This paper seeks to interrogate whether the physically challenged in Nigeria have secured their legal and institutional right of access to education. It is observed that the physically challenged are continually being deprived reasonable access to education and this has contributed negatively to their social conditions in the long run. It is suggested that the Nigerian government should streamline a substantial legal and policy framework with the aim of guaranteeing access to education for all persons with disability. In order for these policies to succeed, the changes should fundamentally transform the educational approach by ensuring that persons with disability are not excluded from the general educational system on the basis of disability. AGS-16 is expressed homogeneously at high levels in primary kidney cancers and in kidney metastases to lymph nodes and lung tissues. Amongst normal tissues, AGS-16 is expressed predominantly in kidney proximal tubules. In vitro studies with a purified extracellular domain (ECD) protein and AGS-16 expressing cells have indicated an involvement of AGS-16 in angiogenesis and tumor cell proliferation and migration. AGS-16 ECD induced human umbilical vein endothelial cell (HUVEC) tube formation on Matrigel in vitro , as well as their migration in a transwell chamber assay. RNAi silencing of AGS-16 expression in the kidney clear cell line RXF-393 and the liver cancer cell line HepG2 significantly reduced their proliferation, survival, migration and invasion. XenoMouse...

SLC44A4 is a member of a family of putative choline transporters which we show to be markedly upregulated in a variety of epithelial tumors, most notably prostate and pancreatic cancer. SLC44A4 is normally expressed on the apical surface of secretory epithelial cells but in cancer we show expression is not restricted to the luminal surface in advanced and undifferentiated tumors. ASG-5ME consists of a human IgG2 anti-SLC44A4 antibody conjugated through a cleavable linker to the microtubule-disrupting agent, monomethylauristatin E. It has potent anti-tumor activity in both cell line and patient-derived xenograft models of pancreatic and prostate cancers. Combination studies with ASG-5ME and nab-paclitaxel demonstrated combination effect in both pancreatic and prostate tumor models. Altogether, the data presented here suggests that ASG-5ME may have the potential to...

Amongst normal tissues, AGS-8 expression is restricted to testis and the red pulp of spleen. In vitro studies with AGS-8 expressing cells indicated an involvement of AGS-8 in tumor cell survival, migration, invasion and cell-cell interaction. Furthermore, a purified AGS-8 extracellular domain (ECD) protein mediated Ca2+-dependent binding to human umbilical vein endothelial cells (HUVEC), supported their survival and induced tube formation in vitro. Recombinant expression of AGS-8 in murine fibroblasts was oncogenic, leading to aggressive tumor growth and metastasis in mice. Additionally, these cells induced significant angiogenesis when implanted in vivo. Recombinant AGS-8 expression in ovarian carcinoma cells (OVCAR-5) led to enhancement of their intrinsic tumor growth in vivo.
XenoMouse® technology was employed to generate a panel of high affinity fully human monoclonal antibodies (MAbs) to AGS-8, targeting diverse epitope groups. One MAb that exhibited potent inhibitory activity in all functional assays was AGS-8M4, an IgG1κ MAb with a Kd = 5 x 10-9 M for cell surface AGS-8. AGS-8M4 dose-dependently inhibited tube formation in vitro. In addition, AGS-8M4 inhibited the migration, invasion and cell-cell interaction of AGS-8 expressing cells. Significant anti-tumor efficacy of AGS-8M4 in vivo was observed in both recombinant models and in endogenous models, either during tumor formation or in well-established tumors. Efficacy of AGS-8M4 was maintained after MAb de-glycosylation, indicating an intrinsic anti-tumor activity of the MAb without significant contribution by immune effector functions. AGS-8M4 inhibited lung metastases in the 3T3/AGS-8 tumor model, further suggesting a role of AGS-8 in tumor cell migration and invasion. Studies evaluating the anti-tumor efficacy of AGS-8M4 with drug combinations are underway. Together, these data establish AGS-8 as a novel and attractive ovarian cancer target and AGS-8M4 as a potential therapeutic MAb for this cancer indication.

Molecular profiles of eight pancreatic ductal adenocarcinoma patient tumors were compared to corresponding passages of xenografts obtained by grafting tumor fragments into immunocompromised mice. Molecular characterization was performed by copy number analysis, gene expression and microRNA microarrays, mutation analysis, short tandem repeat (STR) profiling, and immunohistochemistry. Xenografts were found to be highly representative of their respective tumors, with a high degree of genetic stability observed by STR profiling and mutation analysis. Copy number variation (CNV) profiles of early and late xenograft passages were similar, with recurrent losses on chromosomes 1p,

Analysis of protein expression carried out in an extensive panel of tumor specimens by immunohistochemistry confirmed strong to moderate expression levels of Nectin-4 in bladder, breast and pancreatic cancers (40-60%). A subset of lung and ovarian cancers (up to 30%) also showed significant expression levels of this protein. We have generated an antibody-drug conjugate (ADC) targeting Nectin-4, AGS-22M6E. The highly potent microtubule disrupting drug monomethyl auristatin E (MMAE) was conjugated via valine-citrulline cleavable linker to a fully human IgG1k antibody specific for Nectin-4. AGS-22M6E was characterized for binding affinity, species crossreactivity and cytotoxicity in vitro. AGS-22M6E binds to cell surface Nectin-4 with high affinity and crossreacts with Nectin-4 orthologs of cynomolgus monkey, rat and mu...
