Mycoplasmas detection in milk samples ¿are the techniques reliable?

Food microbiology is an important aspect that must be considered during food manufacturing, which... more Food microbiology is an important aspect that must be considered during food manufacturing, which is, considering the possible sources of contamination, including operators. Highlighting that the group of mycoplasmas are little studied and therefore their presence in foods is not considered. Different mollicutes, particularly mycoplasmas, have been associated with economic losses in the poultry and dairy industries. The objective of this work was to determine the presence of the mycoplasma genus in milk samples through specific microbiological culture and to complement the diagnosis with the PCR technique. Two hundred milk samples were analyzed in Puebla city-Mexico. The samples obtained were centrifuge and reseeded in triplicate on SP4 agar, incubating and analyzed by stereoscopic microscopy. From each milk sample, DNA extraction was carried out with Zymo Research Quick-DNA™ Mini prep kit and primers AR1 and AR2 were used. The microbiological study showed positivity for mycoplasmas of 17% and 39%, in the liquid and solid phase, respectively. With the PCR technique, 53% positivity for mycoplasmas was obtained, which allows us to consider the presence of factors associated with samples of animal origin and that may be interfering with the sensitivity of the technique. Analyzing the viability of techniques to detect microorganisms, particularly mycoplasma, in foods is a practice that is recommended for the well-being of consumers.

This study compares the kinetics of adherence of both the prototype strain PG-18 and the recently... more This study compares the kinetics of adherence of both the prototype strain PG-18 and the recently clinically isolated strain P-140 of Mycoplasma fermentans to HEp-2 cells. M. fermentans may induce respiratory and genitourinary tract infections. We investigated the presence of putative adhesins from strain P-140. A surface protein of 47 kDa, labeled SP47, isolated from M. fermentans P-140, seems to be involved in acterial adherence on HEp-2 cells; this study could allow helping understand the interaction between mycoplasmas and their hosts. Anti-SP47 antibodies inhibited the formation of bacterial clusters and adherence to cultured cells, quantified by ELISA. M. fermentans P-140 was more efficient at adhering to cultured HEp-2 cells than PG-18.

Non invasive are serology and urea breath test. 152 Hp from 19 children's with acute gastritis (46. 1%); 9 Hp from an adult. There had ampicillin resistance, 27. 4%, claritromicin 21. 8%, metronidazol 58. 4% and tetracycline 31. 5%, the 21 % susceptible to four antimicrobial. RAPD-PCR with 4 primers gave 44 profiles, related with clinical profile. In 7 children, there were two profiles RAPD. Three were similar but different each other. AFLP 23 adults 151 Hp; cagA y vacA genotypes gave unique patterns for each patient. The genes cagA, babA and oipA, secuencing and compared with reported Hp (Genbank); gave high polymorfism. Everything indicates that there are colonization with multiple Hp strains in Mexicans. In 645 sera from 352 children: 36.9% with Hp, the 46. 9% gave him/her anti-shits and 16. 2% antibodies to urease. Adults 293 (89. 1%) with Hp, 78. 9% gave anti-CagA and 59% antibodies to urease. The expression of IL-8 was bigger in infected children that in not infected and more significant in peptic ulcer. Genomic library Cag-PAI revealed 90% (Hp) positive, they had the complete PAI, 2 were incomplete and three negatives. PCR-LiPA LiPA (LineProbe Assay) is suggestive for genotyping assays.

Biofilm formation, which has been associated with antimicrobial resistance, can also occur. Objectives: To determine methicillin resistance and to quantify the biofilm production to establish if there is a relationship in clinical isolates of S. aureus and coagulase-negative Staphylococci. Material and methods: A total of 11 strains of S. aureus and 12 of coagulase-negative Staphylococci were studied. Methicillin resistance was determined with cefoxitin discs and the Clinical Laboratory Standards Institute (CSLI), 2018 reference values. Biofilm production was quantified by the crystal violet method. The mecA and icaADBC genes were identified by PCR. A bivariate analysis was performed with chi-square (c 2) and Cramér's V statistical tests, using SPSS™, version 20.0 software.
