The ribonucleotide reductase (RR) 2 gene of the HSV-2 strain G was cloned, sequenced, and express... more The ribonucleotide reductase (RR) 2 gene of the HSV-2 strain G was cloned, sequenced, and expressed in an E. coli cell. The RR2 gene was located on the PstI 2.4 kb fragment, which was cloned and sequenced. The ORF of the gene was 1,011 bp and its termination codon was TAG; also, the CATATAA sequence was present in the promoter of the RR2 gene. A Poly A signal sequence (AATAAA) was found in the 3'-noncoding region. The RR2 proteins that were produced in the E. coli and Vero cells were confirmed using a Western blot analysis. SDS-PAGE revealed that the molecular weights of the fusion-RR2 that was produced in the E. coli cells were approximately 24 kDa and 38 kDa in the Vero cells. The RR2 proteins were soluble. The differences in the molecular weights might be due to modifications in the Vero cells.

The six lumbrokinase fractions (F1 to F6) with fibrinolytic activities were purified from earthwo... more The six lumbrokinase fractions (F1 to F6) with fibrinolytic activities were purified from earthworm Lumbricus rubellus lysates using the procedures of autolysis, ammonium sulfate fractionation, and column chromatography. The proteolytic activities on the casein substrate of the six iso-enzymes ranged from 11.3 to 167.5 unit/mg with the rank activity orders of F2 > F1 > F5 > F6 > F3 > F4. The fibrinolytic activities of the six fractions on the fibrin plates ranged from 20.8 to 207.2 unit/mg with rank orders of F6 > F2 > F5 > F3 > F1 > F4. The molecular weights of each iso-enzyme, as estimated by SDS-PAGE, were 24.6 (F1), 26.8 (F2), 28.2 (F3), 25.4 (F4), 33.1 (F5), and 33.0 kDa (F6), respectively. The plasminogen was activated into plasmin by the enzymes. The optimal temperature of the six iso-enzymes was 50 o C, and the optimal pH ranged from pH 4-12. The four iso-enzymes (F1-F4) were completely inhibited by PMSF. The two enzymes (F5 and F6) were completely inhibited by aprotinin, TLCK, TPCK, SBTI, LBTI, and leupeptin. The N-terminal amino acid (aa) sequences of the first 20 to 22 residues of each fraction had high homology. All six isoenzymes had identical aa residues 2-3 and 13-15. The Nterminal 21-22 aa sequences of the F2, F3, and F4 isoenzymes were almost the same. The N-terminal aa sequences of F5 and F6 were identical.

This study sought to investigate the ameliorating effects of soy 11S protein on the impacts of al... more This study sought to investigate the ameliorating effects of soy 11S protein on the impacts of alcohol consumption in rat hepatocytes and in reducing total cholesterol levels and total lipid levels in the serum. Liver histology and the clinically important enzyme markers (Aspartate Aminotransferase: AST and Alanine Aminotransferase: ALT) of rats, administered with both alcohol and soy 11S protein treatments, were compared with those in the control group. The treatment regimen (11S soy protein extract) significantly reduced serum ALT and AST levels, indicating the hepato-protective effects of soy 11S protein. Furthermore, total cholesterol and total lipid levels were significantly reduced. In addition to preventing the presence of lipid droplets and secondary lysosomes, electron microscopy indicated that the administration of the soy 11S protein treatment preserved important hepatocyte structures. These results indicate that soy 11S protein can positively mediate the effects of alcohol on hepatocytes and general liver functions.

