Este trabalho foi conduzido com o objetivo de avaliar o efeito da temperatura e do tempo de armazenamento sobre a qualidade de ovos de poedeiras comerciais. O delineamento utilizado foi inteiramente casualizado em esquema fatorial 3x3 sendo três temperaturas (temperatura ambiente -26° C, geladeira -10° C e câmara fria -3° C) e 3 períodos de armazenamento (7, 14 e 21 dias), com 6 repetições, distribuídos em esquema inteiramente aleatorizado em que cada ovo representou uma unidade experimental. As variáveis avaliadas foram perda de peso dos ovos (%), gravidade específica, porcentagem de gema e albúmen, coloração da gema e avaliação sensorial. Os ovos mantidos sob temperaturas de refrigeração (geladeira e câmara fria) apresentaram menores perdas de peso e melhores valores para gravidade específica e coloração de gema crua no período de armazenamento (14 e 21 dias). Para porcentagem de gema e albúmen os ovos não apresentaram diferença (p>0,05) entre os tempos de armazenamento, independente da temperatura a qual foram submetidos. Os ovos mantidos sob refrigeração obtiveram melhores características sensoriais que aqueles mantidos sobre temperatura ambiente. Conclui-se que, nas condições experimentais adotadas, os ovos devem ser mantidos em temperatura de refrigeração (3 a 10° C) para que mantenham sua qualidade preservada. Palavras-chave. In contrast with indomethacin, paracetamol and acetyl salicylic acid, much lower doses of dipyrone were necessary for blocking hyperalgesia (ED50 = 19 mg/kg, i.p.) than oedema (180 mg/kg, i.p.) Dipyrone administered intraperitonially or intraplantarly was able to antagonise PGE2-, isoprenaline- and calcium chloride-induced hyperalgesia, effects which are not observed with non-steroid anti-inflammatory drugs. Systemic or local administration of dipyrone had no effect upon Db-cAMP-induced hyperalgesia while a centrally acting analgesic, morphine, given systemically, was highly effective. These results support our suggestion that the mechanism of action of dipyrone is different from that of classical non-steroidal anti-inflammatory drugs. Although the site of action is peripheral its analgesic effect does not derive from inhibition of the synthesis of prostaglandins but is exerted via direct blockade of the inflammatory hyperalgesia.

Relationships between increased vascular permeability, oedema, hyperalgesia and the effect of non-steroid anti-inflammatory drugs

The relationship between oedema, increased permeability and hyperalgesia has been a matter of deb... more The relationship between oedema, increased permeability and hyperalgesia has been a matter of debate for some time. On one side there are those who concluded that oedema formation and protein leakage are closely linked.l,2, but others have evidence indicating that the processes are independent3,4,5. A parallelism between the development of oedema and an increase in permeability during the development of inflammation induced by carrageenan was found in studies utilizing labelled protein6’7. However, Garcia Lerne et al.8 did not find this parallelism during the entire period of observation. When they measured the seepage of protein-bound Evans Blue into a perfusate (coaxial perfusion), they found that the major increase in permeability occurred in the early stages but that only a small residual increase in permeability was demonstrable at the time oedema was maximal (4th hour). These investigators also showed that non-steroid anti-inflammatory drugs, which are prostaglandin synthesis inhibitors, were only effective when given before administration of carrageenan. This observation is not consistent with the suggestion that prostaglandins are the mediators of the late phase of carrageenan oedema6’9. In addition, non-steroid anti-inflammatory drugs such as aspirin and phenylbutazone were shown to be effective in blocking oedema when given 2 h after carrageenan10. Hyperalgesia, on the other hand, had been shown to be more sensitive to curative treatment with aspirin-like drugs than oedema, and that oedema per se does not cause hyperalgesia (for discussion, see References 11 and 12).

Morphine, enkephalins, nalorphine, naloxone and pentazocine are shown to have a peripheral analge... more Morphine, enkephalins, nalorphine, naloxone and pentazocine are shown to have a peripheral analgesic effect. In our modification of the Randall-Selitto test these substances were 50--100 times more potent than a standard local anaesthetic, lidocaine. At this peripheral site, naloxone did not antagonize the effect of morphine. Morphine had a marked analgesic effect on the hyperalgesia induced by PGE2 and PGI2, BaCl2, Ca2+ ionophore A23187, isoprenaline but not on that induced by dibutyryl cyclic AMP. It was suggested that the peripheral analgesic effect of morphine is due to an inhibition of adenylate-cyclase activity.

