Genetic diversity and population structure of Eplingiella species (Lamiaceae) using ISSR markers

The present study evaluated the population structure and genetic diversity using ISSR molecular m... more The present study evaluated the population structure and genetic diversity using ISSR molecular markers in 18 natural populations belonging to three species of Eplingiella (E. cuniloides, E. fruticosa and E. brightoniae), found growing naturally in the semiarid region of Northeast Brazil. Samples of 265 plants were analyzed using nine primer combinations, which generated 131 informative bands. Eplingiella spp. populations showed moderate genetic diversity (percentage of polymorphic bands, PPB = 75.6-96.9%, Nei's genetic diversity He = 0.31-0.39, Shannon's information index I = 0.33-0.48). Molecular variance analysis (AMOVA) revealed that within populations, variations contributed more (74%) to the genetic diversity than between population variations (26%), with percentage of the genetic differentiation coe cient (G ST = 0.29). The mean value of F ST was 0.175, demonstrating good differentiation between populations. The analysis of the structure by the Bayesian method revealed the formation of two groups (K = 2), with many migrant individuals and a high level of miscegenation. The hierarchical cluster dendrogram grouped the 18 populations into two major clusters, with good support for the main clades (100%). According to principal component analysis (PCA), the two main principal components explained 21.06% of the total variation. The ISSR molecular markers used were effective in identifying the variability of natural populations of Eplingiella spp., and population structure demonstrated recent diversi cation of species. The results shed more light on the genetic variation and evolutionary dynamics of Eplingiella, helping to formulate effective breeding strategies.

Annona glabra L. is one of the most important wild annonaceae with a high potential to produce fr... more Annona glabra L. is one of the most important wild annonaceae with a high potential to produce fruits and pharmacological compounds. In vitro cultivation is a viable alternative for large scale production of plantlets of this species, overcoming the natural constraints for its propagation. However, its use in a broad form has been limited by the difficulty to induce various shoots in explants, high rate of leaf abscission and reduced plant survival index during the acclimatization phase. Among the factors that control the in vitro morphogenesis of this species, cytokinins affect diverse morphological responses. However, a better understanding of the action of these substances on the morphogenetic process of the annonaceae is still necessary, in spite of its broad use on in vitro cultivation of plant tissues. Therefore this paper evaluated the effect of four cytokinin sources, one natural and three syntheticis, on leaves and shoots anatomy, delay in leaf senescence, development of autotrophism and mineral accumulation in plants of A. glabra. Among the cytokinin sources tested, 6-benzilaminopurine (BAP) and kinetin induced an expressive increment in palisade and spongy parenchyma thickness, higher retention on leaf area, higher chloroplast differentiation and higher photosynthetic rate in shoots. The use of BAP and kinetin during the multiplication phase promoted a higher dry matter accumulation, higher development of leaf area and higher plant survival rate during the acclimatization and rooting phases. All the sources of cytokinin used stimulated a higher nitrogen and boron accumulation in shoots and lower accumulation of calcium. Lippia origanoides Kunth. is a medicinal plant that is widely available in the Northeast region o... more Lippia origanoides Kunth. is a medicinal plant that is widely available in the Northeast region of Brazil and is known as "alecrim-d'angola". However, there is no information available on the genetic variability of this species in the region. Thus, the current study was aimed to analyze the genetic diversity and structuring of L. origanoides populations occurring in the states of Bahia and Pernambuco, Brazil, using Inter-Simple Sequence Repeat (ISSR) molecular markers. The evaluated Nei's diversity index of the populations varied from 0.162 to 0.237, and the Shannon diversity index varied from 0.247 to 0.350. In molecular variance (AMOVA) analysis, a variation of 31% was observed among the populations, which denotes a high interpopulation structuring. The structure analysis and dendrogram indicated the possibility of classifying the 18 populations into four groups. As their genetic structure is extremely high, it is important to collect L. origanoides germplasm, including as many populations as possible. Since the region of Chapada Diamantina holds the most diverse populations of L. origanoides germplasm, it is a priority area to obtain the germplasm.

