En el caso del VIH/Sida, los problemas a los que se enfrenta una persona infectada han cambiado bastante en los últimos tiempos. Anteriormente, los principales esfuerzos se empleaban en aumentar el tiempo de vida de los pacientes, mientras que ahora se pretende mejorar su calidad de vida, afectada por múltiples factores (médicos, psicológicos y sociales).

APOBEC3G (A3G) is an innate defense protein showing activity against retroviruses and retrotransp... more APOBEC3G (A3G) is an innate defense protein showing activity against retroviruses and retrotransposons. Activated CD4(+) T-cells are highly permissive for HIV-1 replication, while resting CD4(+) T-cells are refractory. Dendritic cells (DCs), especially mature DCs, are also refractory. We investigated whether these differences could be related to a differential A3G expression and/or sub-cellular distribution. We found that A3G mRNA and protein expression is very low in resting CD4(+) T-cells and immature DCs, but increases strongly following T-cell activation and DCs maturation. The Apo-7 anti-A3G monoclonal antibody, which was specifically developed, confirmed these differences at the protein level and disclosed that A3G is mainly cytoplasmic in resting CD4(+) T-cells and immature DCs. Filogenia De La Región Del Gen Pol Que Codifica La Integrasa De Los Retrovirus Phylogeny of the Gene Pol Region That Encodes by Retroviral Integrases
ABSTRACT RESUMEN Contexto: La integrasa de los retrovirus cataliza la inserción covalente de una ... more ABSTRACT RESUMEN Contexto: La integrasa de los retrovirus cataliza la inserción covalente de una copia del cDNA viral en el genoma de la célula hospedera. La enzima es codifica por el extremo 3' de la región proviral Pol; presenta tres dominios estructurales: un amino-terminal (1-46 aa), uno central o catalítico (46-219 aa) y uno carboxi-terminal o de unión al ADN (220-300 aa). Objetivos: Definir las relaciones filogenética intra-específica y inter-específicas de miembros de la familia Retroviridae. Métodos: A partir de 28 secuencias completas de la IN de diferentes géneros de retrovirus se calcularon, utilizando el método de Kimura doble parámetro, las distancias genéticas entre los diferentes dominios de la proteína de la integrasa y se definieron sus relaciones filogenéticas. Los árboles filogenéticos se obtuvieron mediante los métodos de NJ y MP con bootstraps de 500. Resultados: Se obtuvo una filogenia de la familia Retroviridae representada por cinco " clados " diferentes. Esta distribución fue similar a las obtenidas previamente con secuencias LTR y de envoltura. Se caracterizaron secuencias canónicas de aminoácidos en el dominio N-terminal (HHCC) y en el dominio central catalítico (DTDGEK). Se determinó que los distintos dominios de las integrasas de miembros de la familia Retroviridae mostraron una amplia variación genética (5,0 a 0,68). El dominio central catalítico presentó el menor rango de distancias genéticas (1,91 a 0,44). Conclusión: Los resultados confirman aquellos previamente obtenidos para LTR y env; en su conjunto, éstos son una evidencia fuerte de que estarían operando mecanismos similares de evolución para los genes retrovirales pol yenv.

BACKGROUND: Non-occupational post-exposure prophylaxis for human immunodeficiency virus (HIV) inf... more BACKGROUND: Non-occupational post-exposure prophylaxis for human immunodeficiency virus (HIV) infection is widely used, although there is little available scientific evidence to support its effectiveness. The aim of this study is to describe the characteristics of the persons exposed, types of exposures, antiretroviral treatment prescribed, and outcome of HIV infection in cases of non-occupational exposure in Spain.METHOD: The data used included all cases of accidental HIV exposure notified to the Non-occupational Post-exposure Prophylaxis Information System between January 2001 and December 2005. Non-occupational exposure to HIV was defined as accidental contact with blood and/or other biological fluids outside the healthcare setting.RESULTS: A total of 993 cases of exposure were notified (569 men [57.3%]); median age was 30 years (range: 1-87). Exposure was sexual in 53.1%, parenteral in 39.8%, and other types in 7.2%. The source person was identified in 82.7% of cases. Antiretroviral treatment (ART) was prescribed in 528 cases (53.2%), with triple therapy in 68.2%. A total of 54.2% returned for the 6-month visit among patients receiving ART and 61.1% among those without this therapy (P < 0.05). One or more side effects developed in 135 (32.4%) cases, and there were 18 treatment interruptions (4.3%). Three seroconversions to HIV were notified (0.3%).CONCLUSIONS: A national registry for monitoring non-occupational post-exposure prophylaxis to HIV is needed because of the high number of cases notified, the considerable incidence of side effects, and the difficulties of follow-up.

