Collagen arthritis is an experimental model for rheumatoid arthritis which is induced in suscepti... more Collagen arthritis is an experimental model for rheumatoid arthritis which is induced in susceptible DBA/1 mice after immunization with native type II collagen (CII). All immunized mice exhibit a humoral response to CII regardless of the occurrence of articular pathology. In order to further analyze the determinants recognized on the CII molecule, we have investigated the specificity of the antibodies produced by susceptible and resistant strains of mice in response to bovine CII. The sera of both arthritic and non arthritic mice contained distinct antibody populations. One population was exclusively directed to conformational determinants present on the native molecule, another population reacted with epitopes common to native and heat denatured CII but destroyed by cleavage with cyanogen bromide. Type-specificity of the antibodies was mainly restricted to CII although some cross-reactivity with type XI collagen was noted. Using a combination of affinity chromatography procedures anti-CII IgG were fractioned into antibodies reacting with determinants common to bovine and murine CII and those reacting with species specific structures. Sera from arthritic DBA/1 mice contained high levels of antibodies with species cross-reactivity in comparison to the sera from non arthritic DBA/1. Moreover, the mice that developed arthritis after injection of bovine CII were found to secrete also anti-CII antibodies exclusively directed to murine CII epitopes whereas non arthritic immunized mice did not. These findings suggest that the articular pathology is associated with an autoimmune reactivity to autologous CII in the experimental model of collagen induced arthritis.
