Lida Ghazanfari is a PhD student in the Biomedical Engineering Department at the Amirkabir University of Technology the most distinguished university in biomedical science and engineering in Iran. She obtained B.Sc. degree in biomaterials from Biomedical Engineering Department at the Amirkabir University of Technology (2007). Since 2010, she has been engaging in various researches besides the graduate students in different laboratories at Amirkabir University of Technology. She is also passionate about her teaching assistantship and being Lab instructor. Her research interests include:

Preparation and characterization of silica-coated Fe3O4 nanoparticles used as precursor of ferrofluids

Fe3O4 magnetic nanoparticles (MNPs) were synthesized by the co-precipitation of Fe3+ and Fe2+ wit... more Fe3O4 magnetic nanoparticles (MNPs) were synthesized by the co-precipitation of Fe3+ and Fe2+ with ammonium hydroxide. The sodium citrate-modified Fe3O4 MNPs were prepared under Ar protection and were characterized by Fourier transform infrared spectroscopy (FT-IR), transmission electron microscopy (TEM), X-ray powder diffraction (XRD) and vibrating sample magnetometer (VSM). To improve the oxidation resistance of Fe3O4 MNPs, a silica layer was coated onto the modified and unmodified MNPs by the hydrolysis of tetraethoxysilane (TEOS) at 50°C and pH 9. Afterwards, the silica-coated Fe3O4 core/shell MNPs were modified by oleic acid (OA) and were tested by IR and VSM. IR results revealed that the OA was successfully grafted onto the silica shell. The Fe3O4/SiO2 core/shell MNPs modified by OA were used to prepare water-based ferrofluids (FFs) using PEG as the second layer of surfactants. The properties of FFs were characterized using a UV–vis spectrophotometer, a Gouy magnetic balance, a laser particle size analyzer and a Brookfield LVDV-III+ rheometer.

In-vitro Application of Doxorubicin Loaded Magnetoplasmonic Thermosensitive Liposomes for Laser Hyperthermia and Chemotherapy of Breast Cancer

ABSTRACT We describe doxorubicin loaded magnetoplasmonic thermosensitive liposomes (MPTL-DOX), wh... more ABSTRACT We describe doxorubicin loaded magnetoplasmonic thermosensitive liposomes (MPTL-DOX), which are designed to combine features of magnetic drug targeting and laser hyperthermia-triggered drug release. The synthesized magnetite/gold nanoshells are stabilized using polyvinyl pyrolidone (PVP) with mean crystallite size of 15.8 ± 3.5 nm. The liposome formulation DPPC:cholesterol:DSPE-PEG2000 at 80:20:5 molar ratio shows DOX release of less than 5% at 37°C following 24 h incubation. MPTL-DOX shows encapsulation efficiencies of about 95% and 74% for DOX and magnetoplasmonic nanoshells (MPNS), respectively. The MPTL-DOX formulation displays a desired temperature sensitivity with 65% and 100% DOX release following laser irradiation and then 24 h incubation at 37°C, respectively. The rate of DOX release from liposome using this formulation is 0.09 which was obtained by heating to 43°C, and agrees well with the first kinetic model. A temperature rise between 4-12°C was achieved for MNS using 25 μg/ml and 300 μg/ml after 400 s respectively. For cytotoxicity measurement, one untreated (control) and two treatment groups are studied. The first treatment groups are: with MPNS only, with MPTL only, and laser irradiation only. The second treatment groups are: laser hyperthermia using MPTL, MPTL with magnetic field (MF), MPTL-DOX, and MPTL-DOX with MF. MPTL-DOX is targeted to breast cancer cell lines (MCF-7 cells) under a permanent magnetic field and exhibits a substantial increase in cytotoxicity and apoptotic effects. The results suggest that externally guided drug targeting can trigger drug release using an exogenous absorber in laser hyperthermia which can be used advantageously for thermo-chemotherapy of cancers.

Bifunctional magnetic-fluorescent composite nanoparticles (MPQDs) with Fe 3 O 4 MPs and Mn:ZnS/Zn... more Bifunctional magnetic-fluorescent composite nanoparticles (MPQDs) with Fe 3 O 4 MPs and Mn:ZnS/ZnS core-shell quantum dots (QDs) encapsulated in silica spheres were synthesized through reverse microemulsion method and characterized by X-ray powder diffraction, scanning electron microscopy, transmission electron microscopy, vibration sample magnetometer, and photoluminescence (PL) spectra. Our strategy could offer the following features: (1) the formation of Mn:ZnS/ZnS core/ shell QDs resulted in enhancement of the PL intensity with respect to that of bare Mn:ZnS nanocrystals due to the effective elimination of the surface defects; (2) the magnetic nanoparticles were coated with silica, in order to reduce any detrimental effects on the QD PL by the magnetic cores; and (3) both Fe 3 O 4 MPs and Mn:ZnS/ZnS core-shell QDs were encapsulated in silica spheres, and the obtained MPQDs became water soluble. The experimental conditions for the silica coating on the surface of Fe 3 O 4 nanoparticles, such as the ratio of water to surfactant (R), the amount of ammonia, and the amount of tetraethoxysilane, on the photoluminescence properties of MPQDs were studied. It was found that the silica coating on the surface of Fe 3 O 4 could effectively suppress the interaction between the Fe 3 O 4 and the QDs under the most optimal parameters, and the emission intensity of MPQDs showed a maximum. The bifunctional MPQDs prepared under the most optimal parameters have a typical diameter of 35 nm and a saturation magnetization of 4.35 emu/g at room temperature and exhibit strong photoluminescence intensity.

The synthetic procedure was illustrated in Scheme 1. Our strategy could offer the following features: (1) the formation of Mn:ZnS/ZnS core/shell QDs resulted in enhancement in the photoluminescence (PL) intensity with respect to that of bare Mn:ZnS nanocrystals due to the effective elimination of the surface defects, and the QDs’ chemical stability and photostability were also preserved ({26]; (2) the magnetic MPs were coated with silica, so that no interference of the QD PL by the magnetic particles was expected [20, 27]; and (3) both Fe;0, MPs and Mn:ZnS/ ZnS core-shell QDs were encapsulated in silica spheres, and the obtained MPQDs became water soluble. The obtained bifunctional MPQDs were characterized by X-ray powder diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), vibration sample magnetometer (VSM), and PL spectra. Besides the intensive PL, the MPQDs simultaneously exhibited mag- netic properties and could be separated from solution using a permanent magnet.
