Byl sledovan vliv rostouci koncentrace etanolu v přitoku živneho media na růst a fyziologickou ak... more Byl sledovan vliv rostouci koncentrace etanolu v přitoku živneho media na růst a fyziologickou aktivitu buněk Candida utilis. Experimenty byly provaděny v kaskadě tři fermentorů zapojených v serii při konstantni hodnotě zřeďovaci rychlosti, teploty a pH media. V ustalených stavech kontinualnich kultivaci se měřila koncentrace biomasy, etanolu a kyseliny octove v mediu, výtěžnost biomasy, produktivita procesu a respiracni aktivita buněcne populace. Maximalni koncentrace biomasy ve výtoku z fermentoru a maximalni produktivity se dosahlo při koncentraci etanolu v přitoku v rozsahu 20 až 25 g.l-1. Ve vyssich koncentracich působil etanol negativně na spřaženi v procesu oxidativni fosforylace a respiracni kontrolu buněk, což se projevilo poklesem výtěžnosti biomasy i poklesem obsahu bilkovin v buňkach. Bylo prokazano, že přitomnost acetatu v mediu inhibuje respiracni aktivitu buněk rostoucich na etanolu.

An inhibitory ethanol concentration (30 g/L) slows down acidification but has no effect on its extent. The transmembrane Apl 1 depends directly on the ethanol level in the medium and is not lowered at high ethanol concentrations. Changes in membrane potential induced by ethanol are also concentration dependent. The high ethanol level does not increase the passive permeability of cell membrane to H +.

In O2-aerated non-growing cells ethanol causes an H ÷ / K + exchange and an extrusion of acetate and lactate accompanied mostly by K ÷, and their subsequent reimportation together with H +. Cells from continuous culture display generally stronger acidification and more marked K + movements than non-growing ones.

This aim of study was to compare the performance of a biofilter (BF) and trickle bed reactor (TBR... more This aim of study was to compare the performance of a biofilter (BF) and trickle bed reactor (TBR) under increased styrene loading with a constant acetone load, 2 g c /m 3 /h. At styrene loading rates up to 30 g c /m 3 /h, the BF showed higher styrene removal than TBR. However, the BF efficiency started to drop beyond this threshold loading and could never reach steady state, whereas the TBR continued to yield a 50% styrene removal. The acetone removal remained constant (93-98%) in both the reactors at any styrene loading. Once the overloading was lifted, the BF recovered within 26 min, whereas the TBR efficiency bounced back only to 95%, gradually returning to complete removal only in 10 h.
