Group contribution methods are suitable tools for estimating many physicochemical properties of p... more Group contribution methods are suitable tools for estimating many physicochemical properties of pure compounds and mixtures. The classical group contribution method decomposes each chemical compound into first, second, and third-order functional groups based on its molecular structure. The most significant advantage of these methods is that they need only the compounds chemical structure without any other input information. From these approaches, structural fragments and subsequent group contribution methods can be established. The knowledge of the molecular structure and its decomposing into molecular fragments can optimize the production of biosynthesis compounds for chemical and pharmaceutical industries. The current research proposes an innovative group contribution application as selection criteria to culture media carbon and nitrogen sources. The concept is based on group contribution to identifying molecular fragments present in tacrolimus structure. Tacrolimus is a macrolide lactone originally obtained from fermentation of Streptomyces tsukubaensis broth. This drug decreases the occurrence and severity of refractory rejection episodes. In addition, tacrolimus is recommended for the treatment of autoimmune diseases. In order to investigate the application of the group contribution, tacrolimus molecular structure and fermentation results of this macrolide were analyzed. This approach can enhance productivity of this important immunosuppressant.
Tacrolimus is an immunosuppressant used in the treatment of people who have undergone organ and t... more Tacrolimus is an immunosuppressant used in the treatment of people who have undergone organ and tissue transplants, as well as in the treatment of dermatoses and eye diseases. Strategies have been carried out to increase the productivity of tacrolimus, since many compounds have in their composition tacrolimus precursors, and bacteria Streptomyces depend on a carbon source for their growth. One strategy is to change the carbon source of the fermentation medium. The present study aims to evaluate the use of coconut oil for the production of tacrolimus via Streptomyces tsukubaensis, as an alternative in the face of glucose, a traditional source of carbon. The batch fermentation process was done in an orbital shaker at 28°C and 130 rpm. Quantification of tacrolimus was performed by High Performance Liquid Chromatography (HPLC). The sugars and proteins, present in the medium, were measured from Somogyi-Nelson and Bradford methods, respectively. According to the results, coconut oil achieved the production of tacrolimus higher than glucose. This is due to the presence of a portion of monounsaturated and polyunsaturated fatty acids that act as precursors to tacrolimus. On the other hand, glucose is a quick carbon source for the bacteria and helps in its growth, but in high amounts it inhibits the production of the drug.

Pastelike materials are encountered in many technological processes in chemical, pharmaceutical, ... more Pastelike materials are encountered in many technological processes in chemical, pharmaceutical, foodstuff and natural product industries. The most important factor in the drying of this type of materials is the nature of the moisture bonding that occurs. Because of the different characteristics of pastes, it is impossible to recommend a universal type of dryer for all of these materials. Some of the dryers available provide only indirect contact with the drying agent (heat) and also maintain constant moisture with a system of rotating paddles. We evaluated the performance of this type by studying the dryer kinetics curves for ground coffee under a variety of operational conditions of moisture load of material, temperature of the heating plate, intensity of the mechanical mixing of the moisture material, and initial moisture. The effects of these parameters (except for moisture) were studied using a 2 3 factorial design. According the analyses of the kinetics drying curves, was observed that the increase in the temperature of the plate and rotation as well as the decrease in the load facilitates more effective removal of moisture. In statistical analysis was determined that the load of the material and the heating plate temperature influence the final moisture content of the material and plate temperature modifies the final temperature of the solid. Also, was suggested linear models from the factorial design to describe the process of drying coffee grounds satisfactorily.

Abstract The terpene and aromatic ketones are among oxygenated compounds present in essential oil... more Abstract The terpene and aromatic ketones are among oxygenated compounds present in essential oils (Eos). They are substances of great importance to the chemical, pharmaceutical, and the food industries. Because of their applications in these industries, it is extremely necessary to know their thermodynamic properties and to obtain reliable data of these properties. This work aims to determinate experimentally the enthalpy of vaporization (∆Hvap) and normal boiling temperature (Tnb) of some terpene and aromatic ketones and other oxygenated compounds present in essential oils by the gas chromatography technique (GC). The additional goal is to evaluate three group contribution models developed to estimate enthalpies of vaporization. The experimental determination of thermodynamic data using GC involves the use of the chromatographic retention index in combination with Kirchhoff-Rankin type equations. Gas chromatography experiments were carried out within a defined temperature range consistent with the nature of the compounds studied. In all, 11 compounds were evaluated. The ∆Hvap data were determined at 298.15 K. The results obtained agreed with those found in the literature, showing small relative deviations (RD), demonstrating that the CG technique can be a good tool for obtaining the thermodynamic properties studied. In addition, unpublished data of ∆Hvap at 298.15 K and Tnb for the compounds bicyclo[2,2,2]octan-2,5-dione, methyl-jasmonate and 4-(4-hydroxyphenyl)-2-butanone are reported.

The calibration constant value was obtained using thymol as reference compound, due to the fact that compound represents the majority in the essential oil. To check the calibration, the vapor pressure data for carvacrol have been compared with the results reported in the literature and showed a good agreement. The method was used in the determination of the vapor pressure curve for the essential oil. From vapor curves, the Antoine constants for the essential oil were found to be: A = 10.29230, B = 3116.68 and C = 74.23, respectively.
In this paper, a multiphase model applied to a downer reactor is proposed, where the gas-solid mo... more In this paper, a multiphase model applied to a downer reactor is proposed, where the gas-solid modeled with some special characteristics, such as: unsteady and 3D flow for two fluids on an Eulerian-Eulerian approach; dissipation of turbulent kinetic energy only on the gas phase modeled as real fluid; and the assumption that the solid phase behaves like a hypothetical fluid without shear stress from molecular causes. Studies based on first (HIPWIND) and second order (HIGHER UPWIND) interpolation schemes were carried out. The difference in the results using the two schemes was found to be negligible. The Computational Fluid Dynamic Technique (CFD) was adopted as a tool to predict downer reactor dynamic, comparing the proposed model with experimental data available in the literature. Good agreement between experimental data and model predictions was verified. Therefore CFD techniques proved to be a powerful tool to predict the fluid dynamic of inlet changing of downer reactors, and it can be used with success in chemical process optimization.

The aim of this paper it to contextualize the importance of the hope as essential element in chemical design to obtain tacrolimus, and to professionals involved in this research. The effect of a reaction of dissolved ozone in an alkaline water solution with hydrogen peroxide ... more The effect of a reaction of dissolved ozone in an alkaline water solution with hydrogen peroxide was evaluated. Assays were conducted in a bubble column with a gaseous ozone concentration (mixed with oxygen) at 21 and 27 g m-3. Concentration curve of dissolved ozone in the liquid revealed an ozone concentration peak in the liquid phase, subsequent decreasing concentration and constant maintenance in the stationary phase. Highest concentration was not detected for the lowest ozone concentration in the gaseous phase. Peak was due to the self-decomposition of ozone in the liquid phase and the production of hydrogen peroxide. The latter reacted with molecular dissolved ozone and decreased ozone concentration after maximum concentration.