This work describes an in vitro propagation protocol for the large-scale cultivation of Leptohypt... more This work describes an in vitro propagation protocol for the large-scale cultivation of Leptohyptis macrostachys (Benth.) Harley & JFB Pastore and the influence of abiotic factors on podophyllotoxin and yatein production. The plant was established from seeds collected in Chapada Diamantina, BA, Brazil and submitted to different growth mediums and physical conditions. The podophyllotoxin and yatein contents were quantified by HPLC/DAD and with pure standards and these two lignans were present in all experiments. The lignan quantities were evaluated using the Sisvar Program, compared by Tukey's test and hierarchical cluster analysis and principal component analysis. In all experiments, podophyllotoxin and yatein were detected at different concentrations. The best protocol cultivar of L. macrostachys was established from seeds in MS ½ medium supplemented with 1.5% sucrose and 11.55 μM of gibberellic acid A3 (GA 3) at 30°C, which yielded the highest concentration of podophyllotoxin (5.831 mg g −1). These results are important findings for the production of podophyllotoxin from the tissue culture.

Leaf samples were analyzed with light microscopy and scanning electron microscopy to describe the structure and to quantify the trichome frequency. Six types of glandular trichomes were identified: uni, bi and tetracellular, as well as three types of tectorial trichomes. L. bromleyana presents an ornamented epidermis and no tectorial trichomes on the abaxial surface. L. thymoides has glandular trichomes with irregular contours on both leaf faces, with a higher frequency of tectorial trichomes on the adaxial surface and peltate glandular trichomes on the abaxial surface. L. insignis and L. lasiocalycina are anatomically similar, and present a higher tector trichome frequency on the abaxial surface compared to the others species. Hyptis leucocephala e Hyptis platanifolia (Lamiaceae) são espécies aromáticas endêmicas do semiár... more Hyptis leucocephala e Hyptis platanifolia (Lamiaceae) são espécies aromáticas endêmicas do semiárido nordestino e possuem grande importância econômica devido ao seu potencial medicinal. O presente trabalho teve por objetivo obter o protocolo para o estabelecimento inicial in vitro das espécies H. leucocephala e H. platanifolia. O meio de cultura WPM promoveu a maior porcentagem de germinação (96,81%) para H. leucocephala, porém não diferiu estatisticamente dos resultados obtidos no MS½ na última semana de cultivo. Para a germinação de sementes de H. platanifolia, o MS½ foi o meio que proporcionou a maior porcentagem (40,83%). O tipo de esterilização não interferiu na germinação in vitro para as duas espécies. Para a espécie H. platanifolia, quando se utilizou o fechamento dos tubos de ensaio com tampão de algodão, a hiperidricidade foi eliminada; porém, a porcentagem de germinação das sementes foi menor. Ageratum (Asteraceae) é nativo das Américas, sendo A. conyzoides a espécie mais explorada para di... more Ageratum (Asteraceae) é nativo das Américas, sendo A. conyzoides a espécie mais explorada para diversos fins em diferentes partes do mundo. O objetivo deste trabalho foi prospectar publicações científicas e patentes com os representantes do gênero. A consulta foi realizada em bancos de publicações científicas e de patentes, utilizando os termos Ageratum e mentrasto. Foi possível observar um expressivo número de publicações científicas (761) em detrimento dos depósitos de patentes (294) e em ambos os documentos se destaca a área medicinal e agronômica, sendo a China a maior detentora dos depósitos (90%). Nenhuma patente concedida ou em avaliação foi encontrada no Instituto Nacional da Propriedade Industrial (INPI), apesar de terem sido compiladas 37 dissertações/teses no Brasil na área farmacológica e agronômica, campos de grande interesse comercial. Isso demonstra a falta de incentivo à propriedade intelectual na Academia do País e a necessidade das pesquisas estarem alinhadas para solucionar alguma carência da sociedade. Ageratum (Asteraceae) is native to the Americas and A. conyzoides is the most exploited species for a variety of purposes in different parts of the world. The aim of this study was to investigate scientific publications and patents with representatives of the genus. The survey used the terms Ageratum and goat weed. A significant number of scientific publications (761) compared to patent deposits (294) were observed, with China holding the largest number of deposits (90%). No patent was awarded or under consideration at INPI (National Institute of Industrial Property), despite the fact that 37 dissertations/theses have been submitted in Brazil, in the area of pharmacology and agronomy, fields of considerable commercial interest. This demonstrates the lack of support for intellectual property in academic settings and the need for research to bridge societal gaps.

