Massively parallel reporter assays (MPRAs) functionally survey large numbers of cis-regulatory elements for regulatory potential, but do not identify the trans-acting factors that mediate any observed effects. Here we describe transMPRA — a reporter assay that efficiently combines multiplex CRISPR-mediated perturbation and MPRAs to identify trans-acting factors that modulate the regulatory activity of specific enhancers.

Induced pluripotent stem cells (iPSCs) are an essential tool for studying cellular differentiatio... more Induced pluripotent stem cells (iPSCs) are an essential tool for studying cellular differentiation and cell types that are otherwise difficult to access. We investigated the use of iPSCs and iPSC-derived cells to study the impact of genetic variation across different cell types and as models for studies of complex disease. We established a panel of iPSCs from 58 well-studied Yoruba lymphoblastoid cell lines (LCLs); 14 of these lines were further differentiated into cardiomyocytes. We characterized regulatory variation across individuals and cell types by measuring gene expression, chromatin accessibility and DNA methylation. Regulatory variation between individuals is lower in iPSCs than in the differentiated cell types, consistent with the intuition that developmental processes are generally canalized. Studies of the performance of the CMS drift tube barrel muon system are described, with results b... more Studies of the performance of the CMS drift tube barrel muon system are described, with results based on data collected during the CMS Cosmic Run at Four Tesla. For most of these data, the solenoidal magnet was operated with a central field of 3.8 T. The analysis of data from 246 out of a total of 250 chambers indicates a very good muon reconstruction capability, with a coordinate resolution for a single hit of about 260 µm, and a nearly 100% efficiency for the drift tube cells. The resolution of the track direction measured in the bending plane is about 1.8 mrad, and the efficiency to reconstruct a segment in a single chamber is higher than 99%. The CMS simulation of cosmic rays reproduces well the performance of the barrel muon detector.

Commissioning studies of the CMS hadron calorimeter have identified sporadic uncharacteristic noi... more Commissioning studies of the CMS hadron calorimeter have identified sporadic uncharacteristic noise and a small number of malfunctioning calorimeter channels. Algorithms have been developed to identify and address these problems in the data. The methods have been tested on cosmic ray muon data, calorimeter noise data, and single beam data collected with CMS in 2008. The noise rejection algorithms can be applied to LHC collision data at the trigger level or in the offline analysis. The application of the algorithms at the trigger level is shown to remove 90% of noise events with fake missing transverse energy above 100 GeV, which is sufficient for the CMS physics trigger operation.

The CMS Collaboration conducted a month-long data taking exercise, the Cosmic Run At Four Tesla, ... more The CMS Collaboration conducted a month-long data taking exercise, the Cosmic Run At Four Tesla, during October-November 2008, with the goal of commissioning the experiment for extended operation. With all installed detector systems participating, CMS recorded 270 million cosmic ray events with the solenoid at a magnetic field strength of 3.8 T. This paper describes the data flow from the detector through the various online and offline computing systems, as well as the workflows used for recording the data, for aligning and calibrating the detector, and for analysis of the data.

El propósito de la investigación es explicar el funcionamiento y dinámica del criptomercado de... more El propósito de la investigación es explicar el funcionamiento y dinámica del criptomercado de drogas. Con ese propósito se desarrolló un modelo de competencia monopolística con externalidades en red basado en los trabajos Belleflamme y Peitz (2010) y Dixit y Stiglitz (1977). La principal predicción del modelo es que las políticas de interdicción en estos mercados tienden a ser contrarrestadas por la entrada de nuevos vendedores, y la resultante apertura de nuevas páginas web en la red oscura. Otro hallazgo es que, debido a la presencia de efectos de externalidad en red en el consumo de drogas online, el endurecimiento de las políticas de interdicción produce un efecto de retroalimentación que lleva a que a un mayor número efectivo de consumidores decida adquirir los dispositivos para acceder a las plataformas de drogas en la web oscura y, así, compre drogas ilegales.

The above is not in itself a new problem, the novelty is the magnitude and extent achieved in recent years. To this challenge is added the need to generate inclusive public and residential spaces with the population with physical disabilities. These two elements establish a challenge for the housing sector, since this dynamic specifies the impacts against the mitigation of the described phenomena. The State, from this perspective, acquires a leading role since it promotes and regulates access to housing, being the main guarantor of access to housing for vulnerable populations. Defining a general overview of the subject is the main purpose of this document.

En Colombia, el fenómeno de la corrupción política no solo se genera y reproduce por la debilidad institucional del país, sino también por condiciones sociopolíticas y culturales, como el clientelismo y el narcotráfico. Este artículo indaga, desde la economía, el fenómeno de la corrupción política, partiendo del análisis pionero del Public Choice e incorporando un análisis de redes para caracterizar la complejidad estructural que definen los procesos de corrupción política, clientelismo y favorecimientos judiciales en Colombia.
