One method to prevent typhoid fever is vaccination. Nowadays, frequently, vaccine for typhoid fever is Ty 21a and vi polysaccharide vaccines. The aim of the study is to detection S. typhimurium and E. coli by Real Time PCR from milk samples. Gene targeted of S. typhimurium is pef and the E. coli is fimC. Confirmation test of all primers in contaminated milk with S. typhimurium and E. coli gave different cycle threshold (Ct) value in range 12-13. The detection limit for S. typhimurium with primer pef was in 7, 92 pg/μL and detection limit for E. coli with primer fimC in 7, 12 pg/μL. Each primer produces distinctive melting peak. The melting peak curve S. typhimurium with pef primer is 84,17°C and the melting peak curve E. coli with fimC primer is 79,97°C Based on the result of this study; all the primers were successfully applied in detecting S. typhimurium and E. coli bacteria in the milk sample quickly, sensitive and specific as well as can determine the number of bacteria accurately.

Kurzichalcolactone A (1) and B (2), two chalcone derivatives were isolated from two species of In... more Kurzichalcolactone A (1) and B (2), two chalcone derivatives were isolated from two species of Indonesian Cryptocarya: Cryptocarya phoebeopsis and Cryptocarya morotaiense. The structures of the isolated compounds were elucidated based on the analysis of spectroscopic data, including UV, IR, NMR, and mass spectra. On biological evaluation of these compounds for their cytotoxic activity, it was found that (1) exhibited moderate inhibition activities against murine leukemia P388 cells while (2) was inactive.

