My background encompasses a wide range of research areas, including nucleic acid methodologies (RNA and DNA), gene cloning, protein expression and purification, antibiotic resistance, lipid analysis, lipid nanoparticle formulation and characterization, binding affinity and kinetics measurements, structure-activity relationships, bioconjugation, and quality control.

From an early age, I have been captivated by fundamental existential questions, exploring the origins of life, the principles of complexity, and the intricate web of interconnected systems. This passion led me to study biology and physical and analytical chemistry before delving into homeopathy.

My current research focuses on water, its remarkable ability to store information, the tunability of its molecular network, and its function as an antenna capable of receiving and transmitting signals. I am developing a new hypothesis linking water to consciousness, a pursuit that requires bridging disciplines, leveraging analytical techniques, and synthesizing insights across scientific domains.

L’etude des systemes supramoleculaires photoactifs basee sur l’un des derives hydroxyles de la meso (tetraphenyl) porphyrine (m-THPP) utilise comme une sonde de fluorescence dans le but analytique de de��tecter des phospholipides, fait l’objet de la premiere partie de ce travail. La deuxieme partie concerne l’interaction de neuf derives hydroxyles et glucoconjugues de 5,10,15,20-meso-tetraphenyl porphyrine avec les dimyristoyl-phosphatidyl choline (DMPC) liposomes et l’albumine dans le but d’evaluer l’impact de la modification structurale. Ces porphyrines ont ete synthetisees comme photosensibilisateurs potentiels pour la therapie photodynamique des cancers.

Investigating the possible use of a tetra (hydroxyphenyl) porphyrin as a fluorescence probe for the supramolecular detection of phospholipids

The present work explores the interest of using the 5,10,15,20-tetrakis(3-hydroxyphenyl)-21H,23Hp... more The present work explores the interest of using the 5,10,15,20-tetrakis(3-hydroxyphenyl)-21H,23Hporphine (m-THPP), a commercially available porphyrin, as a polarity probe for the detection of phospholipids. Feasibility of the process was first tested by direct fluorimetry and it was established that emission enhancement in the presence of lipids was maximum for a methanol content of 15% (v/v), a neutral pH (6-8) and a probe concentration of 2 Â 10 À 7 M in the working solution. Detection with m-THPP in these conditions presents higher response factor but shorter linear range than diphenylhexatriene (DPH), the reference dye for the supramolecular detection of lipids. Insights on the mechanism of m-THPP-lipid association demonstrated that, at high lipid concentrations, porphyrin is associated to lipid assemblies in monomer form and located in the interior of bilayer; in low lipid content, porphyrin aggregates coexist with the monomer. Finally it was assessed that the method can be successfully coupled with micro-bore liquid chromatography.

Three series of glycoconjugated and hydroxylated derivatives of 5,10,15,20-meso-tetraphenyl porph... more Three series of glycoconjugated and hydroxylated derivatives of 5,10,15,20-meso-tetraphenyl porphyrin (TPP) were studied in order to evaluate the effect of a porphyrin structure on its binding to dimyristoylphosphatidylcholine (DMPC) liposomes and to human serum albumin (HSA). The studied derivatives have been developed as potent photosensitizers for photodynamic therapy (PDT) of cancers. Steady state and time resolved fluorescence emission spectroscopy, Stern-Volmer quenching and fluorescence anisotropy were used for this evaluation. The lipophilicity of the compounds has been deduced from their retention time in reverse phase liquid chromatography. The results demonstrated that the more polar glycoconjugated compounds presented limited aggregation in aqueous media and very rapid binding kinetics to DMPC liposomes and HSA. Derivatives having intermediate or high hydrophobicity showed extensive auto-association in aqueous media and as a consequence slow association kinetics. The strength of porphyrin binding to DMPC liposomes also depended on their lipophilicity and was lower for the polar glycoconjugated analogues. The highest affinity for liposomes was observed for hydroxylated derivatives with intermediate lipophilicity. In contrast, the highest binding constant for albumin was observed for a polar tetra-glycoconjugated analogue. The depth of penetration into the phospholipid bilayer did not appear to be directly related to the global hydrophobicity of the compounds, but depended more on the number of apolar, non-substituted phenyl groups grafted to a tetrapyrrolic macrocycle. Furthermore, liposome-albumin competition studies revealed that the porphyrins were always mainly partitioned into the phospholipid bilayer.

DNA G-Quadruplexes Activate Heme for Robust Catalysis of Carbene Transfer Reactions

Guanine-rich single-stranded DNAs and RNAs that fold into G-quadruplexes (GQs) are known to compl... more Guanine-rich single-stranded DNAs and RNAs that fold into G-quadruplexes (GQs) are known to complex tightly with Fe II -heme and Fe III -heme (hemin), ubiquitous cellular cofactors. Heme-GQ (DNA) complexes, known as heme•DNAzymes, are able to utilize hydrogen peroxide as an oxidant to vigorously catalyze a variety of one-electron (peroxidase) and two-electron (peroxygenase) oxidation reactions. Herein, we show that complexes of Fe II -heme with GQs also robustly catalyze a mechanistically distinct reaction, carbene transfer to an alkene substrate. Significant enhancements were seen in both reaction kinetics and product turnover (∼180) relative to disaggregated Fe II -heme in the absence of DNA or in the presence of other DNA folds, such as single-stranded or double-stranded DNA. Heme binds to GQs by end-stacking. Simple, intramolecularly folded GQs are unable to provide a complexly structured "distal side" environment to the bound heme; therefore, such DNAzymes do not display significant product stereoselectivity. However, intermolecular GQs with multiple pendant nucleotides show increasing stereoselectivity in addition to their enhanced catalytic rates. These results recapitulate the unique functional synergy and highlight the surprising catalytic versatility of complexes formed between heme and DNA/RNA GQs. Our findings suggest that heme•DNAzymes and heme•ribozymes may prove to be useful reagents for carbon-carbon bond forming "green" reactions carried out in vitro and likely within living cells.

Interest of Fluorescence Derivatization and Fluorescence Probe Assisted Post-column Detection of Phospholipids: A Short Review

There are many analytical methods available for the quantitative and qualitative determination of phospholipids, but since these molecules lack chromophores, common absorbance based methods are of limited use. Beside mass spectrometry, some less specific approaches that are routinely used are evaporative light scattering detection or fluorescence, which exhibit sufficient sensitivity. Here, we focus on fluorescence, which remains an interesting way to quantify phospholipids. Two ways of detecting phospholipids by fluorescence are possible coupled with separation techniques such as thin layer chromatography (TLC), high performance liquid chromatography (HPLC) and capillary electrophoresis (CE): firstly, pre-column derivatization procedures and secondly, probe assisted post-column detection with suitable fluorescence reagents. In both cases, the common purpose is to increase the detection sensitivity. It is shown that, whereas pre-column derivatization is characterized by selectivity due to the chemical functionality of the analyte involved in the derivatization process, in supramolecular post-column derivatization, the selectivity only proceeds from the capacity of the lipid to involve supramolecular assemblies with a fluorescence probe. The aim of this review is to summarize available experiments concerning fluorescence detection of phospholipids. The interest and limitation of such detection approaches are discussed.
