The complete nucleotide sequence of insertion element IS492, which causes reversible inactivation... more The complete nucleotide sequence of insertion element IS492, which causes reversible inactivation of extracellular polysaccharide production in the marine bacterium Pseudomonas atlantica, is presented. Insertion of IS492 results in the EPS-phenotype, and excision results in restoration of EPS+. DNA sequencing of the site of insertion in the eps locus showed that insertion of IS492 generates a 5-base-pair repeat and that its excision is precise. IS492 is 1,202 nucleotides in length and contains one large open reading frame encoding a protein of 318 amino acids, a candidate for transposition function. No similarity between IS492 and other transposable elements has been found. Unlike the situation with other insertion sequences, no direct or inverted repeats exist at the termini of IS492.

ABSTRACT The Attentional Blink (AB) is a visual phenomenon demonstrating an apparent limit of the... more ABSTRACT The Attentional Blink (AB) is a visual phenomenon demonstrating an apparent limit of the visual system's ability to process individual items in a rapid serial visual presentation (RSVP). The AB occurs when two items, the first (target) to be identified and the second (probe) to be detected, are presented among other stimuli in a RSVP. The AB describes the interval of time in which attention cannot be focused on the probe which thereby proceeds undetected. Previous research has demonstrated that an iconic happy face attenuates the AB (Mack et al, 2002). We chose to explore whether this finding was face specific or emotionally mediated. Methods: RSVP streams of face stimuli with similar emotional expressions were presented at a rate of 100/ms each to produce an AB. Ss were instructed to identify one of five blue shapes superimposed over one of the faces in the RSVP and detect a subsequent face probe presenting a different emotional expression. Conditions consisted of: 1) a smiling face probe among neutral expressions; 2) a frowning face probe among neutral expressions and; 3) a neutral expression face probe among smiling faces. Results: When Ss were asked solely to detect the probe, performance ranged between 90 & 100%. However when Ss were asked to both identify the target and detect the probe, the results demonstrated a monotonic AB with a lag effect (Awh et al, 2003) rather than the classic U-shaped AB function (Raymond et al, 1992) across all conditions. Notably, significant differences were found between detection rates of the happy face and sad face as compared to the neutral face (p Conclusions: The AB has been demonstrated reliably over numerous conditions using various stimuli. Our experiment differs from these studies in that both the distractors and probes comprised similar forms with different meaning. Our results point to the attenuation of the AB by emotion and offer additional support in the late selection processes mediating perception of the attentional blink.

Genetic and biochemical analysis of mutants defective in the synthesis of flagella in Escherichia... more Genetic and biochemical analysis of mutants defective in the synthesis of flagella in Escherichia coli revealed an unusual class of mutants. These mutants were found to produce short, curly, flagella-like filaments with low amplitude (.0.06 gim). The filaments were connected to characteristic flagellar basal caps and extended for 1 to 2 pm from the bacterial surface. The mutations in these strains were all members of one complementation group, group E, which is located between his and uvrC. The structural, serological, and chemical properties of the filament derived from the mutants closely resemble those of the flagellar hook structure. On the basis of these properties, it is suggested that these filaments are "polyhooks", i.e., repeated end-to-end polymers of the hook portion of the flagellum. Polyhooks are presumed to be the result of a defective cistron which normally functions to control the length of the hook region of the flagellum. The MgSO4-7H2O glucose and thiamine were added aseptically after autoclaving. Amino acids and thymine, if required, were added to a final concentration of 100 mg/liter. Minimal plates were prepared by adding 1.5% agar to minimal medium.

A 54-year-old man from Guyana presented with a painful, nonerythematous, subcutaneous nodule on h... more A 54-year-old man from Guyana presented with a painful, nonerythematous, subcutaneous nodule on his right patella at the site of a remote injury. The patient fell while climbing a coconut tree at 10 years of age, 44 years before presentation. The fall caused him to land on the stump of a 'black sage tree' injuring his right patella. Multiple small splinters from the stump penetrated his skin over and around his patella, which were removed shortly after the initial injury. The patient reported a significant amount of purulent discharge that eventually subsided. He was never treated with antibiotics. For approximately the next 40 years, the patient reported a history of knee swelling that dramatically increased in recent years. He reported no recent trauma. He experienced no fever, chills, sweats or recent weight loss. The patient was a nonsmoker, did not drink alcohol and had no history of injection drug use.

The deletions were then used in an analysis of (i) the relative position on the genome of previously described mutant loci in the flagellar genes, (ii) the relative position of a newly defined cistron, flaN, and (iii) the orientation and direction of transcription of genes previously assigned to multicistronic transcriptional units. As a result of this analysis and previous work, we report the following arrangement of the flagellar genes in the his-aroD region of the E. coli genetic map: his-flaR-flaQ-flaP-flaA-flaE-flaO-flaC-flaB-flaN-hag-flaD-uvrC-flaI-mot-flaG-flaH. The genes flaA, P, Q, and R, flaB, C, 0 and E, and flaG and H are co-transcribed in that order.

In an era of evidence-based practice (EBP), it is becoming increasingly important to distinguish ... more In an era of evidence-based practice (EBP), it is becoming increasingly important to distinguish the quality of research studies and synthesize results so they can be applied to clinical practice. Thus, in an attempt to categorize research and amalgamate results, scholars have developed various hierarchical levels of evidence to differentiate research implications. However, these levels of evidence have not yet been applied to the psychiatric music therapy literature base. The purpose of this paper was to discuss and identify the levels of evidence and apply well-established levels of evidence to the psychiatric music therapy literature base. Results indicated a lack of randomized controlled trials and overall low level of evidence. Further, regardless of taxonomy applied, most studies met criteria for the lowest level of evidence. This finding is congruent with the levels of evidence of other well-established psychosocial treatments for psychiatric consumers. Limitations, generalizations, and implications for research and clinical practice are provided.

The polar flagellum is responsible for motility in a liquid environment (swimming), and the lateral flagella enable the bacteria to move over surfaces (swarming). Synthesis of lateral flagella occurs when V. parahaemolyticus is grown on agar media but not when it is grown in liquid media. We used lux (luminescence gene) fusions to conveniently and sensitively analyze the factors which influence transcription of lateral flagella genes (laf). Transposon mini-Mu lux was used to mutagenize V. parahaemolyticus and to generate laf::lux transcriptional fusions. Mutants with insertions of mini-Mu lux in laf genes were defective in the swarming phenotype and produced light when the bacteria were propagated on agar media, but not when cells were grown in liquid media. Thus, surface-dependent expression of lateral flagella synthesis is controlled by regulation of transcription. Such fusion strains were also used to further define the environmental conditions which induce laf gene expression. Cultivation on media solidified by gelling agents other than agar also induced light production in fusion strains, as did growth on a variety of hydrophilic membrane filters suspended over liquid media. Growth at an air-surface interface was not necessary for expression since embedding the fusion strains in agar was also effective. Furthermore, induction of laf gene transcription could also be accomplished by increasing the viscosity of the liquid medium by the addition of a high-molecular-weight polymer such as polyvinylpyrrolidone. Increase in luminescence of the fusion strains was detected within 30 min of initiation of the inducing circumstance, and reversal of induction, e.g., by dilution of the viscous medium, resulted in a rapid decline in the rate of increase in luminescence. Conditions that induced luminescence in the fusion strains also induced the synthesis of lateral flagella in wild-type V. parahaemolyticus. The growth environment of the fusion strains was manipulated extensively to identify those conditions which resulted in transcription of laf genes, and it appears that the signal that triggers laf expression is physical rather than chemical in nature. Possibilities for a sensing mechanism are discussed.