The genus Pseudo-nitzschia is a chain-forming diatom comprising about 30 species some of which ar... more The genus Pseudo-nitzschia is a chain-forming diatom comprising about 30 species some of which are known to produce domoic acid (DA) that causes amnesic shellfish poisoning (ASP). The current study aimed at assessing the population dynamics of Pseudo-nitzschia in the near shore waters of Dar es Salaam. Samples were collected between August 2008 and July 2009 from two stations. The first station was located in the open waters off Mbudya Island and the second was located at a point in-between Kunduchi and Mbudya Island. Three Pseudo-nitzschia species, i.e., Pseudo-nitzschia pungens, Pseudo-nitzschia seriata and Pseudo-nitzschia cuspidata were encountered with Pseudo-nitzschia pungens being the most abundant. The concentration of Pseudo-nitzschia species ranged from none in various samples to a highest value of 16 cells/L recorded in September 2008. There was no significant difference in the abundance of Pseudonitzschia spp. between site 1 and 2 and between the northeast (NE) and southeast (SE) monsoon periods. The abundance of Pseudo-nitzschia spp. was below concentrations reported elsewhere to cause problems to shellfish consumers implying that shellfish collected near Dar es Salaam may be ASP free. However, further studies are required to ascertain DA production in coastal waters of Tanzania.

Purpose: According to the Body-EU education program, this study investigates changes after conduc... more Purpose: According to the Body-EU education program, this study investigates changes after conducting education with ten subjects with autism symptoms (disability level 2~3) 63 times (126 hours) for nearly two years. Methods: Body-EU habit practice, note writings discussion, singing, Bible study, question and communication, class arrangement, etc., a total of 63 times were educated for 2 hours each week. Results: Subjects felt comfortable breathing and improved energy. Other subject's habit of an out-toed gait step and bending back alleviated. One subject made it easier to speak, so if talking with no strength. When breathing, the stuffy nose improved, the eyes became calm, and the hands and feet were warm. Subject felt like my head was cleaned. Heart tightness disappeared. Subject felt at ease as gaining confidence. The subject gained confidence in seeing whoever the other person was. The subject's pronunciation became more sophisticated, with more confidence, and his mouth cramping alleviated. Subjects with right eye movements showed a relatively high concentration in education time. Conclusions: Subjects are educated and trained in making general habits of their body from morning to going to bed. This study was a research program for ordinary people , but it confirmed with autism. Whether it was genetic autism or acquired autism, they could be somewhat relieved.

Several interesting phenotypes were observed. A large proportion of the mutants were unable to form polyhedral occlusion bodies (polyhedra) at the nonpermissive temperature (32.5°C). At 32.5°C, one mutant formed plaques in which the cells lacked polyhedra. Another mutant type was defective in the production of progeny extracellular nonoccluded virus and produced a “plaque” consisting of only a single cell containing polyhedra at 32.5°C. One mutant was defective in plaque formation, progeny nonoccluded virus formation, and polyhedra formation at 32.5°C. Several mutants produced nonoccluded virus but failed to produce plaques or polyhedra at 32.5°C. Other phenotypes were also distinguished. Purpose: The purpose of this study was to investigate whether Qihyul-therapy was effective agains... more Purpose: The purpose of this study was to investigate whether Qihyul-therapy was effective against subjects of bad Qihyul circulation who had narrowed muscles of the buttocks, a distorted body shape, or pain. Methods: Qihyul-therapy used. Results: The muscles of the buttocks remained almost untouched. The elasticity of the muscles restored. The skin tone brightened, and the anal muscle strengthened after Qihyul-therapy. The subject's body was shifted to the right by −7°. The right arm was about 6 cm longer than the left arm. But after correction, the right arm was about 5 cm shorter. The distorted body was almost balanced. After neck and shoulder correction, the distance between the collarbone and neck was 7~10 cm wider. After the pelvic correction, the pain in the buttocks was relieved, and the position of the hip joint returned correctly. The right leg that was turned 45° to the left was corrected, and the length became the same. Subjects had no knees attached, their right knees were up 5 cm above the left, and their ankles bent at an angle of 30 ° relative to the left, but after correction, the knees of the two legs stably attached. The height of the knee became the normal angle. Conclusion: If the Qihyul-therapy, the spine correction Qihyul-exercise and the pelvic correction Qihyul-exercise performed steadily, the deformed body shape restored adequately, and it is useful to prevent and cure related diseases by alleviating the pains.