Amylase enzymes are widely used in various industrial fields such as textile, food, paper and other industries. Compared to other organisms, yeasts can produce enzymes more effectively and safer for the environment. Amylolytic yeast can be isolated from flower substrates as it contains sugar for the very limited condition of yeast growth. This study aims to isolate, select and characterize amylolytic yeasts on the substrate of Paphiopedilum sp. from Bedugul Botanical Garden, Bali. Yeast isolation was carried out with direct and washing method, followed by screening conducted on YPSA medium with diffusion agar method. Results showed that 19 yeast isolates were obtained with characteristics of 73.9% white-mucoid, 21.05% cream-mucoid and 5.26% light flesh-mucoid. Penelitian ini bertujuan untuk meningkatkan efektivitas pembelajaran Ksp dan koloid siswa SMA Yup... more Penelitian ini bertujuan untuk meningkatkan efektivitas pembelajaran Ksp dan koloid siswa SMA Yuppentek 1 Tangerang.melalui implementasi Lesson Study. Sampel yang digunakan adalah satu kelas XI IPA dari enam kelas XI IPA yang ada di sekolah tersebut. Penelitiian dilakukan sebanyak lima siklus, dan setiap siklus terdiri dari tiga tahapan, yaitu tahap perencanaan (plan), tahap pelaksanaan (do), dan tahap refleksi (see) untuk memberikan tanggapan dan gambaran tentang hasil pelaksanaan. Indikator ketercapaian yang digunakan untuk mengetahui efektivitas pembelajaran pada penelitian ini adalah aktivitas siswa, interaksi siswa, kepuasan siswa, keterlaksanaan pembelajaran oleh siswa, dan keterlaksanaan pembelajaran oleh guru. Tiap pertemuan siswa diberikan angket untuk mengetahui tanggapan siswa mengenai pembelajaran yang dilakukan. Hasil angket siswa dan hasil pengamatan observer dianalisis lebih lanjut sehingga diperoleh data yang akurat tentang ketercapaian indikator peneltian yang disyaratkan. Hasil penelitian menunjukkan adanya peningkatan ketercapaian indikator efektivitas pembelajaran dari siklus pertama ke siklus kedua, dan seterusnya. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa Lesson study dapat meningkatkan efektivitas pembelajaran Ksp dan koloid siswa di SMA Yuppentek 1 Tangerang. Penelitian ini bertujuan untuk meningkatkan kualitas pembelajaran kimia melalui Lesson Study pada... more Penelitian ini bertujuan untuk meningkatkan kualitas pembelajaran kimia melalui Lesson Study pada materi Hidrolisis Garam, Kelarutan dan Hasil Kali Kelarutan (Ksp) di SMA Negeri 7 Tangerang. Metode penelitian yang digunakan dalam penelitian ini adalah Metode deskriftif kualitatif dengan pendekatan Active Learning. Penelitian dilakukan melalui enam kali pertemuan dengan empat siklus, dan setiap siklus terdiri dari tiga tahapan, yaitu tahap perencanaan (plan), tahap pelaksanaan (do), dan tahap refleksi (see). Selanjutnya pada tiap akhir materi pokok dilakukan tes hasil belajar.Kualitas pembelajaran pada penelitian ini diukur melalui empat indikator, yaitu interaksi siswa, motivasi belajar siswa, keaktifan siswa, dan keterlaksanaan proses pembelajaran oleh guru. Hasil analisis menunjukkan bahwa keempat indikator kualitas pembelajaran mengalami perubahan ke arah positif dari empat open lesson yang dilaksanakan. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa Lesson studydengan pendekatan active learning dapat meningkatkan kualitas pembelajaran kimia pada Materi Hidrolisis Garam, Kelarutan dan Hasil Kali Kelarutan (Ksp) di SMA Negeri 7 Tangerang. It started from December 2011 until April 2012. The method was classroom action research in two rounds. This research started with preliminary analysis by teacher interview and student questionnaire to make the lesson plan. The results showed that the learning effectiveness had enhancement from first cycle to second cycle. It could be seen from the average of learning outcomes that increased from 73,89 in first round to 81,28 in second round. To increase learning effectiveness of buffer solution through SAVI approach, first we could know the dominant learning style of student in that class and the characteristic of subject that learnt. Matakuliah Biokimia I merupakan matakuliah wajib yang harus diikuti oleh mahasiswa Jurusan Kimia.... more Matakuliah Biokimia I merupakan matakuliah wajib yang harus diikuti oleh mahasiswa Jurusan Kimia. Mulai semester 093, matakuliah ini disampaikan secara bilingual. Tujuan penelitian ini adalah meningkatkan aktivitas mahasiswa pada pembelajaran Bilingual Biokimia I dengan model pembelajaran Cooperative Learning Tipe Student Team Achievement Division (STAD). Metode penelitian yang digunakan adalah metode Penelitian Tindakan Kelas. Penelitian dilakukan melalui tiga siklus dengan setiap siklus memiliki tahapan identifikasi dan analisis masalah, penetapan fokus permasalahan, perencanaan tindakan, pelaksanaan tindakan, observasi, evaluasi, refleksi dan rencana tindak lanjut. Hasil analisis data menunjukkan bahwa indikator interaksi yang terdiri atas 3 subindikator dari siklus I-III mengalami kenaikan. Subindikator Interaksi antar mahasiswa dari 83-86%. Subindikator Interaksi mahasiswa dengan dosen dari 86-87%, dan Subindikator Interaksi mahasiswa dengan sumber belajar dari 83-84%. Pada indikator kedua yaitu keaktifan mahasiswa yang terdiri atas 5 subindikator secara umum juga mengalami peningkatan. Subindikator kehadiran mahasiswa dalam proses pembelajaran dari 97-100%. Subindikator intensitas mahasiswa dalam penggunaan bahasa Inggris dari 72-76%. Subindikator kemampuan mahasiswa dalam bertanya dan menjawab dengan bahasa Inggris 52-68%. Subindikator kemampuan mahasiswa dalam mengungkapkan pendapat dengan bahasa Inggris 71-73% dan Subindikator kemampuan mahasiswa dalam memahami konsep Biokimia dalam bahasa Inggris sebesar 58-79%. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa proses pembelajaran biokimia I dengan model pembelajaran STAD yang disampaikan secara bilingual berhasil meningkatkan aktivitas mahasiswa dalam proses pembelajaran. Penelitian ini diharapkan menjadi landasan pengembangan proses pembelajaran bilingual biokimia lainnya terutama dalam peningkatan kualitas proses tanya-jawab dan penguasaan konsep biokimia dalam bahasa Inggris.