Apesar do grande potencial farmacologico, trata-se de uma especie ainda nao cultivada, sendo necessario o desenvolvimento de metodologias para sua propagacao e cultivo, o que viabilizara a sua exploracao de forma sustentavel. Este trabalho teve como objetivo avaliar a influencia de diferentes substratos e concentracoes de acido indolbutirico (AIB) na propagacao de L. origanoides por estacas apicais. O delineamento experimental utilizado foi inteiramente casualizado em esquema de parcelas subdivididas, tendo substratos como parcelas e concentracoes de AIB como subparcelas. Avaliou-se porcentagem de enraizamento e sobrevivencia das estacas, comprimento do caule e da maior raiz, numero de folhas por muda, massa da materia seca de folhas, caule, raiz e total. Verificou-se que e possivel a propagacao vegetativa da especie L. origanoides a partir de estacas apicais e que a imersao das estacas em AIB seguida de cultivo em substrato composto por Biomix ® e vermiculita expandida (1:1) proporciona maior sobrevivencia e crescimento inicial das mudas.

Given that it is a fast-growing and highly productive species, determining the most adequate nitrogen (N) doses could contribute to higher crop yields. This study aimed at assessing the influence of N concentrations, in a hydroponic system, on the growth and production of P. angulata , as well as determining the critical N level in leaves. The experiment was conducted in individual pots with nutrient solutions, applying a completely randomized design and twelve replications, using five N doses (0 mg L -1 , 56 mg L -1 , 112 mg L -1 , 168 mg L -1 and 224 mg L -1 ). Despite its richness in bioactive molecules, with several scientifically proven applications, there is little information on the insecticidal potential of its species. This study aimed to assess the insecticidal potential of essential oils from the species Lippia thymoides (Martius & Schauer); Lippia lasiocalycina (Schauer) and Lippia insignis (Moldenk) against Nasutitermes corniger (Motschulsky) (Isoptera-Termitidae). Insecticidal activity was evaluated by exposure to a contaminated surface, whereby plastic pots were lined with filter paper and imbibed in 1.5 ml of solution containing essential oils (10 µl/ml), with 10 N. corniger specimens per pot. Propagation is difficult primarily due, to a reduced seed germination and occurrence of recalcitrant seeds that stimulate the search of rapid and efficient propagation alternatives. In this context, the aim of this work was to study in vitro seed germination conditions in order to produce explants to be used on in vitro culture. The effect of different culture media, sucrose and GA 3 concentrations and three pH levels were evaluated. Seeds were extracted from mature fruits after being harvested and processed. Higher in vitro germination was obtained using WPM and MS/2 media supplemented with 15.0 g L -1 , 0.2 mg L -1 GA 3 and pH adjusted to 5.8.

ABSTRACT Aims In the semi-arid region of Bahia State, Brazil, sisal (Agave sisalana Perrine ex En... more ABSTRACT Aims In the semi-arid region of Bahia State, Brazil, sisal (Agave sisalana Perrine ex Engelm) has been cultivated for fibre production for several decades without the use of chemical fertilizers. The contribution of biological nitrogen fixation (BNF) to this crop and the occurrence of diazotrophic rhizosphere, epiphytic or endophytic bacteria have not been investigated. The aims of this work were to study the occurrence and diversity of diazotrophic bacteria in rhizosphere soil, root and leaf tissues of sisal plants from this region, as well as to test their potential for plant growth promotion. Methods Burk’s nitrogen free semi-solid medium was used for isolation and the MPN method was used for quantification of diazotrophic bacteria. BOX-A1R PCR and 16S rRNA sequence analyses were performed to study the diversity of bacterial isolates harboring the nifH-gene. Six isolates were selected for growth promotion of cucumber plants. Results A high diversity of BNF bacterial isolates was observed in sisal plants and rhizosphere soil. Bacterial populations were higher in roots, followed by soil and sisal leaves. Burkholderia, Leifsonia and Paenibacillus were the predominant genera. All isolates tested were able to promote cucumber growth. Conclusions This study showed that sisal-associated diazotrophic bacteria are diverse and may be further exploited to promote plant growth.

Hypocotyl and epicotyl segments were inoculated into test tubes with WPM medium supplemented with 8.87 µM BAP in the following orientations: horizontally on the medium surface, vertically upright retaining their natural polarity and vertically upright but inverted from their natural polarity (hypocotyl segments only). The test tubes were sealed with normal transparent plastic caps (either covered or not covered with PVC film) or with cotton plugs. After inoculation the tubes with the explants were maintained in a growth room under photosynthetically active radiation levels of 45-56 ìmol.m-2.s-1 and temperatures of 25 ± 3oC. The results showed that explant polarity affected organogenesis, and that the largest numbers of shoots (8.3 per explant) were found on hypocotyl explants placed vertically and retaining their natural polarity.
