However, the drug requires frequent dosing due to limited bioavailability (10–26.7%). The rationale of the present study was to develop a self-dissolving microneedle system for local and systemic delivery of acyclovir using a topical lyophilized wafer on microneedle-treated skin to provide the drug at the site of infection. The microneedles prepared with hydroxypropyl methylcellulose (HPMC) (8% w/w) or HPMC (8% w/w)-polyvinyl pyrrolidone (PVP) (30% w/w) penetrated excised rat skin, showing sufficient mechanical strength and rapid polymer dissolution. The topical wafer was prepared with acyclovir (40% w/w; equivalent to 200 mg of drug), gelatin (10% w/w), mannitol (5% w/w), and sodium chloride (5% w/w). The in vitro skin permeation study was conducted on Franz diffusion cell using phosphate buffer pH 7.4 as receptor fluid. Abstract Metal-doped zinc oxide nanostructures (ZONSs) of schemes CuxZn1−xO and MgxZn1−xO (x = 0,... more Abstract Metal-doped zinc oxide nanostructures (ZONSs) of schemes CuxZn1−xO and MgxZn1−xO (x = 0, 0.01, 0.1 and 0.5 M) were synthesized via co-precipitation technique. The structural parameters of as-prepared ZONSs were investigated from XRD technique and doping concentration limit for both types of dopant was found, causing a shift in the Eg value based on the availability of density of states for that dopant. SEM analysis confirmed thin sheet-like nanocrystalline morphologies of all samples within size range 78–450 nm. Optical analysis revealed that higher concentration of Cu in the sample resulted in overlapped impurity states following a considerable increase in its Eg value. Among all samples, Cu0.1 and Mg0.01 samples with reduced PL emission, exhibited maximum degradation of MB dye under direct-sunlight exposure of few seconds. As positively charged metal-doped ZONSs attract more toward Gram-negative bacterial membrane, the suggested samples have shown improved antibacterial performance against Klabsiella strains.

The present study evaluated the genotoxic and mutagenic effect of Kalanchoe laciniata to access the safety and usefulness of the medicinal plant. Aqua-methanolic and n-hexane extracts of K. laciniata were evaluated for the genotoxic potential using Ames assay and cytotoxicity was evaluated using MTT assay. Ames assay was conducted using two strains of Salmonella typhimurium TA-100 and TA-102 whereas MTT assay was performed on baby hamster kidney cell line BHK-21. Aqua-methanolic extract of K.laciniata exhibited significant mutagenicity when exposed to TA-102 strain with a mutagenic index of 50.66 and 54.74 at maximum dose 150 mg/plate. The extract was also mutagenic to TA-100 strain but to a lesser extent. Optical analysis revealed that higher concentration of Cu in the sample resulted in overlapped impurity states following a considerable increase in its Eg value. Among all samples, Cu0.1 and Mg0.01 samples with reduced PL emission, exhibited maximum degradation of MB dye under direct-sunlight exposure of few seconds. As positively charged metal-doped ZONSs attract more toward Gram-negative bacterial membrane, the suggested samples have shown improved antibacterial performance against Klabsiella strains.

Many aspects of this condition remain elusive, including its underlying mechanisms, ultimately hindering the development of a cure. Interdisciplinary approaches are required to overcome long-established research challenges. This review summarizes current knowledge in various tinnitus-relevant research fields including tinnitus generating mechanisms, heterogeneity, epidemiology, assessment, and treatment development, in an effort to highlight the main challenges and provide suggestions for future research to overcome them. Polypill was made by the compression coating technique (CCT) with multiple active ingredients along with different concentrations of mucoadhesive and sustained release polymers, i.e., Carbopol 934 (CAB), Methocel k15 (MTH) and sodium carboxymethyl cellulose (NaCMC). The present study evaluated the genotoxic and mutagenic effect of Kalanchoe laciniata to access the safety and usefulness of the medicinal plant. Aqua-methanolic and n-hexane extracts of K. laciniata were evaluated for the genotoxic potential using Ames assay and cytotoxicity was evaluated using MTT assay. Ames assay was conducted using two strains of Salmonella typhimurium TA-100 and TA-102 whereas MTT assay was performed on baby hamster kidney cell line BHK-21. Aqua-methanolic extract of K.laciniata exhibited significant mutagenicity when exposed to TA-102 strain with a mutagenic index of 50.66 and 54.74 at maximum dose 150 mg/plate. The extract was also mutagenic to TA-100 strain but to a lesser extent.