The mortality rate of typhoid disease in Indonesia is increasing. The most common detection method currently used is serological test. However, this method is often resulted in less accurate, less sensitive and specific detection. The previous research has successfully discovered fim-C-S. typhi gene which is able to code protein to contribute in adherent or colonization in human epithelial cell. The information of fim-C-S. typhi gene is used to develop more specific and more accurate detection method for S. typhi bacteria by amplification, characterization, specificity and sensitivity assay, and continue with clinical assay. The PCR result was the amplified 0.2 kilo base (kb) DNA fragment using the fimbriae-C-S. typhi primer pairs. The specificity assay was conducted by comparing the amplicon in size of 0.2 kb S. typhi with Gram negative bacteria genome with similar characteristics. The results indicated the fimbriae-C-S. typhi primers can differentiate amplicon in size of 0.2 kb S. typhi against Enterobacter aerogenes, Klebsiella pneumonia, Vibrio cholera, Escherichia coli, and Pseudomonas aeruginosa. However, they cannot differentiate S. typhimurium which have higher homology in their genome. Sensitivity assay was conducted by determining the detection level of fim-C-S. typhi primers to S. typhi genome. The sensitivity assay showed the detected fimbriae-C primers S. typhi with 1.295 x 10-39 µg/mL DNA chromosome. The clinical assay was evaluated by comparing the PCR product of suspected patient blood, the isolated bacteria S. typhi genome served as positive control, and uninfected human blood as negative control. The PCR product from 10 suspected patient bloods showed the band size 0.2 kb equals to the size of band produced by the PCR product from isolated S. typhi bacteria genome. Meanwhile, the band was not detected in the uninfected blood. Band size 0.2 kb from the suspected patient blood sample indicated positive infection of S. typhi bacteria. The above results demonstrated the developed clinical detection for typhoid disease using fimbriae-C-S. typhi primer pairs.

That is due to strengthen students' understanding of the environment. This study aimed to develop an electronic module for enrichment programs in the form of science, environment, technology, and society (SETS) called EM-SETS. The research method used a research and development model. The instrument used in this study was a questionnaire. The module developed was validated by 3 learning topics and language experts and 3 media experts. The module was tested by 94 Class X students at Vocational School and 8 chemistry subject teachers. The results of the EM-SETS assessment by experts, teachers, and students are classified as high to very high, so it can be concluded that the EM-SETS that was developed was feasible to be used as a learning medium for students at Vocational School.

The aim of this research was to develop Salmonella typhimurium detection by Real Time Polymerase Chain Reaction (RT-PCR) using two pairs of primers. The ability of primer pairs to detect S. typhimurium is seen from cycle threshold or Ct. Artificially contaminated milk sample with 24 ng each microliter can be detected with fljB (flagellin gene) primers on Ct 12,933 and with STM4497 (hypothetical protein code) primers on Ct 13,665. The specificity test of both primers showed that melting temperature (Tm) of fljB was 80,5 degree Celsius, and STM449 was 81,6 degree Celsius. FljB and STM4497 primers gave an average detection limit respectively of 11,75 Colony Forming Unit (CFU) each milliliter and 6,8 CFU each milliliter. It is an important plant for novel pharmaceuticals since most of the pathogens are developing resistance against currently available antimicrobial drugs. Thus, this review is summarizing the active compounds which are responsible for bioactivity of the plant and provides insight into genetic regulation of alkaloids containing in the extracts to serve as antimicrobial agents. Further research on Catharanthus roseus is highly recommended to explore its full potential as phytomedicine specifically to be developed as antimicrobial drug.