목적: 본 연구는 임상대상자 30명에게 미생물발효용액인 Zen발효용액을 8주간 섭취시킨 후에 임상대상자들의 변에서 중요한 장내 유익세균 및 저해세균의 증식이 촉진되는지 또는 ... more 목적: 본 연구는 임상대상자 30명에게 미생물발효용액인 Zen발효용액을 8주간 섭취시킨 후에 임상대상자들의 변에서 중요한 장내 유익세균 및 저해세균의 증식이 촉진되는지 또는 감소하는지를 연구하는 것이 목적이었다. 방법: 장내세균은 특정 Primer를 이용하여 PCR 증폭기로 동정 검색하였다. 결과: Zen발효액을 섭취한 임상군의 Bifidobacterium genus 유전자복제지수(gi%)는 섭취 전 수치는 55.15%, 섭취 후에는 70.1%로 나타났으며, 섭취 후에 14.95% 차이로 유의성이 있게 증가하였다. 아래 모든 대조군은 유의성이 없었다. 임상군의 Lactobacillus genus지수는 섭취 전이 46.87%, 섭취 후가 60.91%로 나타났으며, 섭취 후에 14.04% 차이로 유의성은 있게 증가하였다(p<.01). 임상군의 Clostridium genus지수는 사전이 85.64%, 사후가 65.99%로 나타났으며, 섭취 후에 -19.65% 차이로 유의성이 있게 감소를 하였다(p<.017). 임상군의 Bacteroides genus지수는 사전이 17.11%, 사후가 20.22%로 나타났으며, 섭취 후에 3.11% 차이로 유의성이 있게 증가하였다. 임상군의 Prevotella genus지수는 사전이 14.01%, 사후가 16.79%로 나타났으며, 섭취 후에 2.78%차이로 증가하였으나 유의성은 없었다. 결론: 장내세균은 혼합미생물의 발효액Zen을 섭취 후에 장내에서 유익균은 증식이 증가하고, 유해균은 억제되는 현상을 발견하였다. Zen발효액은 장 건강에 유익한 음료라 평가한다.

In this research, the total polyphenol and flavonoid contents in the hot water extract (HE) and m... more In this research, the total polyphenol and flavonoid contents in the hot water extract (HE) and methanol extract (ME) of the Coriolus versicolor fruiting body and their partial bioactivities were investigated from March, 2015 to February, 2016. For the analysis of the data, the software used was SPSS 2.0. The results indicate that the total polyphenol content in the HE (129.60 mg/dl) was 85,8 mg/dl higher than that in the ME (43.80 mg/dl). The total flavonoid content in the HE (30.50 mg/dl) was 9.5 mg/dl higher than that in the ME (20.90 mg/dl). The DPPH scavenging activities of the HE and ME at a concentration of 3 mg/ml were 41.03% and 16.36% higher than those at a concentration of 1 mg/ml. The tyrosinase inhibitory activities of the HE and ME at a concentration of 3 mg/ml were 8.29% and 10.32% higher than those at a concentration of 1 mg/ml. The elastase inhibitory activities of the HE and ME at a concentration of 3 mg/ml were 3.12% and 21.53% higher than those at a concentration of 1 mg/ml. The collagenase inhibitory activities of the HE and ME at a concentration of 3 mg/ml were 5.44% and 4.07% higher than those at a concentration of 1 mg/ml. In conclusion, these two kinds of extracts are judged to be useful as skin wrinkle relaxants, due to their anti-oxidative and bioactive functions.

The purpose of the study is to investigate the effectiveness of the concurrent thermotherapy and ... more The purpose of the study is to investigate the effectiveness of the concurrent thermotherapy and intermittent fasting therapy for obesity. The experimental therapy conducted at the same time for 11 days, and had 35 adult participants. Six variables(girth, body weight, body mass index, body fat ratio, abdominal fat, and body basal metabolic rate) of participants were measured initially before the experimental therapy, and respectively after two consecutive experimental therapies (S1 and S2). The differences between the averages of the six variables in the initial measurement (C group), S1 group, and S2 group are significant and correlate each other. The differences between variables also were significant within groups and between groups. Additionally, the results show that the effectiveness of the therapy was greater after the 2nd therapy than the 1st therapy. In conclusion, these findings suggest that the concurrent therapy is effective and useful as a natural healing therapy for obesity.