Este estudo investigou evidências de validade e precisão de um instrumento de diferencial semânti... more Este estudo investigou evidências de validade e precisão de um instrumento de diferencial semântico que tem sido utilizado para medir a transferência de signifi cados entre estímulos equivalentes. Um total de 196 estudantes universitários avaliou fotografi as de faces expressando emoções e também fi guras abstratas, utilizando um instrumento composto por 13 escalas bipolares ancoradas por adjetivos opostos. A análise fatorial dos dados coletados forneceu indicações da validade do instrumento e os coefi cientes alfa obtidos indicam que os dois fatores extraídos nesta análise possuem níveis de precisão aceitáveis, embora um dos fatores deva ser revisto. Discute-se que o modelo de equivalência de estímulos, entendido como um modelo experimental de relações simbólicas, pode se benefi ciar de instrumentos capazes de fornecer medidas válidas de transferência de signifi cado. Nove matrizes receberam dietas compostas basicamente de plantas do ciclo fotossintético C4 durante toda a gestação (d‰13C = 17,12) e lactação (d‰13C = -19,26). Os leitões foram desmamados aos 21 dias de idade e 68 deles foram alimentados com ração composta de plantas do ciclo fotossintético C3 (d‰13C = -25,12). Os leitões foram aleatoriamente distribuídos em duas dietas: uma sem suplementação de glutamina; e outra suplementada com 1% de glutamina. Aos 0; 1; 1,5; 2; 2,5; 3; 3,5; 4; 4,5; 5; 5,5; 8; 11; 15, 20; 29 e 46 dias pós-desmame, dois leitões de cada dieta foram abatidos para coleta de amostras da mucosa intestinal, que foram analisadas quanto à composição isotópica de d‰13C e mensurada a velocidade de substituição do carbono no tempo. Açai fruit (Euterpe oleracea) has significant antioxidant and anti-inflammatory actions. The present study aimed to determine whether oral administration of an açai stone extract (ASE) could reduce lung inflammation induced by CS. Thirty C57BL/6 mice were assigned to three groups (n=10 each): the Control+A group was exposed to ambient air and treated orally with ASE 300 mg/kg/day; the CS group was exposed to smoke from 6 cigarettes per day for 5 days; and the CS+A group was exposed to smoke from 6 cigarettes per day for 5 days and treated orally with ASE (300 mg/kg/day). The antinociceptive effect of clonidine was dose-dependent and was abolished by local administration of the selective a2-adrenoceptor blocker, yohimbine or of the opioid antagonists naloxone or quaternary nalorphine. St-91, a clonidine analog which does not cross the blood-brain barrier also promoted significant antinociception. Repeated administration of drugs possessing a central mechanism of analgesic action leads to the development of tolerance in this test. Significant analgesic tolerance was observed following repeated (5 days) morphine (8 mg/kg) or high doses of clonidine (0.5 mg/kg). In contrast, no tolerance was detected to the analgesic effect of low doses of clonidine (0.15 mg/kg) or of St-91 (0.5 mg/kg). These results suggest that, in addition to its central analgesic action, clonidine can induce peripheral antinociception by an a2-adrenoceptor-mediated local release of enkephalin-like substances.

In the present investigation, we showed that the NTG gel reduces prostaglandin E2-induced hyperalgesia and blocks neurogenic inflammation induced in rat skin by antidromic electrical stimulation of the saphenous nerve. These results offer an explanation for the effects of topical application of NTG observed in thrombophlebitis, which may be common to other cutaneous pathologies. In addition to the central and spinal sites of action of morphine both our laboratory and others ... more In addition to the central and spinal sites of action of morphine both our laboratory and others have demonstrated that opiates can also cause analgesia through a peripheral mechanism . However, the molecular basis of the mechanism of the analgesic actions of opiates remains unknown. In vitro studies show that morphine is able to inhibit activation of adenylate cyclase (Collier and Roy, I974) as well as to stimulate formation of cGMP (Minneman and Iversen, I976). Recently, using the rat paw hyperalgesia test, we showed that in the periphery, acetylcholine-induced analgesia was mediated via the activation of the nitric oxide/cGMP pathwny (Duarte et al., I990). This conclusion was based upon the fact that intraplantar injection of sodium nitroprusside, a substance which nonenzymatically releases nitric oxide (NO), caused analgesia. Furthermore, the analgesic effects of acetylcholine (ACh) and sodium nitroprusside were enhanced by intraplantar injection of an inhibitor of cyclic GMP phosphodiesterase, My5445. In addition, methylene bk:e (MB), an inhibitor of guanylate cyclase, blo1.:ked the analgesia induced by acetylcholine and sodium nitroprusside. On the other hand, the analgesia induced by acetylcholine, but not by sodium nitroprusside, was blocked by N°-monomethyl-L-arginine (L-NMMA), an inhibitor of the formation of NO from L-arginine. Due to the similarity of the local action of ACh and opiates we investigated whether agents which affect the arginine/NO-cGMP pathway also interfere with morphine-induced peripheral analgesia as tested with our modification of the Randall-Selitto rat paw pressure test (see Duarte et al., I990

Glucocorticoids inhibit the migration of neutrophils induced by endotoxin (E. coli lipopolysaccha... more Glucocorticoids inhibit the migration of neutrophils induced by endotoxin (E. coli lipopolysaccharide, LPS) in vivo. Macrophage monolayers stimulated by LPS showed a dose-dependent release into the supernatant of a chemotactic factor for neutrophils, MNCF, which was active in vivo and in vitro. Dexamethasone reduced the neutrophil migration into the abdominal cavities of rats that was induced by LPS, but did not affect the migration induced by MNCF. The release of MNCF by LPS-pretreated macrophage monolayers was inhibited by dexamethasone and hydrocortisone but not by indomethacin and BW755C. MNCF was stable at 56 degrees C and did not release histamine from mast cells. Thus, MNCF activity seems not to be due to arachidonic acid metabolites or C5/C5a. MNCF shares some properties with interleukin-I, such as the blockade of its release by glucocorticoids, the association of its activity with a material of a molecular weight greater than 10 000 Daltons and the abolition of its activity by incubation with phenylglyoxal. However MNCF liberation was not blocked by cycloheximide and the time course of its release by macrophage monolayers stimulated by LPS differed from that described for interleukin-1. In addition human interleukin-1 when tested in dexamethasone-treated test rats failed to induce neutrophil migration. It is suggested that inhibition by glucocorticoids of LPS-induced neutrophil migration in vivo is not due to a direct effect upon the migrating cells but rather to an indirect one, i.e. through blockade of MNCF release by macrophages.