In this study, we evaluated the incidence and management of depressive symptoms during IFN-α therapy in HIV-infected patients with CHC. Methods: HIV-infected patients with CHC who began IFNα and ribavirin therapy during the recruitment period April 2001 to April 2003 were included in the study. Patients with a history of major depressive disorder were excluded. Results: Of 113 co-infected patients who started IFN-α therapy during the recruitment period, 45 (40%) developed symptoms of depression (sadness, tiredness and apathy). Twenty of them (44%) were treated with citalopram, a selective serotonin re-uptake inhibitor, resulting in a significant improvement in their symptoms. Most of the patients (60%) showed depressive side effects in the first 3 months after initiation of IFN-α. In addition, during the study, three patients developed psychotic symptoms and one committed suicide.

The genetic variation and population structure of three populations of Anopheles darlingi from Co... more The genetic variation and population structure of three populations of Anopheles darlingi from Colombia were studied using random amplified polymorphic markers (RAPDs) and amplified fragment length polymorphism markers (AFLPs). Six RAPD primers produced 46 polymorphic fragments, while two AFLP primer combinations produced 197 polymorphic fragments from 71 DNA samples. Both of the evaluated genetic markers showed the presence of gene flow, suggesting that Colombian An. darlingi populations are in panmixia. Average genetic diversity, estimated from observed heterozygosity, was 0.374 (RAPD) and 0.309 (AFLP). RAPD and AFLP markers showed little evidence of geographic separation between eastern and western populations; however, the F ST values showed high gene flow between the two western populations (RAPD: F ST = 0.029; Nm: 8.5; AFLP: F ST = 0.051; Nm: 4.7). According to molecular variance analysis (AMOVA), the genetic distance between populations was significant (RAPD:Φ ST = 0.084; AFLP:Φ ST = 0.229, P < 0.001). The F ST distances and AMOVAs using AFLP loci support the differentiation of the Guyana biogeographic province population from those of the Chocó-Magdalena. In this last region, Chocó and Córdoba populations showed the highest genetic flow.

ABSTRACT Predicting the need for fertilizer‐nitrogen (N) topdressing based on plant N status is a... more ABSTRACT Predicting the need for fertilizer‐nitrogen (N) topdressing based on plant N status is an important N management strategy for increasing both grain yield and N‐use efficiency of irrigated rice. Plant N analysis by conventional‐oven drying and micro‐Kjeldahl procedure generally requires several days. Accurate estimation of leaf N content per unit dry weight (Ndw) at different growth stages by the chlorophyll meter requires that meter‐reading values (SPAD values) be adjusted for specific leaf weight (SLW). This study demonstrated that a microwave oven can be used for drying rice leaves for quick estimation of leaf dry weight without significantly influencing the estimation of Ndw by micro‐Kjeldahl procedure. Microwave oven drying of 0.5 g (dry weight) leaf samples required only two minutes. Extended exposure duration up to six minutes did not alter the measured Kjeldahl N concentration. Specific leaf weight determined with microwave‐oven drying improved the prediction of Ndw by SPAD. The chlorophyll meter with SLW adjustment can provide another tool to make rapid, in‐season diagnosis of crop N status.
