT, ALAT were analyzed in serum samples. High fat diet showed significant increase in body weight, lipid profile in serum. The polyherbal formulation and atorvastatin treated groups showed significantly increased HDL levels and decreased total cholesterol, triglyceride, LDL, VLDL, HDL ratio and atherogenic index. End of the study atorvastatin elevated the ASAT and ALAT levels. The PHF has been found significant anti‐hyperlipidemic and hepatoprotective action.

The effect of plumbagin, a naphthoquinone from the roots of the Indian medicinal plant Plumbago r... more The effect of plumbagin, a naphthoquinone from the roots of the Indian medicinal plant Plumbago rosea, and Cobalt-60 gamma radiation was studied on Ehrlich ascites carcinoma in vivo, taking cytogenetic damage and cell cycle changes as experimental endpoints. Plumbagin (5 mg/kg body wt, P1) administered intraperitoneally produced a significant increase in the percentage of S-phase as well as G2-M cells with a corresponding decrease in the G1 phase at different post-treatment times. Radiation (7.5 Gy, RT) alone produced the classical G2 block at 1 hr, which persisted with a continuous increase throughout the post-treatment observation period. The combination treatment produced a similar effect as that of RT on G2-M cells, but its effect on the G1 phase was more pronounced than the latter. While P1 treatment produced a small increase in the percentage of labeled S-phase cells, combination treatment significantly reduced the labeled S-phase cells with a corresponding increase in the unl...

Materials and Methods: The hepatoprotective potential of Clearliv was evaluated in thioacetamide-induced liver necrosis, DL-galactosamine [GalN]-induced liver injury, and carbon tetrachloride [CCl 4 ]-induced hepatitis models in Wistar rats. In all the models, Clearliv (at the dose levels of 800, and 1000 mg/kg) was administered for 3 days orally followed by single intraperitoneal administration of the hepatotoxicant on the last day after one hour of Clearliv administration. After 24 h of toxicant administration blood sample was collected by sino-orbital puncture in sodium EDTA tubes. The efficacy of Clearliv was evaluated by plasma biochemical parameters (AST, ALT, and ALP), and antioxidant enzyme (lipid peroxidation, catalase, superoxide dismutase, glutathione peroxidase, and hydroxyproline) levels. Results: In thioacetamide-induced necrosis, and GalN-induced liver injury models, Clearliv at 1000 mg/kg showed significant reduction in the elevated plasma liver markers, and elevated antioxidants levels. In CCl 4 -induced hepatitis, the Clearliv had favorable hepatoprotective effect, but the results were not significant. Conclusion: Clearliv 800, and 1000 mg/kg showed significant hepatoprotective effect against thioacetamide- and GalN-induced liver necrosis and injury, respectively.

To determine the in vitro free radical scavenging property and in vivo diuretic effect of Trigliz... more To determine the in vitro free radical scavenging property and in vivo diuretic effect of Triglize TM , a marketed polyherbal formulation in experimental models. Materials and Methods: The aqueous extract of polyherbal formulation (PHF) triglize was used for the experiment. The free radical scavenging property and antioxidant effect of PHF were studied by LPS-induced free radicals in rat macrophages cells and DPPH (2, 2-Diphenyl-1-Picrylhydrazyl) methods, respectively. The diuretic effect of a PHF was studied with Lipschitz model using male Wistar rats. Results: PHF signifi cantly inhibited lipopolysaccharide-induced free radicals in rat macrophages and it showed moderate antioxidant potential in DPPH model. Polyherbal formulation at 50, 200 and 400 mg/ kg signifi cantly increased potassium excretion in urine at 0-5 h and 5-24 h. The diuretic effect of PHF was as similar as furosemide. Conclusion: The PHF has signifi cant diuretic effect and free radical scavenging properties.
